Immunopathological Roles of Cytokines, Chemokines, Signaling Molecules, and Pattern-Recognition Receptors in Systemic Lupus Erythematosus

Abstract: Systemic lupus erythematosus (SLE) is an autoimmune disease with unknown etiology affecting more than one million individuals each year. It is characterized by B- and T-cell hyperactivity and by defects in the clearance of apoptotic cells and immune complexes. Understanding the complex process involved and the interaction between various cytokines, chemokines, signaling molecules, and pattern-recognition receptors (PRRs) in the immune pathways will provide valuable information on the development of novel therapeutic targets for treating SLE. In this paper, we review the immunopathological roles of novel cytokines, chemokines, signaling molecules, PRRs, and their interactions in immunoregulatory networks and suggest how their disturbances may implicate pathological conditions in SLE.
Down-Regulated NOD2 by Immunosuppressants in Peripheral Blood Cells in Patients with SLE Reduces the Muramyl Dipeptide-Induced IL-10 Production

Abstract: Background Pattern recognition receptors (PRRs) such as Toll-like receptors are aberrantly expressed of peripheral blood mononuclear cells (PBMCs) in systemic lupus erythematosus (SLE) patients, for playing immunopathological roles. Methodology/Principal Findings We investigated the expression and function of the PRR nucleotide-binding oligomerization domain (NOD2) in SLE. NOD2 expression in T, B lymphocytes, monocytes, myeloid dendritic cells (mDCs) and plasmacytoid dendritic cells (pDCs) was assessed in SLE patients and healthy controls (HCs) using flow cytometric analysis. Ex vivo production of cytokines from PBMCs upon NOD2 agonist muramyl dipeptide (MDP) stimulation was assessed using Cytometric Bead Array. Over-expression of NOD2 in monocytes was observed in immunosuppressant na?ve SLE patients, and was positively associated with longer disease duration. Immunosuppressive therapy was an independent explanatory variable for downregulating NOD2 expression in CD8+ T, monocytes, mDCs and pDCs. Ex vivo basal productions of cytokines (IL-6, IL-8 and IL-10) were significantly increased in immunosuppressant na?ve patients and patients with active disease despite immunosuppressants compared with HCs. Upon MDP stimulaiton, relative induction (%) of cytokines (IL-1β) from PBMC was significantly increased in immunosuppressant na?ve patients with inactive disease, and patients with active disease despite immunosuppressant treatment compared with HCs. Immunosuppressant usage was associated with a decreased basal production and MDP induced relative induction (%) of IL-10 in patients with inactive disease compared with immunosuppressant na?ve patients and HCs. Conclusions/Significance Bacterial exposure may increase the NOD2 expression in monocytes in immunosuppressant na?ve SLE patients which can subsequently lead to aberrant activation of PBMCs to produce proinflammatory cytokines, implicating the innate immune response for extracellular pathogens in the immunopathological mechanisms in SLE. Immunosuppressant therapy may downregulate NOD2 expression in CD8+ T lymphocytes, monocytes, and DCs in SLE patients which subsequently IL-10 reduction, contributing towards the regulation of immunopathological mechanisms of SLE, at the expense of increasing risk of bacterial infection.

Antagonist-mediated down-regulation of toll-like receptors increases the prevalence of human papillomavirus infection in systemic lupus erythematosus

Abstract: Protein levels of TLRs 3, 7, 8 and 9 in cervical epithelial cells of lupus patients and controls with or without HPV infection were assessed using flow cytometry. Characteristics associated with the differential expression of TLRs in systemic lupus erythematosus (SLE) were elucidated. The effect and interferon-stimulated genes (ISGs) (ISG15 and Mx-1) gene expressions were then measured in oncogenic HeLa (HPV18), CaSki (HPV) and C33A (HPV negative) cell lines using flow cytometry and quantitative real-time PCR. Ex vivo productions of cytokines and interferon-gamma (IFN-γ) upon TLR ligands stimulations were subsequently measured using cytometric bead array and ELISA.For subjects with HPV infection, levels of TLR3 and TLR7 were significantly lower in lupus patients compared with controls. Significantly decreased TLRs 7, 8 and 9 levels were observed in HPV-negative SLE compared to healthy controls. For SLE with and without HPV infection, TLR7 and 9 levels were significantly lower in infected SLE than those in HPV-negative patients. Independent explanatory variables associated with down-regulation of TLR7 level included HPV infection and a higher cumulative dose of prednisolone; while a higher cumulative dose of hydroxychloroquine and HPV infection were associated with down-regulation of TLR9 level. In cervical cell lines, TLRs 3, 7, 8, 9 protein levels and antiviral ISG15 and Mx-1 gene expressions were inhibited in two oncogenic HPV types. Functional data showed that the induction of pro-inflammatory cytokines by TLR ligands (R837, ssRNA and ODN2395) was greatly impaired in CaSki and HeLa than C33A cells.In conclusion, prednisolone and TLR antagonist (hydroxychloroquine) may down-regulate protein levels of TLR7 and TLR9 in lupus patients, thereby decreasing the innate immune response against HPV infection. Upon infection, HPV further down-regulate TLR7 and 9 levels for viral persistence. Furthermore, reduction of nucleic acid-sensing TLRs 7, 8 and 9 in carcinogenic HPVs

Abstract: Effects of the multilayer heterostructures as the feedback end facets and quantum well structure as the active medium on the lasing behavior in the vertical cavity surface emitting laser (VCSEL) have been analyzed theoretically and systematically. It is shown that the degree of phase matching and the effective length of the distributed reflector have prominent effects on the laser characteristics, and the available low threshold VCSEL may not be due to the microcavity effect. The possible ways to improve the design and the device characteristics have been proposed, both for low threshold and simplified device technology, and for satisfying the condition of microcavity effect in VCSEL.

