Trypanosome Prereplication Machinery: A Potential New Target for an Old Problem

Abstract: Approximately ten million people suffer from Chagas disease worldwide, caused by Trypanosoma cruzi, with the disease burden predominately focused in Latin America. Sleeping sickness is another serious health problem, caused by Trypanosoma brucei, especially in sub-Saharan countries. Unfortunately, the drugs currently available to treat these diseases have toxic effects and are not effective against all disease phases or parasite strains. Therefore, there is a clear need for the development of novel drugs and drug targets to treat these diseases. We propose the trypanosome prereplication machinery component, Orc1/Cdc6, as a potential target for drug development. In trypanosomes, Orc1/Cdc6 is involved in nuclear DNA replication, and, despite its involvement in such a conserved process, Orc1/Cdc6 is distinct from mammalian Orc1 and Cdc6 proteins. Moreover, RNAi-mediated silencing of trypanosome Orc1/Cdc6 expression in T. brucei decreased cell survival, indicating that Orc1/Cdc6 is critical for trypanosome survival. 1. Introduction Currently, approximately ten million people have Chagas disease worldwide, with the disease burden being centered in Latin America. In 2008, approximately ten thousand people died from this disease. Sleeping sickness is another serious health problem, particularly in sub-Saharan countries; in the first half of the twentieth century, this disease practically decimated entire communities in central Africa [1]. After many surveillance programs, 2009 marked the first time in 50 years that less than ten thousand new cases of sleeping sickness were reported in Africa (http://www.who.org/). The causal agents of Chagas disease and sleeping sickness are, respectively, the protozoan parasites Trypanosoma cruzi and T. brucei of the Kinetoplastida order. Two subspecies of T. brucei, T. b. rhodesiense, and T. b. gambiense, are responsible for acute sleeping sickness in Eastern and Southern Africa and chronic sleeping sickness in Western and Central Africa, respectively. Chronic sleeping sickness accounts for approximately 95% of all reported cases. These protozoa have life cycles that alternate between a mammalian host and an insect host. T. cruzi epimastigotes are a noninfective life cycle stage of the parasite that proliferate by binary fission in the guts of Triatoma infestans insects, which are more commonly known as kissing bugs. These epimastigotes then transform into the infective metacyclic form in the insect hindgut. Abstract: the objective of this systematic review was to retrieve topics of relevance to the implications of leptin in modulating blood pressure and in the genesis, prevention and treatment of hypertension associated with obesity. considering the hormone as a common link between the two pathological conditions, were identified actions to reduce the leptinemia, proposed and discussed in the literature. it was found that nursing lacks production of knowledge to subsidize the assistance to hypertensive obesity that present alterations in serum leptin. were described signs and multifactorials symptoms dynamically interrelated and paradoxical in the manifestation of the clinical case studied, with effective management interventions in caring for those clients.

Abstract: objective: assess the prevalence of white coat hypertension in the municipality of dumont, in s?o paulo state, and map study participants' demographics, and physiologic and metabolic changes. methods: one hundred and nine (109) users of the municipal health service were selected and then divided into three groups: normotension, essential hypertension, and white coat hypertension after blood pressure was measured through oscilometry and blood pressure monitoring exam at outpatient unit. variables found between the groups were compared based on interview, data measurements, and laboratory exams. anova and tukey tests were used for statistical data. results were expressed as means ± standard error of means. p<0.05 was considered statistically significant. results: the prevalence of white coat hypertension was 34.1%. females were predominant: mean age 45.3 years, increased body mass index, waist/hip ratio, plasma glucose level and creatinine when compared to hypertensives and/or normotensives. no correlation was found between white coat hypertension and demographic variables. conclusion: the differences found between the groups and clinical and biochemical variations lead to the conclusion that white coat hypertension is a condition that should be investigated in normotensive and hypertensive individuals distinctively.

Abstract: using a questionnaire with open and closed questions, this study aimed to: verify among nursing professionals which are the most used regions for intramuscular medication application; identify the method they use for delimiting the puncture regions and site; identify their knowledge of complications and contraindications of using the regions and verifying the use of the ventrogluteal region. thirty-two professionals participated in the study, which used the dorsogluteal (65.62%) and deltoid (31.25%) regions. the need for updating these professionals was disclosed, especially regarding to anatomy, the adequate terminology used for designating the regions and the knowledge of complications and contraindications. investments in training these professionals are essential for using the ventrogluteal region.

Abstract: historically, nurses' management functions have evidenced that their conduct is compatible with programmatic commitments. this fact was observed in the analysis of 18 studies carried out in healthcare services located in different brazilian regions. since the 1990's, management experts have alerted that traits such as commitment, interdisciplinarity, risk, uncertainty, spirituality, and ethical involvement will frame the characteristics of the human capital of 21st century organizations. in this much more humane environment, people's integrity and their way of relating at work must be based on the dimension of being. in order to face the new century institutions' healthcare demands, nurses are now urged to present a conduct permeated by the ethics of the attitudinal commitment. thus, this study aims to stimulate a critical reflection on this ethical trend of nurses and finally propose that this trend be an essential value in nursing management processes.

Abstract: this descriptive and exploratory study with a qualitative design aimed to describe and analyze discussions in online chats about mental disorders in a psychiatric nursing course as part of an undergraduate nursing program. the sample consisted of 32 undergraduate students who attended the course. data analysis showed that the discussions through online chat sessions permeated the acquisition of knowledge, procedures, attitudes and values and promoted students' active participation. the results reaffirm the discussions' importance for students' learning and showed the potential of technology resources, such as real-time communication tools, to support and improve teaching possibilities in psychiatric nursing.
Marketing profissional e visibilidade social na enfermagem: uma estratégia de valoriza o de recursos humanos

Abstract: En el ámbito internacional hay un movimiento de valorización de la enfermería entre las profesiones de salud, que usa para ello, herramientas de marketing profesional. Por medio de una revisión integradora de la literatura, se discutió el marketing como herramienta para el incremento de la visibilidad de la enfermería. Se utilizaron las bases LILACS, MEDLINE, Web of Science y SciELO. Se seleccionaron publicaciones que asociaban el marketing a la enfermería en el período de 2000 a 2010. Se encontraron 91 artículos, y después de la lectura exhaustiva de los títulos y resúmenes se seleccionaron ocho publicaciones que abordan, en su mayoría, el uso de los medios de comunicación como estrategia de marketing de la profesión para promover la salud y la imagen profesional transmitida en medios como la internet y televisión. El tema es poco explorado en la investigación en enfermería, pero hay perspectivas de avance por el interés que despierta y por el reconocimiento creciente de su importancia.

Abstract: Este estudo teve como objetivo determinar a influência da obesidade infantil no desenvolvimento da hipertens o arterial do adulto. Fizeram parte da amostra 46 usuários da Unidade de Saúde de um município paulista atendidos no período de setembro de 2008 a fevereiro de 2009. A classifica o do estado nutricional infantil foi obtida por meio do registro do percentil. A press o arterial foi mensurada por método indireto, obedecendo às especifica es das Diretrizes Brasileiras de Hipertens o. Os valores antropométricos atuais foram obtidos de acordo com as recomenda es da Organiza o Mundial da Saúde. Os dados foram exportados e analisados no programa Statistical Package for Social Science - SPSS, para cálculo defrequências absolutas e porcentagens. Observou-se ocorrência significativa de altera es pressóricas e fatores de risco para adoen a hipertensiva, independente de condi o nutricional na infancia. Estes achados confirmam a importancia do controle eavalia o destes indicadores de risco em toda a popula o.
