Aqueous Extract from Hibiscus sabdariffa Linnaeus Ameliorate Diabetic Nephropathy via Regulating Oxidative Status and Akt/Bad/14-3-3γ in an Experimental Animal Model

Abstract: Several studies point out that oxidative stress maybe a major culprit in diabetic nephropathy. Aqueous extract of Hibiscus sabdariffa L. (HSE) has been demonstrated as having beneficial effects on anti-oxidation and lipid-lowering in experimental studies. This study aimed at investigating the effects of Hibiscus sabdariffa L. on diabetic nephropathy in streptozotocin induced type 1 diabetic rats. Our results show that HSE is capable of reducing lipid peroxidation, increasing catalase and glutathione activities significantly in diabetic kidney, and decreasing the plasma levels of triglyceride, low-density lipoprotein (LDL) and increasing high-density lipoprotein (HDL) value. In histological examination, HSE improves hyperglycemia-caused osmotic diuresis in renal proximal convoluted tubules (defined as hydropic change) in diabetic rats. The study also reveals that up-regulation of Akt/Bad/14-3-3γ and NF-κB-mediated transcription might be involved. In conclusion, our results show that HSE possesses the potential effects to ameliorate diabetic nephropathy via improving oxidative status and regulating Akt/Bad/14-3-3γ signaling.
Adrenal Dysfunction in Portal Hypertensive Rats with Acute Hemorrhage

Abstract: Nitric oxide (NO) participates in shock and poorer portal hypotensive effect to vasoconstrictors in portal hypertension with hemorrhage, the so-called splanchnic hyposensitivity. Relative adrenal insufficiency accompanies hemorrhagic shock and is found in liver disease, the ‘hepatoadrenal syndrome’, but the relevant interactions remain unsettled. Portal hypertensive rats were induced by partial portal vein ligation (PVL). Experiments were performed on the 14th day post PVL: (I) ACTH stimulation test for rats without or with hemorrhage; (II) Glypressin response (mean arterial pressure, MAP; portal pressure, PP) in rats (a) without hemorrhage or with hemorrhage, injected with (b) distilled water (DW), (c) dexamethasone 3 mg/kg; (III) To survey the dose-dependent effects of glucocorticoid without being confounded by endogenous adrenal hormone, glypressin response was surveyed in PVL rats with adrenalectomy: (a) without hemorrhage or with hemorrhage, injected with (b) DW; (c) dexamethasone 3 mg/kg; (d) dexamethasone 5 mg/kg. Plasma tumor necrosis factor-α (TNF-α) concentrations and abdominal aorta (AA), superior mesenteric artery (SMA) NO synthases (NOS) mRNA expressions were determined. The results showed that ACTH induced corticosterone release similarly in PVL rats with or without hemorrhage. In bleeding PVL rats, dexamethasone (1) down-regulated AA NOS and enhanced glypressin-induced MAP elevation; (2) did not influence glypressin-induced PP reduction; (3) reduced TNF-α. In bleeding PVL and adrenalectomized rats, high-dose dexamethasone (1) down-regulated AA/SMA NOS; (2) enhanced glypressin-induced MAP elevation and PP reduction; (3) reduced TNF-α. In conclusion, bleeding portal hypertensive rats failed to enhance corticosterone release, suggesting a relative adrenal insufficiency. High-dose dexamethasone reversed systemic hypotension and splanchnic hyporesponsiveness to glypressin in adrenalectomized PVL rats accompanied by TNF-α and NOS down-regulation, suggesting the importance of adequate adrenocorticoid supplement in portal hypertension with hemorrhage and adrenal dysfunction.
Feature Extraction Techniques of Non-Stationary Signals for Fault Diagnosis in Machinery Systems  [PDF]

Abstract: Previously, fault diagnosis of fixed or steady state mechanical failures (e.g., pumps in nuclear power plant turbines, engines or other key equipment) applied spectrum analysis (e.g., fast Fourier transform, FFT) to extract the frequency features as the basis for identifying the causes of failure types. However, mechanical equipment for increasingly instant speed variations (e.g., wind turbine transmissions or the mechanical arms used in 3C assemblies, etc.) mostly generate non-stationary signals, and the signal features must be averaged with analysis time which makes it difficult to identify the causes of failures. This study proposes a time frequency order spectrum method combining the short-time Fourier transform (STFT) and speed frequency order method to capture the order features of non-stationary signals. Such signal features do not change with speed, and are thus effective in identifying faults in mechanical components under non-stationary conditions. In this study, back propagation neural networks (BPNN) and time frequency order spectrum methods were used to verify faults diagnosis and obtained superior diagnosis results in non-stationary signals of gear-rotor systems.
The Association between Genetic Polymorphism and the Processing Efficiency of miR-149 Affects the Prognosis of Patients with Head and Neck Squamous Cell Carcinoma

