Production of proinflammatory cytokines in the human THP-1 monocyte cell line following induction by Tp0751, a recombinant protein of Treponema pallidum

Abstract: The tissue destruction characteristic of syphilis infection may be caused by inflammation due to Treponema pallidum and the ensuing immune responses to the pathogen. T. pallidum membrane proteins are thought to be potent inducers of inflammation during the early stages of infection. However, the actual membrane proteins that induce inflammatory cytokine production are not known, nor are the molecular mechanisms responsible for triggering and sustaining the inflammatory cascades. In the present study, Tp0751 recombinant protein from T. pallidum was found to induce the production of proinflammatory cytokines, including TNF-α, IL-1βand IL-6, in a THP-1 human monocyte cell line. The signal transduction pathways involved in the production of these cytokines were then further investigated. No inhibition of TNF-a, IL-1β, or IL-6 production was observed following treatment with the SAPK/JNK specific inhibitor SP600125 or with an ERK inhibitor PD98059. By contrast, anti-TLR2 mAb, anti-CD14 mAb, and the p38 inhibitor SB203580 significantly inhibited the production of all three cytokines. In addition, pyrrolidine dithiocarbamate (PDTC), a specific inhibitor of NF-κB, profoundly inhibited the production of these cytokines. Tp0751 treatment strongly activated NF-κB, as revealed by Western blotting. However, NF-κB translocation was significantly inhibited by treatment with PDTC. These results indicated that TLR2, CD14, MAPKs/p38, and NF-κB might be implicated in the inflammatory reaction caused by T. pallidum infection.
Evaluation of imaging protocol for ECT based on CS image reconstruction algorithm

Abstract: SPECT (Single-photon Emission Computerized Tomography) and PET (Positron Emission Tomography) are essential medical imaging tools, for which the sampling angle number, scan time should be chosen carefully to compromise between image quality and the radiopharmaceutical dose. In this study, the image quality of different acquisition protocol was evaluated via varied angle number and count number per angle with Monte Carlo simulation data. It was shown that when similar imaging counts were used, the factor of acquisition counts was more important than that of the sampling number in ECT (Emission Computerized Tomography). To further reduce the activity requirement and the scan duration, an iterative image reconstruction algorithm for limited-view and low-dose tomography based on compressed sensing theory has been developed. The total variation regulation was added in the reconstruction process to improve SNR (Signal to Noise Ratio) and reduce the artifacts caused by the limited angle sampling. Maximization of maximum likelihood of the estimated image and the measured data and minimization of the total variation of the image are alternative implemented. By using this advanced algorithm, the reconstruction process is able to achieve image quality matching or exceeding that of normal scan with only half of the injection radiopharmaceutical dose.
Assessment of the immune responses to Treponema pallidum Gpd DNA vaccine adjuvanted with IL-2 and chitosan nanoparticles before and after Treponema pallidum challenge in rabbits

Abstract: Syphilis is a multistage, sexually transmitted disease caused by the spirochete, Treponema pallidum (Tp). A significantly high incidence of syphilis has been reported in several countries, including China, and there is an urgent need for the development of efficacious vaccines against syphilis. DNA vaccines are a major breakthrough in the field of vaccination with several advantages over traditional vaccines. Animal model studies of Tp DNA vaccines have not been reported elsewhere but our previous reports describe the development of a single-gene Tp DNA vaccine and preclinical immunization study. In this study, chitosan (CS) nanoparticles were used as a vector and an interleukin-2 expression plasmid (pIL-2) as an adjuvant to enhance a TpGpd DNA vaccine candidate (pTpGpd) in a rabbit Tp skin challenge model. At week 8 after the first immunization, three rabbits from each group were used to determine cytokine measurements and spleen lymphocyte proliferation assay. pTpGpd in combination with pIL-2 wrapped with CS led to the greatest enhancement of anti-TpGpd antibodies and T-cell proliferation. During infection, levels of anti-TpGpd antibodies and T-cell proliferation were measured. Both the serum special IgG and IL-2, interferon-γ were significantly increased by the co-injection of the IL-2 plasmid compared with the injection of TpGpd DNA alone (P<0.05). Furthermore, IL-2 plasmid coinjection efficiently enhanced the antigen-specific lymphocyte proliferation response. Additionally, the ratios of positive skin lesions and ulcer lesions in groups immunized with pTpGpd were significantly lower than those of the pIL-2, CS or pIL-2 mixed with CS control groups (P<0.001). CS vectored and pIL-2 adjuvanted pTpGpd immunized animals exhibited the lowest rates of positive skin tests (8.33%) and ulcer lesions (4.17%) and the fastest recovery (42 d). These experiments indicate that co-injection of a pIL-2 plasmid with pTpGpd DNA vaccine wrapped with CS can significantly strengthen the long-term stability of immune response during infection, efficiently improve the protective effect against T. pallidum spirochetes infection and attenuate syphilitic lesion development.

Abstract: Pollination biology is the study of the various biological features in relation to the event of pollen transfer. It is one of the central concerns of plant reproductive ecology and evolutionary biology. In this paper, we attempt to introduce the main advances and some new interests in pollination biology and make a brief review of the research work that has been done in China in recent years. We also give some insights into the study that we intend to carry out in this field in the future.

