Abstract: O Biodiesel é um biocombustível derivado de fontes renováveis como óleos vegetais, gordura animal, óleos egorduras residuais, que na presen a de um catalisador, reagem com o álcool etílico ou metílico, resultando emum combustível de alta qualidade e grande potencial na substitui o de óleo diesel. Essas rea es podem serrealizadas tanto pela rota química (processo convencional) utilizando catalisadores químicos, ácidos ou básicos,quanto pela rota biológica (processo n o convencional) com catalisadores biológicos, enzimas ou célulasimobilizadas. Neste processo químico, a glicerina é separada da gordura ou do óleo gerando dois produtos: umamistura de ésteres alquílicos de ácidos graxos (biodiesel) e glicerina (resíduo da rea o). No entanto, o biodieselpode ser obtido por rota química ou enzimática, ou inclusive através de uma diversidade de processos queincluem, entre outros, microemuls es, pirólise, rea es de esterifica o e transesterifica o. Sendo esta últimaalternativa a de maior ado o do ponto de vista tecnológico por apresentar eficaz convers o de triglicerídeos emésteres metílicos ou etílicos em menor tempo de rea o. Neste contexto, dada à diversidade de processosimplementados atualmente e sendo o conhecimento da matéria prima de qualquer processo industrial umelemento essencial para definir rotas e alternativas tecnológicas eficientes, o presente trabalho discute algumasconsidera es sobre a produ o de diferentes matérias-primas e rotas tecnológicas de processo.
STABILITY AND ACTIVITY OF THE PARTIALLY PURIFIED SPRAY-DRIED PROTEASE FROM BACILLUS SP SMIA-2 AND ITS CHARACTERIZATION AS A LAUNDRY DETERGENT ADDITIVE

Abstract: Enzymes, and particularly proteases, have become an important and indispensable part of industrial processes such as laundry detergents and food products. In this study, after production by Bacillus sp SMIA-2 in the complex medium and separation of solids from the fermentation broth, protease was dried and some biochemical properties of the enzyme after rehydration was investigated for the purposes of exploiting its potential as detergent additive. Studies on the enzyme characterization revealed that the optimum temperature of protease was 70oC. The enzyme retained about 55% of its maximum activity after 10 min incubation at 70oC. The optimum pH of the enzyme was found to be 8.5. After incubation of the enzyme solution for 2h at 25oC at pH 10.0, the activity was reduced to 53% of its maximum activity. Activity of the enzyme was stimulated in the presence of Ca2+. The spray-dried protease retained more than 90% activity after incubation for 30 min at 40oC with 0.25% (w/v) SDS and RENEX-95 and more than 60% when incubated with 0.25 and 0.5% TRITON x-100, H2O2 and RENEX-60. The stability of protease was good as the enzyme retained 64% of its activity after 30 minutes incubation at 40oC in the presence of the detergent Campeiro . The enzyme improved the washing performance of a detergent for removing stains from uniformly soiled clothes with cocoa, milk and sugar (EMPA 112).

Abstract: the thermostable proteolytic enzymes produced by the genus bacillus are commercially very important, being predominantly applied (35%) in detergents. in this work the production of proteases by a thermophilic bacillus sp. smia-2 using low-cost substrates was evaluated. in order to assess the use of the protease for industrial use, the stability of the enzyme activity at different ph values and temperatures was also studied. the protease activity secreted by bacillus sp. smia-2 in submerged cultures containing 0.5% (w/v) apple pectin, 0.1% (w/v) whey protein concentrate and 0.3% (w/v) corn steep liquor reached a maximum after 24 h, with levels of 54.3 u.mg-1 protein. a reduction in the pectin concentration to 0.3% (w/v) and increase in the whey protein levels to 0.3% (w/v) in the culture medium improved the protease production, which reached a maximum after 30 h, with levels of 72.2 u.mg-1 protein. studies on the characterization of the protein revealed that the most important kinetic properties of the enzyme were the high optimal temperature (70 °c) and high stability in a very broad ph range. the protease retained about 80% of its original activity when incubated at room temperature for 2 h in a ph range between 6.0 - 12.0. these properties represent important advantages for the possible use of the enzyme in detergent industries.
Production and partial characterization of alkaline polygalacturonase secreted by thermophilic Bacillus sp. SMIA-2 under submerged culture using pectin and corn steep liquor

Abstract: polygalacturonase production by the thermophilic bacillus sp. smia-2 cultivated in liquid cultures containing 0.5% (w/v) apple pectin and supplemented with 0.3% (w/v) corn steep liquor, reached its maximum after 36 hours with levels of 39 u.ml-1. the increase in apple pectin and corn steep liquor concentrations in the medium from 0.5 and 0.3%, respectively, to 0.65%, markedly affected the production of polygalacturonase, whose activity increased four times, reaching a maximum of 150.3 u.ml-1. studies on polygalacturonase characterization revealed that the optimum temperature of this enzyme was between 60-70 °c. thermostability profile indicated that the enzyme retained about 82 and 63% of its activity at 60 and 70 °c, respectively, after 2 hours of incubation. the optimum ph of the enzyme was found to be 10.0. after incubation of crude enzyme solution at room temperature for 2 hours at ph 8.0, a decrease of about 29% on its original activity was observed. at ph 10.0, the decrease was 25%.

