Determinación de alcoholes grasos en el ingrediente activo D004 por cromatografía de gases-espectrometría de masas

Abstract: introduction: d004, a new lipid extract purified from the cuban royal palm (roystonea regia [kunth] f. cook) fruits, has been shown to be effective in experimental models of prostate hyperplasia, and to exhibit antioxidant and anti-inflammatory effects. objectives: to identify the fatty alcohols that could be present in d004 active ingredient using gas chromatography-mass spectrometry. methods: samples of nine batches of d004 were submitted to saponification with koh/etoh solution and the unsaponifiable fractions were extracted with n-hexane. the alcohols were analyzed as trimetilsilil derivatives, which were obtained by reaction with mstfa. the quantification was carried out by the internal standard method using 1-eicosanol. results: the total content of alcohols in d004 ranged from 0.18 to 5.10 %. this fraction was mainly composed of 1-octacosanol (52.94 %), 1-triacontanol (25.91 %), and 1-hexacosanol (14.40 %); whereas the minority components were 1-docosanol, 1-tricosanol, 1-tetracosanol, 1-pentacosanol, 1-heptacosanol, 1-nonacosanol, 1-dotriacontanol 1-tetratriacontanol, 1-tetradecanol, 1-pentadecanol, 1-hexadecanol, 1-octadecanol and 1-eicosanol were minor components, being the last six alcohols present in trace levels. conclusions: fatty alcohols were identified and quantified in d004 active ingredient by gas chromatography-mass spectrometry. the major components were 1-octacosanol, 1-triacontanol and 1-hexacosanol.

Abstract: Introducción: el D004, nuevo extracto lipídico purificado a partir de los frutos de la palma real cubana (Roystonea regia [Kunth] F. Cook), ha mostrado ser efectivo en modelos experimentales de hiperplasia prostática, y ha mostrado efectos antioxidantes y antiiflamatorios. Objetivos: identificar los alcoholes que pudieran estar presentes en el ingrediente activo D004 utilizando la cromatografía de gases acoplada a espectrometría de masa. Métodos: muestras de 9 lotes de D004 se sometieron a saponificación con una disolución de KOH/EtOH y las fracciones insaponificables fueron extraídas con n-hexano. Los alcoholes se analizaron como derivados trimetilsilil, obtenidos por reacción con N-metil, N-trimetilsilil trifluoroacetamida. La cuantificación se basó en el empleo de 1-eicosanol como patrón interno. Resultados: el D004 presenta un contenido total de alcoholes entre 0,18 y 5,10 %. Esta fracción está compuesta principalmente por 1-octacosanol (52,94 %), 1-triacontanol (25,91 %) y 1-hexacosanol (14,40 %); y presenta como componentes minoritarios: 1-docosanol, 1-tricosanol, 1-tetracosanol, 1-pentacosanol, 1-heptacosanol, 1-nonacosanol, 1-dotriacontanol, 1-tetratriacontanol, 1-tetradecanol, 1-pentadecanol, 1-hexadecanol, 1-octadecanol y 1-eicosanol, los seis últimos en niveles de trazas. Conclusiones: se identificaron y se cuantificaron mediante cromatografía de gases acoplada a espectrometría de masa los alcoholes grasos en el ingrediente activo D004. Abstract: this study summarizes the main findings of the preliminary characterization of the vegetal material obtained from dry and ground roystonea regia (kunth) o.f. cook mature fruits, used to obtain an active ingredient named d-004, a lipid extract from roystonea regia mature fruits that has shown to prevent testosterone or phenylephrine-induced prostate hyperplasia in rodents. this characterization of the starting vegetal material assessed the content of total and acid-insoluble ash, moisture and oil. the results support preliminary the following acceptance limits, all in dry weight basis, except the moisture: total ash £ 4,6 %, acid-insoluble ash £ 1,9 %; moisture £ 10,8 %; oil content 14,0-36,6 %.

Abstract: this paper shows the results of preliminary phytochemical studies conducted in ripe fruits of roystonea regia (kunth) o.f. cook. first, it was proved that 15 days were enough to guarantee the drying of the fruits in a well-ventilated place, at ambient temperature and protected from the direct exposure to the sun light. the results of the phytochemical screening demonstrated the presence of lipids, tripertenes and steroids, flavonoids, phenols and tannins, among others. it was determined that the extraction in s?xhlet with n-hexane, at a vegetal material-dissolvent ratio of 1:10 for 0.5 h, is an adequate method to calculate the lipid content of these fruits. these results will contribute to the phytochemical knowledge of this plant.

