RNA Packing Specificity and Folding during Assembly of the Bacteriophage MS2

Abstract: Using a combination of biochemistry, mass spectrometry, NMR spectroscopy and cryo-electron microscopy (cryo-EM), we have been able to show that quasi-equivalent conformer switching in the coat protein (CP) of an RNA bacteriophage (MS2) is controlled by a sequence-specific RNA–protein interaction. The RNA component of this complex is an RNA stem-loop encompassing just 19 nts from the phage genomic RNA, which is 3569 nts in length. This binding results in the conversion of a CP dimer from a symmetrical conformation to an asymmetric one. Only when both symmetrical and asymmetrical dimers are present in solution is assembly of the T = 3 phage capsid efficient. This implies that the conformers, we have characterized by NMR correspond to the two distinct quasi-equivalent conformers seen in the 3D structure of the virion. An icosahedrally-averaged single particle cryo-EM reconstruction of the wild-type phage (to ∼9 Å resolution) has revealed icosahedrally ordered density encompassing up to 90% of the single-stranded RNA genome. The RNA is seen with a novel arrangement of two concentric shells, with connections between them along the 5-fold symmetry axes. RNA in the outer shell interacts with each of the 90 CP dimers in the T = 3 capsid and although the density is icosahedrally averaged, there appears to be a different average contact at the different quasi-equivalent protein dimers: precisely the result that would be expected if protein conformer switching is RNA-mediated throughout the assembly pathway. This unprecedented RNA structure provides new constraints for models of viral assembly and we describe experiments aimed at probing these. Together, these results suggest that viral genomic RNA folding is an important factor in efficient assembly, and further suggest that RNAs that could sequester viral CPs but not fold appropriately could act as potent inhibitors of viral assembly.
Exploring Proteins in Anopheles gambiae Male and Female Antennae through MALDI Mass Spectrometry Profiling

Abstract: MALDI profiling and imaging mass spectrometry (IMS) are novel techniques for direct analysis of peptides and small proteins in biological tissues. In this work we applied them to the study of Anopheles gambiae antennae, with the aim of analysing expression of soluble proteins involved in olfaction perireceptor events. MALDI spectra obtained by direct profiling on single antennae and by the analysis of extracts, showed similar profiles, although spectra obtained through profiling had a richer ion population and higher signal to noise ratio. Male and female antennae showed distinct protein profiles. MALDI imaging experiments were also performed and differences were observed in the localization of some proteins. Two proteins were identified through high resolution measurement and top-down MS/MS experiments. A 8 kDa protein only present in the male antennae matched with an unannotated sequence of the An. gambiae genome, while the presence of odorant binding protein 9 (OBP-9) was confirmed through experiments of 2-DE, followed by MS and MS/MS analysis of digested spots. This work shows that MALDI MS profiling is a technique suitable for the analysis of proteins of small and medium MW in insect appendices, and allows obtaining data for several specimens which can be investigated for differences between groups. Proteins of interest can be identified through other complementary MS approaches.

Environmental Impact of the Construction and Use of a House: Assessment of Building Materials and Electricity End-Uses in a Residential Area of the Province of Norte de Santander, Colombia

Abstract: the objective of this research is to assess the environmental impacts associated with the construction and use of a residential house. building materials and electricity end-uses were assessed. the study was used to highlight some initiatives for the residential-building industrial sector and also some of the aspects that should be considered when trying to improve sustainability indicators. as a study case, a colombian dwelling located in the city of pamplona was chosen. the findings of this research showed that the highest environmental impacts during the dwelling's life cycle take place during the operation phase of the dwelling, while construction phase generates about 7-28% of the impact. transportation and maintenance combined account for less than 10% of the life cycle impacts.

