Mdb1, a Fission Yeast Homolog of Human MDC1, Modulates DNA Damage Response and Mitotic Spindle Function

Abstract: During eukaryotic DNA damage response (DDR), one of the earliest events is the phosphorylation of the C-terminal SQ motif of histone H2AX (H2A in yeasts). In human cells, phosphorylated H2AX (γH2AX) is recognized by MDC1, which serves as a binding platform for the accumulation of a myriad of DDR factors on chromatin regions surrounding DNA lesions. Despite its important role in DDR, no homolog of MDC1 outside of metazoans has been described. Here, we report the characterization of Mdb1, a protein from the fission yeast Schizosaccharomyces pombe, which shares significant sequence homology with human MDC1 in their C-terminal tandem BRCT (tBRCT) domains. We show that in vitro, recombinant Mdb1 protein binds a phosphorylated H2A (γH2A) peptide, and the phospho-specific binding requires two conserved phospho-binding residues in the tBRCT domain of Mdb1. In vivo, Mdb1 forms nuclear foci at DNA double strand breaks (DSBs) induced by the HO endonuclease and ionizing radiation (IR). IR-induced Mdb1 focus formation depends on γH2A and the phospho-binding residues of Mdb1. Deleting the mdb1 gene does not overtly affect DNA damage sensitivity in a wild type background, but alters the DNA damage sensitivity of cells lacking another γH2A binder Crb2. Overexpression of Mdb1 causes severe DNA damage sensitivity in a manner that requires the interaction between Mdb1 and γH2A. During mitosis, Mdb1 localizes to spindles and concentrates at spindle midzones at late mitosis. The spindle midzone localization of Mdb1 requires its phospho-binding residues, but is independent of γH2A. Loss of Mdb1 or mutating its phospho-binding residues makes cells more resistant to the microtubule depolymerizing drug thiabendazole. We propose that Mdb1 performs dual roles in DDR and mitotic spindle regulation.

Abstract: 用叔丁醇作为反应介质,利用固定化脂肪酶催化油脂原料甲醇醇解反应制备生物柴油,消除了甲醇和甘油对酶的负面影响,酶的使用寿命显著延长. 用菜籽油作原料,叔丁醇和油脂的体积比为1:1,甲醇与油脂的摩尔比为4:1,3%的Lipozyme TLIM和1%的Novozym 435结合使用,35℃下130 r/min反应12 h,生物柴油得率可达95%. 该工艺在200 kg/d的规模下制得的生物柴油产品完全满足美国和德国生物柴油标准,脂肪酶重复使用200批次,酶活性基本没有下降. 且在叔丁醇介质体系中大豆油、桐籽油、棉籽油、乌桕油、泔水油、地沟油和酸化油都能被有效转化成生物柴油且脂肪酶保持很好的稳定性.
Psychological Typhoon Eye in the 2008 Wenchuan Earthquake

Abstract: Background On May 12, 2008, an earthquake measuring 8.0 on the Richter scale jolted Wenchuan, China, leading to 69,227 deaths and 374,643 injured, with 17,923 listed as missing as of Sept. 25, 2008, and shook the whole nation. We assessed the devastating effects on people's post-earthquake concern about safety and health. Methodology/Principal Findings From June 4 to July 15, 2008, we surveyed a convenience sample of 2,262 adults on their post-earthquake concern about safety and health. Residents in non-devastated areas (Fujian and Hunan Provinces, and Beijing) and devastated areas (Sichuan and Gansu Provinces) responded to a questionnaire of 5 questions regarding safety measures, epidemic disease, medical workers, psychological workers, and medication. The ANOVAs showed a significant effect of residential devastation level on the estimated number of safety measures needed, the estimated probability of the outbreak of an epidemic, and the estimated number of medical and psychological workers needed (Ps<0.001). The post-earthquake concern decreased significantly as the level of residential devastation increased. Because of the similarity with the meteorological phenomenon of the eye of a typhoon, we dubbed these findings a “Psychological Typhoon Eye”: the closer to the center of the devastated areas, the less the concern about safety and health a resident felt. Conclusions/Significance Contrary to common perception and ripple effect that the impact of an unfortunate event decays gradually as ripples spread outward from a center, a “Psychological Typhoon Eye” effect was observed where the post-earthquake concern was at its lowest level in the extremely devastated areas. The resultant findings may have implications for Chinese governmental strategies for putting “psychological comfort” into effect.

Abstract: In 2008 after a massive earthquake jolted Wenchuan, China, we reported an effect that we termed a “Psychological Typhoon Eye”: the closer to the center of the devastated area, the lower the level of concern felt by residents about safety and health. We now report on the progression of this effect and the development of new variations after the quake as well as investigating potential explanations.

Abstract: Similar to research on risky choice, the traditional analysis of intertemporal choice takes the view that an individual behaves so as to maximize the discounted sum of all future utilities. The well-known Allais paradox contradicts the fundamental postulates of maximizing the expected value or utility of a risky option. We describe a violation of the law of diminishing marginal utility as well as an intertemporal version of the Allais paradox.

Abstract: The problem of forward production of hadrons in heavy-ion collision at RHIC is revisited with modification of the theoretical treatment on the one hand and with the use of new data on the other. The basic formalism for hadronization remains the same as before, namely, recombination, but the details of momentum degradation and quark regeneration are improved. Recent data on the p/pi and pbar/p ratios are used to constrain the value of the degradation parameter. The p_T spectrum of the average charged particles is well reproduced. A prediction on the p_T dependence of the pbar/p ratio at eta=3.2 is made.

Abstract: Recent research progress on regulation network and biological roles of LFY gene in Arabidopsis thaliana and its homologue genes in floral development are reviewed emphatically in the present paper. LFY gene expresses widely in both vegetative and reproductive tissues in different higher plants, therefore investigation on role of LFY gene on flowering is of general significance. LFY gene plays an important role to promote flower formation by interaction and coordination with other genes,such as TFL, EMF, AP1, AP2, CAL, FWA, FT, AP3, PI, AG, UFO, CO, LD, GA1 etc, and a critical level of LFY expression is essential. LFY gene not only controls flowering-time and floral transition,but also plays an important role in inflorescence and floral organ development. It was situated at the central site in gene network of flowering regulation,positively or negatively regulates the level or activities of flowering-related genes. Some physiological factors, such as carbon sources, phytohormones, affect directly or indirectly the expression and actions of LFY gene. This indicates that level of LFY expression can also be regulated with physiological methods. It is probable that we can explain the principal mechanism of flowering by regulation network of LFY gene.
