Characterization of New Otic Enhancers of the Pou3f4 Gene Reveal Distinct Signaling Pathway Regulation and Spatio-Temporal Patterns

Abstract: POU3F4 is a member of the POU-homedomain transcription factor family with a prominent role in inner ear development. Mutations in the human POU3F4 coding unit leads to X-linked deafness type 3 (DFN3), characterized by conductive hearing loss and progressive sensorineural deafness. Microdeletions found 1 Mb 5′ upstream of the coding region also displayed the same phenotype, suggesting that cis-regulatory elements might be present in that region. Indeed, we and others have recently identified several enhancers at the 1 Mb 5′ upstream interval of the pou3f4 locus. Here we characterize the spatio-temporal patterns of these regulatory elements in zebrafish transgenic lines. We show that the most distal enhancer (HCNR 81675) is activated earlier and drives GFP reporter expression initially to a broad ear domain to progressively restrict to the sensory patches. The proximal enhancer (HCNR 82478) is switched later during development and promotes expression, among in other tissues, in sensory patches from its onset. The third enhancer (HCNR 81728) is also active at later stages in the otic mesenchyme and in the otic epithelium. We also characterize the signaling pathways regulating these enhancers. While HCNR 81675 is regulated by very early signals of retinoic acid, HCNR 82478 is regulated by Fgf activity at a later stage and the HCNR 81728 enhancer is under the control of Hh signaling. Finally, we show that Sox2 and Pax2 transcription factors are bound to HCNR 81675 genomic region during otic development and specific mutations to these transcription factor binding sites abrogates HCNR 81675 enhancer activity. Altogether, our results suggest that pou3f4 expression in inner ear might be under the control of distinct regulatory elements that fine-tune the spatio-temporal activity of this gene and provides novel data on the signaling mechanisms controlling pou3f4 function.
Organogénesis directa in vitro a partir de hojas de la planta Antiplasmodial Solanum nudum Dunal

Abstract: solanum nudum dunal (solanaceae) is a plant with a potential for developing chemotherapeutic treatments against malaria. this is the first report of a fast, efficient, and reproducible direct organogenesis protocol from leave segments from in vitro seed-grown plantlets. leaves segments from 5 months old germinated plantlets were placed on half concentration murashige and skoog medium (ms), supplemented with several concentrations of bencilaminopurin (bap) combinated with indolacetic acid (iaa). dark/ light incubation effect in periods 0/45, 15/30 and 30/15 dark/light days were evaluated on the buds induction. high frequency buds formation was shown (4,83) in explants cultured on ms supplemented with bap 2,0 mg/l and aia 0,1 mg/l under a period of 30 days of dark condition incubation. after induction, buds obtained were transferred to ms medium supplemented with gibberellic acid (ga3) 0,3 mg/l and maintained under artificial cool light, there the plantlets rooted. regenerated plantlets were placed under greenhouse conditions and these were morphologically similar to donor plants.
Intensive culture of Litopenaeus vannamei without water exchange and with an artificial substrate

Abstract: aiming to determine the effect of the periphyton growing on artificial substrates, juveniles (3 g initial weight and 440 g m-3 stocking biomass) of the whiteleg shrimp litopenaeus vannamei (boone, 1931) , were grown during 32 days in eight 1 m3 cylindrical tanks with 3.7 m2 of total submerged surface. two culture treatments (with and without artificial substrate or control) were tested with four replicates each. artificial substrate (aquamatstm) provided an additional surface area of 7.2 m2. the mean dissolved ammonium (nh4+) and ammonia (nh3) concentrations for the aquamats group were 39 and 22% lower than the respective values obtained for the control cultures. the artificial substrate stimulated nutrient recycling among the biological components (shrimp, biofilm, bottom microfauna, etc.) since mean shrimp biomass yield was 13% higher for the aquamats group, and it contained a significantly higher percentage of the total nitrogen and phosphorus inputs than the control treatment. the protein content of shrimp cultured with aquamats was 21.4% higher than that obtained for the control group, which is explained by the higher availability (and diversity) of the natural food of the periphyton. in view of these results, the use of closed cultures added this artificial substrate seems a viable alternative for shrimp culture.

Abstract: Título en inglés: Direct organogenesis in vitro from leaves of Solanum nudum Dunal – Antiplasmodial plant Resumen Solanum nudum Dunal (Solanaceae), es una especie vegetal con potencial para desarrollar un tratamiento quimioterapéutico contra la malaria. Este es el primer reporte de un protocolo rápido, eficiente y reproducible de organogénesis directa a partir de segmentos de hoja de plántulas in vitro de esta especie. Los segmentos de hojas de plántulas de 5 meses de germinadas fueron cultivados sobre medio Murashige y Skoog (MS) a mitad de sales y vitaminas, con diferentes concentraciones de Bencilaminopurina (BAP), en combinación con Acido Indolacético (AIA). Se evaluó también el efecto de la iluminación en periodos 0/45, 15/30 y 30/15 días oscuridad/ luz, sobre la inducción de brotes. Se registró un promedio alto de formación de brotes (4,83) en explantes cultivados en medio suplementado con 2,0 mg/L de BAP y 0,1 mg/L de AIA, bajo condición de iluminación por un periodo de 30/15 días oscuridad/luz. Luego de la inducción, los brotes obtenidos fueron transferidos a medio MS suplementado con 0,3 mg/L de Giberelina (GA3), y mantenidos en condiciones de luz donde también enraizaron. Las plántulas regeneradas se llevaron a condiciones de invernadero y fueron morfológicamente similares a las plantas madres. Con el fin de verificar el efecto del perifiton presente en sustratos artificiales sobre la calidad del agua y el reciclaje de nutrientes, se cultivaron en sistemas cerrados durante 32 días juveniles de camarón blanco Litopenaeus vannamei (Boone, 1931) (peso inicial: 3 g y biomasa inicial de 440 g m-3) en ocho estanques cilíndricos de 1 m3 y 3.7 m2 de superficie sumergida total. Se utilizaron dos tratamientos (con o sin sustrato artificial), cada uno con cuatro repeticiones. En las repeticiones de uno de los tratamientos se adicionaron 7.2 m2 de sustrato artificial (AquamatsTM). Los niveles medios de amonio (NH4+) y de amoniaco (NH3) en los cultivos con Aquamats fueron equivalentes al 39% y al 22% respectivamente, de los valores registrados en los cultivos control. El tratamiento con sustrato artificial favoreció el reciclaje de nutrientes, ya que la biomasa final fue 13% superior al control, y su contenido de nitrógeno y fósforo representó porcentajes significativamente mayores de los respectivos ingresos totales. El mayor nivel de proteína en la biomasa de camarón (21.4%) en los tratamientos con sustrato adicional, se explica por la mayor disponibilidad de alimento natural representado por el perifiton. Abstract: the aim of this article is to inquire into the emergence of a new catholic economic ethic, as well as examine whether this ethic differs from that conceptualized by max weber as the protestant ethic. the theoretical starting point is weber's classic work the protestant ethic and the spirit of capitalism, in which weber argued that catholic entrepreneurs only participated marginally in the development of modern capitalism. early, many catholic intellectuals refused any merits of the capitalist system, particularly in its liberal version. the market economy has commonly been associated with individualism, utilitarianism, and egotism. the late pope john paul ii, as head of the vatican state, was active in promoting a change of ethical attitude in the face of capitalism, particularly through the centesimus annus encyclical. this encyclical attempts to formulate a bridge between catholic thinking and capitalism, by accepting, and even promoting, important aspects of capitalism, albeit a capitalism imbued by a new catholic economic ethic.