Abstract: MicroRNAs (miRNAs) play important roles in modulating the neoplastic process of cancers including head and neck squamous cell carcinoma (HNSCC). A genetic polymorphism (rs2292832, C>T) has been recently identified in the precursor of miR-149; nevertheless its clinicopathological implications remain obscure. In this study, we showed that miR-149 is down-regulated in HNSCC compared to normal mucosa and this is associated with a poorer patient survival. In addition, HNSCC patients with the T/T genotype have more advanced tumors and a worse prognosis. Multivariate analysis indicated that patients carried the T/T genotype have a 2.81-fold (95% CI: 1.58–4.97) increased risk of nodal metastasis and 1.66-fold (95% CI: 1.05–2.60) increased risk of mortality compared to other groups. T/T genotype also predicted the worse prognosis of buccal mucosa carcinoma subset of HNSCC. In vitro analysis indicated that exogenous miR-149 expression reduces the migration of HNSCC cells. Moreover, HNSCC cell subclones carrying the pri-mir-149 sequence containing the T variant show a low processing efficacy when converting the pre-mir-149 to mature miR-149. These findings suggest that miR-149 suppresses tumor cell mobility, and that the pre-mir-149 polymorphism may affect the processing of miR-149, resulting in a change in the abundance of the mature form miRNA, which, in turn, modulates tumor progression and patient survival.
Diabetes Diminishes the Portal-Systemic Collateral Vascular Response to Vasopressin via Vasopressin Receptor and Gα Proteins Regulations in Cirrhotic Rats

Abstract: Liver cirrhosis may lead to portal-systemic collateral formation and bleeding. The hemostatic effect is influenced by the response of collateral vessels to vasoconstrictors. Diabetes and glucose also influence vasoresponsiveness, but their net effect on collaterals remains unexplored. This study investigated the impact of diabetes or glucose application on portal-systemic collateral vasoresponsiveness to arginine vasopressin (AVP) in cirrhosis. Spraque-Dawley rats with bile duct ligation (BDL)-induced cirrhosis received vehicle (citrate buffer) or streptozotocin (diabetic, BDL/STZ). The in situ collateral perfusion was done after hemodynamic measurements: Both were perfused with Krebs solution, D-glucose, or D-glucose and NaF, with additional OPC-31260 for the BDL/STZ group. Splenorenal shunt vasopressin receptors and Gα proteins mRNA expressions were evaluated. The survival rate of cirrhotic rats was decreased by STZ injection. The collateral perfusion pressure changes to AVP were lower in STZ-injected groups, which were reversed by OPC-31260 (a V2R antagonist) and overcome by NaF (a G protein activator). The splenorenal shunt V2R mRNA expression was increased while Gα proteins mRNA expressions were decreased in BDL/STZ rats compared to BDL rats. The Gαq and Gα11 mRNA expressions also correlated with the maximal perfusion pressure changes to AVP. Diabetes diminished the portal-systemic collateral vascular response to AVP in rats with BDL-induced cirrhosis, probably via V2 receptor up-regulation and Gα proteins down-regulation.

Abstract: In the title dioxidomolybdenum(VI) complex, [Mo(C17H16N2O5)O2(CH3OH)], the MoVI atom is coordinated by the phenolate O, imine N and enolic O atoms of the tridentate hydrazone ligand, one methanol O atom, and two oxide O atoms, forming a distorted octahedral coordination geometry. The oxide O atoms adopt a cis conformation: one is trans to the methanol O atom and the other is trans to the ligand N atom. The dihedral angle between the two benzene rings in the hydrazone ligand is 4.0 (3)°. In the crystal, molecules are linked by O—H...N and O—H...O hydrogen bonds.

The Movement Strategy in Taiwan’s Judicial Independence Reform

Abstract: Judicial independence reform in Taiwan was pioneered by a group of reform-spirited judges from Room 303 of the Taichung (台中, Taizhong) District Court, in 1993. Rather than joining the mass movement that was unfolding on the streets, the reformers formed a coalition with other judges to trigger reform from within the judiciary. The reformers appealed to the rule of law and democracy as a movement strategy for mobilization. As a result, the movement strategy turned out to be a great success, and Room 303 became the chief engine for further judicial reforms in subsequent years. However, the movement strategy in itself also presents some limitations. This paper examines why the movement strategy was successful and how its limitations eventually created problems that hinder further judicial reforms in Taiwan.
University Student Sample Is Unable to Accurately Assess Their Calorie Needs: Implications for Weight Management and Menu Labeling  [PDF]

Abstract: National surveys have shown that over 80% of adults do not know their recommended calorie levels. Lack of knowledge about calorie needs could be contributing to the high prevalence of obesity in the US. Young adulthood is a crucial period for the development of dietary behaviors that continue into later adulthood and influence the risk of obesity and chronic disease. This study examined university students’ knowledge of their recommended calorie needs. Subjects (N = 153) were students at Texas Tech University in Fall 2010. Students were given a survey to assess perceived daily calorie need (PDCN) and perceived daily calorie intake (PDCI). Their recommended daily calorie needs (RDCN) and actual calorie intakes (ACI) were determined using MyPyramid.gov. PDCN, PDCI, RDCN, and ACI were compared to determine students’ ability to accurately estimate and consume recommended daily calorie levels. The range of their PDCN was 120 kcal to 10,000 kcal. Only 19.7% of students estimated their RDCN accurately. There were significant differences between PDCN and RDCN (t [152] = ?3.223, P = 0.002); PDCI and ACI (t [114] = 3.246, P = 0.002); and ACI and RDCN (t [114] = ?5.6, P = 0.000). Nearly 40% of these university students were overweight. BMI had a significant effect (P = 0.001) on students’ estimation of their RDCN as students with underweight/normal BMI were more accurate. Nutrition education programs focused on calorie needs should be implemented with university students so they will be able to effectively use calorie information on food labels and menus for weight management.
