Abstract: the d002, an antioxidant active ingredient extracted from the apis mellifera bees wax was characterized from the physicochemical point of view analyzing its interaction with excipients of pharmaceutical interest. the d-002 is a creamy white odourless fluid powder with losses by £ 1 % dry; it is water and ethanol insoluble and very slightly soluble in other organic solvents. its composition, determined by gas chromatography was: 1-tetracosanol (6-15 %), 1-hexacosanol (7-20 %), 1-octacosanol (12-20 %), 1-triacontanol (25-35 %, 1-dotriacontanol (18-25 %) and 1-tetratriacontaol (£ 7,5 %) for 3 85 % of purity. it remained stable during 5 years in the iv climatic zone and its analysis by differential scanning calorimetry showed 2 fusion transitions at 59.0 and 81.1 °c. without decomposition, a high thermal stability up to 200 °c, as well as a lack of interactions with lactose, starch, sodium croscarmelosa, polyvinylpyrrolidone, microcrystalline cellulose, and magnesium stearate allowing the use of these excipients in tablets formula.

Abstract: El D-002, ingrediente activo antioxidante extraído de la cera de abejas Apis mellifera, fue caracterizado desde el punto de vista físicoquímico, de igual forma se analizó su interacción con excipientes de interés farmacéutico. El D-002 es un polvo fluido inodoro de color blanco a crema, con pérdidas por secado £ 1 %; es insoluble en agua y etanol, y muy ligeramente soluble en otros disolventes orgánicos. Su composición, determinada por cromatografía de gases, fue: 1-tetracosanol (6-15 %), 1-hexacosanol (7-20 %), 1-octacosanol (12-20 %), 1-triacontanol (25-35 %) 1-dotriacontanol (18-25 %) y 1-tetratriacontanol (£ 7,5 %), para una pureza 3 85 %. Fue estable durante 5 a os en la zona climática IV y su análisis por calorimetría diferencial de barrido mostró 2 transiciones de fusión a 59,0 y 81,1 °C sin descomposición, una alta estabilidad térmica hasta 200 °C, así como la ausencia de interacciones con lactosa, almidón, croscarmelosa sódica, polivinil pirrolidona, celulosa microcristalina y estearato de magnesio, lo que posibilita el empleo de estos excipientes en la formulación de las tabletas. Abstract: introduction: d004, a new lipid extract purified from the cuban royal palm (roystonea regia [kunth] f. cook) fruits, has been shown to be effective in experimental models of prostate hyperplasia, and to exhibit antioxidant and anti-inflammatory effects. objectives: to identify the fatty alcohols that could be present in d004 active ingredient using gas chromatography-mass spectrometry. methods: samples of nine batches of d004 were submitted to saponification with koh/etoh solution and the unsaponifiable fractions were extracted with n-hexane. the alcohols were analyzed as trimetilsilil derivatives, which were obtained by reaction with mstfa. the quantification was carried out by the internal standard method using 1-eicosanol. results: the total content of alcohols in d004 ranged from 0.18 to 5.10 %. this fraction was mainly composed of 1-octacosanol (52.94 %), 1-triacontanol (25.91 %), and 1-hexacosanol (14.40 %); whereas the minority components were 1-docosanol, 1-tricosanol, 1-tetracosanol, 1-pentacosanol, 1-heptacosanol, 1-nonacosanol, 1-dotriacontanol 1-tetratriacontanol, 1-tetradecanol, 1-pentadecanol, 1-hexadecanol, 1-octadecanol and 1-eicosanol were minor components, being the last six alcohols present in trace levels. conclusions: fatty alcohols were identified and quantified in d004 active ingredient by gas chromatography-mass spectrometry. the major components were 1-octacosanol, 1-triacontanol and 1-hexacosanol.