Abstract: This paper aimed to explore the literature on security issues that digital libraries should consider in managing digital resources. Books on information security and network security were consulted as well as several databases such as ERIC, Ebrary, LISA, Science Direct, EbscoHost, ISI, Google Scholar, ProQuest, Emerald Insight, ACM were searched to understand what particular aspect of information security and privacy in digital libraries exist from 2000 - 2010. Security in digital libraries is an issue of the most important, and should be considered carefully in creating policies and strategic plans of institutions wanting to set up a digital library. This paper focused on the four main streams that concerns security in the digital environment, namely: infrastructure, digital content, users and standards and legal issues. Abstract: This work proposes a 3D Virtual World environment and a didactic experience for training young students in an environment capable of supporting the engineering practices based on technical drawing. The main difficulty of technical drawing consists in representing a 3D object on a 2D medium. This restriction imposes to human mind to be able to summarize the spatial properties of objects on the paper. The proposed system trains these capabilities by requiring students to build, in the simulated environment, simple objects represented with 2D drawings. In this way, the students are not only pushed to move themselves between different dimensionality spaces, but also they benefit of the 3D spaces for moving and exploring the models they are building. An empirical evaluation, conducted as a controlled experiment, has provided enthusiastic results in terms of user performances and impressions.
Limited Bandwidths and Correlation Ambiguities: Do They Co-Exist in Galileo Receivers  [PDF]

Abstract: Galileo is the Global Navigation Satellite System that Europe is building and it is planned to be operational in the next 3-5 years. Several Galileo signals use split-spectrum modulations, such as Composite Binary Offset Carrier (CBOC) modulation, which create correlation ambiguities when processed with large or infinite front-end bandwidths (i.e., in wideband receivers). The correlation ambiguities refer to the notches in the correlation shape (i.e., in the envelope of the correlation between incoming signal and reference modulated code) which happen within +/– 1 chip from the main peak. These correlation ambiguities affect adversely the detection probabilities in the code acquisition process and are usually dealt with by using some form of unambiguous processing (e.g., BPSK-like techniques, sideband processing, etc.). In some applications, such as mass-market applications, a narrowband Galileo receiver (i.e., with considerable front-end bandwidth limitation) is likely to be employed. The question addressed in this paper, which has not been answered before, is whether or not this bandwidth limitation can cope inherently with the ambiguities of the correlation function, to which extent, and which the best design options are in the acquisition process (e.g., in terms of time-bin step and ambiguity mitigation mechanisms).
Efficacy and tolerability of propolis special extract gh 2002 as a lip balm against herpes labialis: a randomized, double-blind three-arm dose finding study  [PDF]

Abstract: A dose-finding study was performed with respect to the clinical applicability and tolerability of three different concentrations of propolis special extract GH 2002 in a lip balm (0.1%, 0.5% and 1%). The trial was designed as a double-blind, randomized dermatological study in 150 outpatients with Herpes labialis. The primary parameter was the duration in days until painless incrustation in 50% or 90% of the patients (observable in 121 patients). Secondary parameters were local pain (assessed on a visual analogue scale), itching, burning and tension/ swelling on a verbal rating scale, and tolerability. Visits were performed on days 2/3, 5/6 and 8/9. Best efficacy results with shortest healing time (3.4 and 5.4 days in the 50th and 90th percentile, respectively; p = 0.008 vs. 1% and 0.09 vs. 0.1%) and good tolerability were observed with the 0.5% concentration. All three concentrations achieved highly significant therapeutic results in comparison with baseline values (p < 0.0005) for all secondary parameters as early as day 2/3. Analgesia was the most prominent effect for the patients. Conclusion: The 0.5 % concentration of propolis special extract GH 2002 in a lip balm was found to have the best risk-benefit ratio for the treatment of Herpes labialis.
Analysis on Long Precipitation Series in Piedmont (North-West Italy)  [PDF]

Abstract:

This study analyses thirteen daily precipitation series of Piedmont, region of North-West Italy. The meteorological series have been chosen because they were meteorological observatories operating continuously from the beginning of 1900 until 2011. As the first step an historical research over each station has been carried out. In this way, the potential breaks, in the series, either due to changes of locations or instruments, have been determined and the missing values have been recovered. On the precipitation daily series a quality control have been effectuated and by metadata identification it was possible to assess the homogeneity of the meteorological series. In this way we have obtained the complete and correct series on which trends have been computed. In order to better understand the consequences of climate variations on our environment and society, we have calculated the climate indices proposed by “CCL/CLIVAR Working Group on Climate Change Detection” (dry and wet days, rainy days, intensity of precipitation…) over the time. The values of precipitation have also allowed beginning the climatic analysis with the aim at defining the principle local peculiarity in Piedmont.
