Fish oil supplementation reverses the effect of cholesterol on apoptotic gene expression in smooth muscle cells

Abstract: An in vivo/in vitro cell model was used, culturing SMC isolated from chicks exposed to an atherogenic cholesterol-rich diet with 5% of cholesterol (SMC-Ch) alone or followed by an anti-atherogenic fish oil-rich diet with 10% of menhaden oil (SMC-Ch-FO) and from chicks on standard diet (SMC-C). Cells were exposed to 25-HC, studying apoptosis levels by flow cytometry (Annexin V) and expressions of caspase-3, c-myc, and p53 genes by quantitative real-time reverse transcriptase-polymerase chain reaction. Results: Exposure to 25-HC produced apoptosis in all three SMC cultures, which was mediated by increases in caspase-3, c-myc, and p53 gene expression. Changes were more marked in SMC-Ch than in SMC-C, indicating that dietary cholesterol makes SMC more susceptible to 25-HC-mediated apoptosis. Expression of p53 gene was elevated in SMC-Ch-FO. This supports the proposition that endogenous levels of p53 protect SMC against apoptosis and possibly against the development of atherosclerosis. Fish oil attenuated the increase in c-myc levels observed in SMC-C and SMC-Ch, possibly through its influence on the expression of antioxidant genes.Replacement of a cholesterol-rich diet with a fish oil-rich diet produces some reversal of the cholesterol-induced changes, increasing the resistance of SMC to apoptosis.Gene expression is influenced by dietary fat [1], gene transcription is regulated by dietary polyunsaturated fatty acids [2], and the nutritional control of gene regulation guides the transformation of SMC into foam cells in atherosclerosis [3]. Abstract: Smooth muscle cells (SMCs) undergo changes related to proliferation and apoptosis in the physiological remodeling of vessels and in diseases such as atherosclerosis and restenosis. Recent studies also have demonstrated the vascular cell proliferation and programmed cell death contribute to changes in vascular architecture in normal development and in disease. The present study was designed to investigate the apoptotic pathways induced by 25-hydroxycholesterol in SMCs cultures, using an in vivo/in vitro cell model in which SMCs were isolated and culture from chicken exposed to an atherogenic cholesterol-rich diet (SMC-Ch) and/or an antiatherogenic fish oil-rich diet (SMC-Ch-FO). Cells were exposed in vitro to 25-hydroxycholesterol to study levels of apoptosis and apoptotic proteins Bcl-2, Bcl-XL and Bax and the expression of bcl-2 and bcl-xL, genes. The quantitative real-time reverse transcriptase-polymerase chain reaction and the Immunoblotting western blot analysis showed that 25-hydroxycholesterol produces apoptosis in SMCs, mediated by a high increase in Bax protein and Bax gene expression. These changes were more marked in SMC-Ch than in SMC-Ch-FO, indicating that dietary cholesterol produces changes in SMCs that make them more susceptible to 25-hydroxycholesterol-mediated apoptosis. Our results suggest that the replacement of a cholesterol-rich diet with a fish oil-rich diet produces some reversal of cholesterol-induced changes in the apoptotic pathways induced by 25-hydroxycholesterol in SMCs cultures, making SMCs more resistant to apoptosis.

Abstract: in order to propose the necessary moral reflection about the personal responsibility that each researcher assumes when he performs scientific research in human beings, an essay of moral and scientific themes is presented, using information from the relevant scientific and ethical literature. around the concepts of heteronomous and autonomous ethics, it is proposed that ultimately and beyond informed consent and training courses on research ethics, the behavior of the researcher will depend on his/her own moral responsibility, tested in different situational contexts. two explanatory models are used in order to understand this dynamic, the one of normality in mental health and the one of moral development of men. we conclude that the research process in human beings is a scientific and moral activity that, depending in various situations of conflicts of interest, will always test the researcher’s moral controls.

Abstract: Con el objetivo de plantear la necesaria reflexión moral sobre la responsabilidad personal que asume cada investigador cuando realiza investigación científica en seres humanos, se presenta un ensayo de temática científica y moral, utilizando información de la literatura científica y ética pertinentes. En torno a los conceptos de ética heterónoma y ética autónoma, se plantea que, en última instancia y allende el consentimiento Informado y los cursos de adiestramiento sobre ética en investigación, la conducta del/de la investigador/a dependerá de su propia responsabilidad moral puesta a prueba en diversos contextos situacionales. Se utilizan dos modelos explicativos para comprender esta dinámica, el de normalidad en salud mental y el del desarrollo moral del hombre. Se concluye que el proceso de investigación en seres humanos constituye una actividad científica y moral que, dependiendo de diversas situaciones de conflicto de interés, pondrá siempre a prueba los controles morales del investigador. Abstract: We prove a laplacian comparison theorem in the barrier sense for the function distance to the boundary of Riemannian manifolds with nonnegative Ricci curvature, area and mean curvature of the boundary bounded above. As an application we get volume estimates, different to the ones obtained by Heintz-Karcher, and area estimates. With those estimates we prove Sormani-Wenger Intrinsic Flat (SWIF) compactness theorems for sequences of such manifolds with uniform bounds.
Convergence of Manifolds and Metric Spaces with Boundary

Abstract: We study sequences of oriented Riemannian manifolds with boundary and, more generally, integral current spaces and metric spaces with boundary. We prove theorems demonstrating when the Gromov-Hausdorff [GH] and Sormani-Wenger Intrinsic Flat [SWIF] limits of sequences of such metric spaces agree. Thus in particular the limit spaces are countably $\mathcal{H}^n$ rectifiable spaces. From these theorems we derive compactness theorems for sequences of Riemannian manifolds with boundary where both the GH and SWIF limits agree. For sequences of Riemannian manifolds with boundary we only require nonnegative Ricci curvature, upper bounds on volume, noncollapsing conditions on the interior of the manifold and diameter controls on the level sets near the boundary.