Abstract: thermophilic bacillus sp. smia-2, produced protease when grown on apple pectic, whey protein and corn step liquor medium, whose concentration was varied from 3 to 10 gl-1, according to the central composite design 23. the experiments were conducted in shaker, at 50 °c, 150 rpm and initial ph 6.5. the results revealed that the culture medium affected both, cell growth and enzyme production. after graphical and numerical optimization procedure, the enzyme production reached its maximum value at 30 h fermentation, reaching, approximately, 70 u protein mg-1, suggesting that this process was partially associated to the growth.
Qualidade e composi??o química de cortes comerciais de carne de jacaré-do-pantanal (Caiman yacare)

Abstract: with the objective of characterize the alligator swampland meat, a total of 20 parts (commercial cuts) were taken of 5 carcass from these animals kept in captivity (cáceres mt) slaughtered with the weight ranged 2.5 to 3 kg. the commercial cuts were tail filet, back filet, loin filet and members. the physical chemistry characterizes of weight loss (ranged means from 38.99 to 42.28%) shear force (2.29 to 2.50 kgf) and the color component luminosity (54.01 to 56.02) were similar in the different cuts. however, the tenor of red was higher (p<0.05) for members (2.38), compared with loin filet, tail filet and back filet (0.29, 0.40 and 0.34% respectively). in the chemistry composition, any difference (p>0,05) among the cuts to total protein content. therefore the results shows different in moisture, fat and ashes . the cut tail filet showed (p>0,05) higher fat percentage (0,54%) than the others cuts loin filet, back filet and members (0,29; 0,40, and 0,34% respectively) these results show that the meat of alligator presents cuts with appearance very clear, similar to the color of fish, and when cooked, presents high tenderness. also the cuts shows low percentage of total lipids and the cut tail filet show to have more fat than the others cuts.

Abstract: the objective of this work was to evaluate the effect of different culture media, used over grafting spot, in ex vitro micrografting, in order to eliminate the cowpea aphid-borne mosaic virus (cabmv) in sour passion fruit plants (passiflora edulis sims f. flavicarpa deg.). cauline apexes, originated from infected plants, were micrografted in seedlings obtained by the germination of seeds in commercial sterilized substrate. experiments were conducted with the micrografting performed on the hypocotyl and epicotyl using five culture media, which differed on their concentration of phytoregulators applied on the grafting spot. the average rate of micrografting showing expanded leaf was 27.22 and 32.22%, when micrografting was performed on hypocotyl and epicotyl, respectively. in the micrografting performed on the hypocotyl, there was no effect of the culture media applied. in the micrografting performed on epicotyl, the ms medium supplemented with 0.1 mg l-1 3-indolbutiric acid (iba) and 1 mg l-1 6-benzylaminopurine (bap) resulted in 53.3% of micrografting showing expanded leaf, a number superior to the other treatments, and in a larger development of shoots. the indexing performed by the indirect elisa test, within 80 to 100 days after micrografting, showed that 93% of the plants tested presented no detectable virus.
Retrograde autologous priming in cardiopulmonary bypass in adult patients: effects on blood transfusion and hemodilution

Abstract: introduction: retrograde autologous priming (rap) is a cardiopulmonary bypass (cpb) method, at low cost. previous studies have shown that this method reduces hemodilution and blood transfusions needs through increased intra-operative hematocrit. objective: to evaluate rap method, in relation to standard cpb (crystalloid priming), in adult patients. methods: sixty-two patients were randomly allocated to two groups: 1) group rap (n = 27) of patients operated using the rap and; 2) control group of patients operated using cpb standard crystalloid method (n = 35). the rap was performed by draining crystalloid prime from the arterial and venous lines, before cpb, into a collect recycling bag. the main parameters analyzed were: 1) cpb hemodynamic data; 2) hematocrit and hemoglobin values; 3) the need for blood transfusions. results: it was observed statistically significant fewer transfusions during surgery and reduced cpb hemodilution using rap. the cpb hemodynamic values were similar, observing a tendency to use lower cpb flows in the rap group patients. conclusion: this investigation was designed to be a small-scale pilot study to evaluate the effects of rap, which were demonstrated concerning the cpb hemodilution and blood transfusions.

Abstract: this article wants to describe the interaction process of the anthropologist with teenagers in a lan house in porto alegre, rio grande do sul, brazil. first, the study will point out the peculiarities of the lan house, its location in the neighbourhood, as well as the everyday life of the young people who use this space for online playing or just for sociability. secondly, it will discuss as youth people establish their sense of belonging and strengthen their social bounds through the folga??es (joking relationships) and their online interactions in the mmorpg tibia game.