Abstract: 新疆杏产区的光、热、水、土等条件都能很好地满足杏生长、发育的需求,从萌芽到果实成熟期只需≥10℃积温2500℃·d,杏树就能正常开花结果。杏树开花、幼果形成期间的低温霜冻对杏的产量和经济效益有很大影响。天山以北冬季寒冷,杏树越冬保证率低,限制了其发展。用自然灾害风险分析原理,以霜冻灾损率为区划指标,把新疆杏产区划分为4个花期霜冻灾害气候风险区,分析了各区的霜冻发生频率和冻害程度以及发展杏产业的利弊条件。

Abstract: 以不同品种桃(Amygdlus persica(L.)Batsch)"大久保(Okubao)"和"丽春(Lichun)"为试材,研究了不同土壤镉水平下桃树对镉及矿质养分的吸收.结果表明:随着镉浓度增大,品种间镉、大量元素、微量元素的含量差异显著(P<0.05);桃树器官中镉积累量为根>茎>叶>果实;土壤中镉浓度为10 mg·kg-1时,大久保外果皮中镉积累量最大,较对照增加了226.78%,而丽春桃果实组织中镉积累量在处理前后没有明显变化;当土壤镉浓度超过20 mg·kg-1时,2品种均不能正常座果.土壤镉浓度为100 mg·kg-1,胁迫时间为9个月后,大久保桃树体死亡;土壤Cd浓度<20 mg·kg-1时促进大量元素K、N、Ca、Mg 和微量元素Zn、Cu、Mn、Fe吸收,>20 mg·kg-1时抑制其吸收,但丽春根和大久保茎中Fe 元素含量呈现持续上升趋势,且Cd 100 mg·kg-1时Fe的吸收值达最大.
Effects of exogenous betaine on physiological responses of peach tree under water stress

Abstract: With four-year-old potted Prunus persica L. cv. Qingfeng as test material, this paper studied the change pattern of its leaf betaine content under water stress, and its physiological responses under effects of foliage-spraying exogenous betaine. The results showed that under normal water supply, the betaine content in Qingfeng' s leaf was 75.9-80.5 microg x g(-1) FM, which was increased with increasing water stress, and up to 278.9 microg x g(-1) FM on the 16th day after cutting off the water supply. The leaf plasma membrane permeability was 8.06% - 8.61% under normal water supply, but increased to 28.62% under water stress. When 100 and 500 mg x L(-1) of betaine were applied exogenously, the plasma membrane permeability was 26.25% and 21.79% after 16 days, respectively. The hydrogen peroxide (H2O2) content increased from 27.2-32.5 micromol g(-1) FM to 76.4 micromol x g(-1) FM in the course of water stress, and decreased to 73.2 and 68.5 micromol x g(-1) FM after spraying 100 and 500 mg betaine x L(-1), respectively. During the period of intensified water stress, the peak value of ascorbate peroxidase (AsA-POD) activity was 0.435 mg x g(-1) FM, and up to 0.490 mg x g(-1) FM when treated with exogenous betaine. When the peach tree was subjected to water stress, the contents of free proline and soluble sugar accumulated dramatically, but produced on approximately decrease in 500 mg x L(-1) endogenous betaine application on the 16th day which was slightly less than that of control and 100 mg x L(-1) betaine application. There was a gradual decline in the content of soluble protein under water stress, and an increment of 20. 3% was observed when betaine was applied exogenously. These results strongly suggested that foliage-spraying exogenous betaine could increase the drought resistance of peach tree through decreasing its leaf plasma membrane permeability and H2O2, free proline and soluble sugar contents and increasing its leaf AsA-POD activity and soluble protein content.
Neuroprotective Effects of Daphnetin against NMDA Receptor-Mediated Excitotoxicity

Abstract: The accumulation of glutamate can excessively activate the N-methyl-d-aspartate (NMDA) receptors and cause excitotoxicity. Daphnetin (Dap), a coumarin derivative, is a protein kinase inhibitor that exhibits antioxidant and neuroprotective properties. However, little is known about the neuroprotective effects of Dap on glutamate-induced excitotoxicity. We evaluated the neuroprotective activities in the primary cultured cortical neurons against NMDA-induced excitotoxicity. Pretreatment with Dap significantly prevented NMDA-induced neuronal cell loss. Dap significantly inhibited the neuronal apoptosis by regulating balance of Bcl-2 and Bax expression. Furthermore, pretreatment of Dap reversed the up-regulation of NR2B-containing NMDA receptors and inhibited the intracellular Ca 2+ overload induced by NMDA exposure. In addition, Dap prevented cerebral ischemic injury in mice induced via a 2 h middle cerebral artery occlusion and a 24 h reperfusion in vivo. The findings suggest that Dap prevents the excitotoxicity through inhibiting the NR2B-containing NMDA receptors and the subsequent calcium overload in cultured cortical neurons.