Abstract: Introducción: los esteroles han sido ampliamente estudiados por su importancia nutricional y farmacéutica. Sin embargo, para nuestro conocimiento, la composición de esteroles del aceite de los frutos de la palma real cubana (Roystonea regia) no ha sido determinada hasta el momento. Objetivos: determinar la composición de esteroles que pudieran estar presentes en la fracción insaponificable del aceite de R. regia mediante CG-EM. Métodos: muestras de dos lotes de aceite de R. regia fueron sometidas al procedimiento para la determinación de esteroles establecido por el del Instituto de Nutracéuticos de los EE.UU., el cual consiste, fundamentalmente, en una saponificación con disolución de KOH/EtOH y posterior extracción con n-hexano de las fracciones insaponificables. Las fracciones obtenidas fueron analizadas por CG-EM como derivados TMS e identificadas por comparación de sus espectros con los de patrones comerciales y los de la base de espectros Wiley. Para la cuantificación se utilizó el colestano como patrón interno. Resultados: en la fracción insaponificable del aceite de R. regia se encontró un contenido total de esteroles de 66,1%; lo que representa un 0,14 % del aceite. La fracción de esteroles, estuvo compuesta principalmente por a -sitosterol (51,2 %), estigmasterol (9,6 %), campesterol (9,2 %), 24-metilen-cicloartanol (9,2 %), 5-avenasterol (8,9 %), cicloartanol (7,5 %); además de otros componentes minoritarios como cicloartenol, -sitosterol y colesterol. Conclusiones: se identificaron y se cuantificaron mediante CG-EM los esteroles de la fracción insaponificable del aceite de R. regia, en la cual el a-sitosterol resultó el componente mayoritario. Estos resultados son una contribución al estudio de la composición química de dicho aceite y pudieran avalar su posible utilidad nutricional y seguridad. Abstract: Introducción: el D004, nuevo extracto lipídico purificado a partir de los frutos de la palma real cubana (Roystonea regia [Kunth] F. Cook), ha mostrado ser efectivo en modelos experimentales de hiperplasia prostática, y ha mostrado efectos antioxidantes y antiiflamatorios. Objetivos: identificar los alcoholes que pudieran estar presentes en el ingrediente activo D004 utilizando la cromatografía de gases acoplada a espectrometría de masa. Métodos: muestras de 9 lotes de D004 se sometieron a saponificación con una disolución de KOH/EtOH y las fracciones insaponificables fueron extraídas con n-hexano. Los alcoholes se analizaron como derivados trimetilsilil, obtenidos por reacción con N-metil, N-trimetilsilil trifluoroacetamida. La cuantificación se basó en el empleo de 1-eicosanol como patrón interno. Resultados: el D004 presenta un contenido total de alcoholes entre 0,18 y 5,10 %. Esta fracción está compuesta principalmente por 1-octacosanol (52,94 %), 1-triacontanol (25,91 %) y 1-hexacosanol (14,40 %); y presenta como componentes minoritarios: 1-docosanol, 1-tricosanol, 1-tetracosanol, 1-pentacosanol, 1-heptacosanol, 1-nonacosanol, 1-dotriacontanol, 1-tetratriacontanol, 1-tetradecanol, 1-pentadecanol, 1-hexadecanol, 1-octadecanol y 1-eicosanol, los seis últimos en niveles de trazas. Conclusiones: se identificaron y se cuantificaron mediante cromatografía de gases acoplada a espectrometría de masa los alcoholes grasos en el ingrediente activo D004. Abstract: the d004 is a new active ingredient extracted from the cuban real palm (roystonea regia), promising in the treatment of the benign prostatic hyperplasia. to determine its stability in amber glass flasks studies were conducted where samples underwent different conditions. in stress situations d004 is sensitive to the thermal and oxidative degradation. under accelerated conditions (40 ± 2 oc ad 75 ± 5 % of relative humidity) there weren't significant changes in any assessed parameters during 12 months allowing predicting a useful life time of at least 2 years under less drastic conditions. it was corroborated under climatic conditions ii (25 ± 2oc and 60± 5% of relative humidity and iv (30 ± 2 oc and 70 ± 5 % of relative humidity).

Abstract: El D004 es un nuevo ingrediente activo extraído de los frutos de la palma real cubana (Roystonea regia), promisorio en el tratamiento de la hiperplasia prostática benigna. Para determinar su estabilidad en frascos de vidrio ámbar, se realizaron estudios donde las muestras se sometieron a diferentes condiciones. En condiciones de estrés se observó que el D004 es susceptible a la degradación térmica y oxidativa. En condiciones aceleradas (40 ± 2 oC y 75 ± 5 % humedad relativa) no se observaron cambios significativos en ninguno de los parámetros evaluados durante 12 meses, lo que permitió predecir un tiempo de vida útil de al menos 2 a os en condiciones menos drásticas. Esto fue corroborado en las condiciones de las zonas climáticas II (25 ± 2 oC y 60 ± 5 % humedad relativa) y IV (30 ± 2 oC y 70 ± 5 % humedad relativa). Validación de un nuevo método analítico por CG con columna capilar para la determinación de alcoholes de alto peso molecular en policosanol ingrediente activo

Abstract: policosanol is a mixture of 8 long chain primary aliphatic alcohols (c24-c34), isolated and purified from sugar cane (saccharum officinarum l.) wax, wich cholesterol-lowering efficacy, safety and tolerability has been demonstrated. several methods for determing policosanol by packed gc column have been previously validated. however, the advantages achieved with the capillary gc make it worldwide currently extended and overcome to the packed ones. a new gas chromatographic method using a capillary column was developed and validated for the determination of the fatty alcohols that compose policosanol active ingredient. the alcohols were analyzed as trimethylsilyl derivatives. good linearity (r2 = 0,9954, cvresponse=1,07%, cvslope=1,89% and the confidence interval included the zero, then there was no bias) and accuracy (mean recovery = 100,45%) of the method were proven over a range of 80-120% of the nominal concentration. it was also proved its specificity even when samples were subject to stress conditions. repeatablility and intermediate precision at the nominal 100% value fulfilled the acceptance criteria (< 2%). ruggedness was evaluated through an intralaboratory experimental design, in which seven operational changes were made, and there was not found any effect on the observed results. the method was sussefully validated being suitable for the quality control process and the stability studies of this active ingredient.

Abstract: this study summarizes the main findings of the preliminary characterization of the vegetal material obtained from dry and ground roystonea regia (kunth) o.f. cook mature fruits, used to obtain an active ingredient named d-004, a lipid extract from roystonea regia mature fruits that has shown to prevent testosterone or phenylephrine-induced prostate hyperplasia in rodents. this characterization of the starting vegetal material assessed the content of total and acid-insoluble ash, moisture and oil. the results support preliminary the following acceptance limits, all in dry weight basis, except the moisture: total ash £ 4,6 %, acid-insoluble ash £ 1,9 %; moisture £ 10,8 %; oil content 14,0-36,6 %.

Abstract: this paper shows the results of preliminary phytochemical studies conducted in ripe fruits of roystonea regia (kunth) o.f. cook. first, it was proved that 15 days were enough to guarantee the drying of the fruits in a well-ventilated place, at ambient temperature and protected from the direct exposure to the sun light. the results of the phytochemical screening demonstrated the presence of lipids, tripertenes and steroids, flavonoids, phenols and tannins, among others. it was determined that the extraction in s?xhlet with n-hexane, at a vegetal material-dissolvent ratio of 1:10 for 0.5 h, is an adequate method to calculate the lipid content of these fruits. these results will contribute to the phytochemical knowledge of this plant.
