Drosophila muscleblind Codes for Proteins with One and Two Tandem Zinc Finger Motifs

Abstract: Muscleblind-like proteins, Muscleblind (Mbl) in Drosophila and MBNL1-3 in vertebrates, are regulators of alternative splicing. Human MBNL1 is a key factor in the etiology of myotonic dystrophy (DM), a muscle wasting disease caused by the occurrence of toxic RNA molecules containing CUG/CCUG repeats. MBNL1 binds to these RNAs and is sequestered in nuclear foci preventing it from exerting its normal function, which ultimately leads to mis-spliced mRNAs, a major cause of the disease. Muscleblind-proteins bind to RNAs via N-terminal zinc fingers of the Cys3-His type. These zinc fingers are arranged in one (invertebrates) or two (vertebrates) tandem zinc finger (TZF) motifs with both fingers targeting GC steps in the RNA molecule. Here I show that mbl genes in Drosophila and in other insects also encode proteins with two TZF motifs, highly similar to vertebrate MBNL proteins. In Drosophila the different protein isoforms have overlapping but possibly divergent functions in vivo, evident by their unequal capacities to rescue the splicing defects observed in mbl mutant embryos. In addition, using whole transcriptome analysis, I identified several new splicing targets for Mbl in Drosophila embryos. Two of these novel targets, kkv (krotzkopf-verkehrt, coding for Chitin Synthase 1) and cora (coracle, coding for the Drosophila homolog of Protein 4.1), are not muscle-specific but expressed mainly in epidermal cells, indicating a function for mbl not only in muscles and the nervous system.

Abstract: Der Fokus qualitativer Sozialforschung und der eingesetzten computergestützten Forschungsinstrumente richtet sich immer noch weitgehend auf die Analyse verbaler Daten. Das Zusammenwachsen von Digitaltechnologien und traditionellen Technologien zur Video- und Audioaufzeichnung er ffnet neue Chancen für qualitative Forschungsprozesse durch die M glichkeit zur Analyse multicodaler Videodaten. Zugleich entstehen aber auch neue Anforderungen an Forschende und Forschungsinstrumente. Der Beitrag besch ftigt sich im ersten Teil mit Chancen und Grenzen der Nutzung multicodaler Daten für die Analyse unter besonderer Berücksichtigung der M glichkeiten, die sich durch den Einsatz von Digitaltechnologien bei der Erhebung und Analyse ergeben k nnen. In einem zweiten Teil werden Hinweise für Auswahl und Einsatz von Digitaltechnologien zur Aufnahme multicodaler Daten gegeben. Der dritte Teil des Beitrags besch ftigt sich mit M glichkeiten der Datenaufbereitung für die computerunterstützte Analyse multicodaler Daten. Reine Transkriptionsverfahren werden eher für deduktiv ausgerichtete Studien empfohlen. Die Integration von Videodaten als Datenquelle wird nur von wenigen für die Datenanalyse entwickelten Programmen unterstützt. Ein m glicher Weg um die Beschr nkung auf diese Programme zu durchbrechen wird mit modular eingesetzten Analysewerkzeugen vorgeschlagen und an einem Beispiel verdeutlicht. The growing use of digital technology in traditional video- and audio-work has opened up new possibilities for the analysis of multimedia data in qualitative research. This development presents new challenges for researchers and the research tools they use. This article examines the possibilities and limitations associated with the use of multimedia data in qualitative research and the new opportunities in data collection and analysis are discussed. The second section deals with the collection of multimedia data. The third section examines the computer aided data analysis of multimedia data. It is argued that the use of special methods of transcription is more relevant to deductively oriented research projects. There are few software applications for data analysis of multimedia data. A possible way to create more applications is the modular use of software tools. This proposal is illustrated with an example. Towards Quantitative Characterisation of the Small Force Transducer Used in Nanoindentation Instruments  [PDF]

Quantitative characterization of the mechanical properties of materials in micro-/nano-scale using depth-sensing indentation technique demands high performance of nanoindentation instruments in use. In this paper, the efforts to calibrate the capacitive force transducer of a commercial nanoindentation instrument are presented, where the quasi-static characteristic of the force transducer has been calibrated by a precise compensation balance with a resolution of ~1 nN. To investigate the dynamic response of the transducer, an electrostatic MEMS (Micro-Electro-Mechanical System) based on nano-force transfer standard with nano-Newton (10-9 Newton) resolution and a bandwidth up to 6 kHz have been employed. Preliminary experimental results indicate that 1) the force transducer under calibration has a probing force uncertainty less than 300 nN (1σ) in the calibration range of 1 mN; 2) the transient duration at contact points amounts to 10 seconds; 3) the overshoot of engagement is pre-load dependent.

Abstract: introduction: studies about the use of sheep in surgical training and experimental otologic surgery are rare. this study intends to contribute to the knowledge on this field. aim: to study sheep inner ear structures using computerized tomography and serial cross-sections to collect more accurate morphometric data to compare sheep and human ears. material and methods: this descriptive study compared the inner structures of sheep and human ears. measurements were made using computerized tomography, and they were stored in a dicom compact disc for later analysis and manipulation, with a program used for medical image analysis (osíris 4.16). results: mean measures for sheep and human ears were found to be similar in this morphological study. most structures (10 out of 15) maintained the 2/3 ratio of sheep to human ear. conclusion: the results of this morphometric study of sheep ear are an important contribution to the development of an animal model to be used for surgical training and experimental otologic surgeries.

Abstract: 本文目的在于说明有限区域内潮坪小环境沉积物的矿物学和地球化学特点及沉积过程的改造作用。沉积物样品取自汪额诺格岛与联邦德国的德国湾南岸之间的障壁潮坪。粒度分析表明,障壁潮坪由大体平行于大陆的泥坪、混合坪到砂坪三个带组成。粘土矿物以伊利石为主,其次为蒙脱石、高岭石和绿泥石,与北海粘土矿物组合一致。粘土矿物组成和重金属Fe、Mn、Cu、Pb、Zn含量在潮坪不同部位相差无几,说明沉积物受到潮汐水流反覆侵蚀和再沉积作用的混合作用。近潮口附近有机碳含量增高是潮汐水流从北海通过进潮口携入有机物的结果。