Assume $\{s_k\}_{k=1}^m$ and $\{r_l\}_{l=1}^m$ are unordered singular values of $\mathbf{H}$ and $\mathbf{G}$, respectively, and $s$ and $r$ are randomly selected from $\{s_k\}_{k=1}^m$ and $\{r_l\}_{l=1}^m$, respectively. In this paper, exact analytical closed-form expressions are derived for the joint probability distribution function (PDF) of $\{s_k\}_{k=1}^m$ and $\{r_l\}_{l=1}^m$ using an Itzykson-Zuber-type integral, as well as the joint marginal PDF of $s$ and $r$, by a bi-orthogonal polynomial technique. These PDFs are of interest in multiple-input multiple-output (MIMO) wireless communication channels and systems.
Statistical Properties of Eigen-Modes and Instantaneous Mutual Information in MIMO Time-Varying Rayleigh Channels

Abstract: In this paper, we study two important metrics in multiple-input multiple-output (MIMO) time-varying Rayleigh flat fading channels. One is the eigen-mode, and the other is the instantaneous mutual information (IMI). Their second-order statistics, such as the correlation coefficient, level crossing rate (LCR), and average fade/outage duration, are investigated, assuming a general nonisotropic scattering environment. Exact closed-form expressions are derived and Monte Carlo simulations are provided to verify the accuracy of the analytical results. For the eigen-modes, we found they tend to be spatio-temporally uncorrelated in large MIMO systems. For the IMI, the results show that its correlation coefficient can be well approximated by the squared amplitude of the correlation coefficient of the channel, under certain conditions. Moreover, we also found the LCR of IMI is much more sensitive to the scattering environment than that of each eigen-mode.
On the Second-Order Statistics of the Instantaneous Mutual Information in Rayleigh Fading Channels

Abstract: In this paper, the second-order statistics of the instantaneous mutual information are studied, in time-varying Rayleigh fading channels, assuming general non-isotropic scattering environments. Specifically, first the autocorrelation function, correlation coefficient, level crossing rate, and the average outage duration of the instantaneous mutual information are investigated in single-input single-output (SISO) systems. Closed-form exact expressions are derived, as well as accurate approximations in low- and high-SNR regimes. Then, the results are extended to multiple-input single-output and single-input multiple-output systems, as well as multiple-input multiple-output systems with orthogonal space-time block code transmission. Monte Carlo simulations are provided to verify the accuracy of the analytical results. The results shed more light on the dynamic behavior of the instantaneous mutual information in mobile fading channels.
Studies on plant-pollinator interaction and its significances

Abstract: The majority of flowering plants and crops rely in whole or part on animals for pollination. The mutualism between plants and pollinators has attracted ecologists and evolutionists to use this type of inter-specific interaction as a model system to study species adaptation and diversification since Charles Darwin. Recent debate on the nature of pollination systems call for studies of this interaction at different levels, rang-ing from single species to entire communities in a given area. At the species level, detailed studies suggest that floral traits are under selection from mutualists and antagonists as well as the physical environment. In contrast, studies at community-level are rare, but recent analyses indicate considerable spatial and temporal variation in both generalized and specialized pollination systems. This special issue of Biodiversity Science focuses on plant-pollinator interaction, presenting current research status in this area from China. Papers in-clude floral traits and pollinator behaviors addressed by phenotypic manipulation, estimates of pollen re-moval and receipt, anatomy of flowers, histochemistry analysis and spatial and temporal comparison. The taxa being investigated include wild orchid and cultivated legume, endemic, endangered and invasive species with diverse sexual systems. These thirteen experimental studies and three reviews show the development of pollination biology in China and expose how to facilitate our understanding of the critical ecological proc-esses underlying interspecific interaction in both natural and agricultural ecosystems.

Abstract: 以污水处理站脱水污泥和煤为原料,通过共热解法制备污泥吸附剂,并将其用于吸附含Pb2+的模拟废水.同时,考察了污泥吸附剂制备过程中热解温度、配比、热解时间、粒径对碘吸附值及产率的影响,以及吸附时间、温度、pH值、污泥吸附剂投加量对吸附效果的影响,并对其吸附动力学和热力学特性进行了探讨.结果表明,制备污泥吸附剂的最佳条件为热解温度900 ℃,煤在混合样中的比例为20%,热解时间保持60 min,粒径为10~20目.所制备的污泥吸附剂的碘吸附值为321.62 mg · g-1,产率为44.85%,比表面积为189.23 m2 · g-1.污泥吸附剂对Pb2+的吸附量均随着吸附时间、温度、pH、投加量的增加而增大,并逐渐趋于平衡.污泥吸附剂对Pb2+的吸附动力学比较符合伪二级吸附动力学方程、二阶段吸附动力学方程,颗粒内扩散过程是吸附速率的控制步骤,但不是唯一的速率控制步骤.Langmuir等温方程比Freundlich等温方程更适合于描述此吸附行为.

Abstract: 首先用碱熔融法分解试样,然后加抗坏血酸将试液中的Fe3+还原为Fe2+,再用离子色谱法测定试液中的Fe2+,Mg2+和Ca2+。采用Shim-packIC-Cl阳离子交换柱分离样品离子,以0.25mmol/L乙二胺-0.50mmol/L柠檬酸(pH4.50)为流动相,以CDD-6A电导检测器检测,使被测组分得到满意的分离和检测。Fe,Mg和Ca的加标回收率均在90%以上。测定结果与原子吸收光谱法测定结果相近。