Abstract: El presente trabajo resume los aspectos principales de la caracterización preliminar del material vegetal obtenido de los frutos maduros, secos y molidos de Roystonea regia (Kunth) O.F. Cook, utilizado en la obtención del ingrediente activo denominado D004, extracto lipídico de los frutos de Roystonea regia, que previene la hiperplasia prostática inducida con testosterona o con fenilefrina en roedores. La caracterización del material de partida determinó los contenidos de cenizas totales, cenizas insolubles en ácido, humedad y aceite. Los resultados sustentan, de modo preliminar, los límites de aceptación siguientes, todos en base anhidra, exceptuando la humedad residual: cenizas totales £ 4,6 %, cenizas insolubles en ácido £ 1,9 %; humedad residual £ 10,8 %; contenido de aceite 14,0-36,6 %. Abstract: El presente trabajo muestra los resultados de estudios fitoquímicos preliminares realizados a los frutos maduros de Roystonea regia (Kunth) O.F. Cook. Primeramente se demostró que 15 días eran suficientes para garantizar el secado de los frutos en un local bien ventilado, a temperatura ambiente y protegido de la exposición directa a la luz solar. Los resultados del tamizaje fitoquímico mostraron principalmente la presencia de lípidos, además de triterpenos y esteroides, flavonoides, fenoles y taninos, entre otros. Se determinó que la extracción en S xhlet con n-hexano, a una relación material vegetal-disolvente de 1:10 durante 0,5 h, es un método adecuado para determinar el contenido lipídico de estos frutos. Estos resultados contribuirán al conocimiento fitoquímico de esta planta. This paper shows the results of preliminary phytochemical studies conducted in ripe fruits of Roystonea regia (Kunth) O.F. Abstract: introduction: the cuban royal palm tree oil, roystonea regia (kunth) o.f. cook, has possibilities to be used as raw material in the cuban pharmaceutical industry. objectives: to characterize the cuban royal palm tree oil and to study its fatty acid composition. methods: the official methods described in usp 27 were used and the composition of fatty acids was estimated by gas chromatography. results: relative density was 0.874 - 0.970 g/ml; refraction index 1.454 - 1.463; the saponification index ranged 190.0 to 221.8; the acidity index ranged 5.2 to 26.4; iodine index was 49.2 to 55.8; and the contents of non-saponificable material ranged 2.0 to 13.4 %. the total of fatty acids in the analyzed oils varied from 74.4 to 89.4 %. the composition of fatty acids standardized at 100 % was given by the following concentrations: c8:0 y c10:0 (0.1-0.9 %); c12:0 (17.9-27.8 %); c14:0 (9.3-12.7 %); c16:0 (9.3-15.0 %); c16:1 (0.1-0.6 %); c18:0 (2.5-3.7 %); c18:1 (27.3-37.3 %); c18:2 (12.8-17.4 %); y c18:3 (0.1-0.5 %). conclusions: some of the physical and chemical characteristics of r. regia fruit oil were studied in this paper.

Abstract: introduction: d004, an promissory active ingredient in treatment and prevention of begnin prostatic hyperplasia, is obtained from the fruit oil of roystonea regia (kunth) o.f. cook. it is composed by a mixture of free fatty acids including lauric and myristic acids, both very important by their pharmacological effects. objective: to determine the potential influence of harvest season on the oily content and the concentration of lauric and myristic acids from r. regia fruits. methods: each month for two years, it was possible to collect mature fruits from r. regia in a selected group. in dry and milled samples it was determined the oily content in a gravimetric way and that of fatty acids by gas chromatography. results: there were differences statistically significant in oily contents and of lauric and myristic acids; however, the contents determined in all the cases remained within the quality specifications established for the plant material. conclusions: the plant material collected for all the year may be used in obtaining of r. regia oil, raw material used in the production of d004.

Abstract: INTRODUCCIóN: el D004, un ingrediente activo promisorio en el tratamiento de la hiperplasia prostática benigna, se obtiene a partir del aceite del fruto de Roystonea regia (Kunth) O.F. Cook. Está compuesto por una mezcla de ácidos grasos libres que incluye los ácidos láurico y mirístico, ambos de gran interés por sus efectos farmacológicos. OBJETIVO: determinar la posible influencia de la época de colecta sobre el contenido de aceite y la concentración de los ácidos láurico y mirístico de los frutos de R. regia. MéTODOS: se colectaron cada mes, durante 2 a os, frutos maduros de R. regia en una población seleccionada. En las muestras secas y molidas se determinó el contenido de aceite gravimétricamente y el de ácidos grasos por cromatografía de gases. RESULTADOS: se encontraron diferencias estadísticas significativas en los contenidos de aceite y de los ácidos láurico y mirístico; no obstante, los contenidos determinados en todos los casos se mantuvieron dentro de las especificaciones calidad establecidas para el material vegetal. CONCLUSIONES: el material vegetal colectado durante todo el a o puede ser empleado en la obtención del aceite de R. regia, materia prima empleada en la producción de D004. Abstract: INTRODUCCIóN: el aceite de la palma real cubana, Roystonea regia (Kunth) O.F. Cook, tiene perspectivas de aplicación como materia prima, en la industria farmacéutica cubana. OBJETIVOS: caracterizar el aceite de palma real cubana y estudiar su composición de ácidos grasos. MéTODOS: se emplearon métodos oficiales descritos en la USP 27. La composición de ácidos grasos se determinó mediante cromatografía de gases. RESULTADOS: la densidad relativa estuvo entre 0,874 y 0,970 g/mL; el índice de refracción entre 1,454 y 1,463; el índice de saponificación entre 190,0 y 221,8; el índice de acidez entre 5,2 y 26,4; el índice de yodo entre 49,2 y 55,8; y el contenido de material no saponificable entre 2,0 y 13,4 %. El total de ácidos grasos en los aceites analizados varió entre 74,4 y 89,4 %. La composición de ácidos grasos, normalizada 100 %, estuvo dada por las concentraciones siguientes: C8:0 y C10:0 (0,1-0,9 %); C12:0 (17,9-27,8 %); C14:0 (9,3-12,7 %); C16:0 (9,3-15,0 %); C16:1 (0,1-0,6 %); C18:0 (2,5-3,7 %); C18:1 (27,3-37,3 %); C18:2 (12,8-17,4 %); y C18:3 (0,1-0,5 %). CONCLUSIONES: se estudiaron algunas de las características físicas y químicas del aceite obtenido de los frutos enteros de R. regia.
