Formate Excretion in Urine of Rats Fed Dimethylaminoazobenzene-rich Diets: The Possibility of Formate Formation from D-Lactate

Abstract: This experiment was carried out to evaluate the possibility of degradation of d-lactate into formate and acetaldehyde. In order to induce hyperproduction of d-lactate in rats. Donryu male albino rats were fed diets containing 0.064% 3'-methyl-4-dimethylaminoazobenzene (3'-MDAB), 4'-methyl-4-dimethylaminoazobenzene (4'-MDAB) or 2-methyl-4-dimethylaminoazobenzene (2-MDAB) for 10 weeks. During the experiment, body mass, food and water intake and volume of urine were documented. Methylglyoxal, d-lactate and formate in the urine samples were determined. On the first day of the eleventh week, methylglyoxal, d-lactate, glutathione and enzymatic activities of demethylation and glyoxalase I and II in liver were measured. Methylglyoxal, d-lactate and clinical chemistry parameters of blood plasma were also measured. The levels of methylglyoxal and d-lactate in livers of rats fed 3'-MDAB were very high, while those of 2-MDAB fed-rats and the control group were the same. The fact that glyoxalase I activity and the level of glutathione, a cofactor of glyoxalase I, were high in the livers of the 3'-MDAB-fed rats can explain the elevated levels of methylglyoxal and d-lactate in the liver. The most striking results were the elevated formate levels in the urine of rats fed 3'- and 4'-MDAB in a precancerous state. The degradation of d-lactate, an end product of the methylglyoxal bypass, into acetaldehyde and formate was suggested as a possible way to explain the results.
Comparative studies of serum-free media and detection techniques for in vitro drug sensitivity assessment of Plasmodium falciparum  [PDF]

Abstract: Malaria continues to be a devastating disease. In a previous study, we formulated a chemically defined culture medium that is able to sustain the complete intraerythrocytic growth of Plasmodium falciparum. We tested the feasibility of using the medium (CDRPMI) as well as human serum-free media enriched with commercially available human-serum substitutes (GFSRPMI and ALBRPMI) to assess the drug sensitivity of P. falciparum, using chloroquine diphosphate (CQ) and dihydroartemisinin (DHART) as conventional antimalarial drugs. Growth inhibition was measured by four different methods: flow cytometry with SYBR Green I (FCM), microscopy (Giemsa method), enzymatic estimation of parasite lactate dehydrogenase (pLDH), and histidine-rich protein 2 (HRPII) determination. In drug sensitivity tests on asynchronous parasites cultured for 96 h in the presence of drugs, the dose-response curves were similar and differences in the 50% growth inhibition concentrations for the drugs, which were estimated by the four methods, were not statistically significant for the three culture media. The effect of the drugs on the growth of synchronous parasites at the ring stage was also assessed in micro-volume tests by three different methods of FCM: tracking fluorescent erythrocytes, schizont test, and merozoite test. Dose-response curves for the drugs were similar, and differences in the 50% growth inhibition concentrations were not statistically significant for CDRPMI and GFSRPMI. Thus CDRPMI as well as GFSRPMI and ALBRPMI can be similarly useful media for drug sensitivity testing of P. falciparum. The FCM, pLDH and HRPII estimations were fast and reliable detection methods, with FCM allowing schizont and merozoite tests to be performed with shorter periods of culture.

Abstract: Hepatocellular carcinoma (HCC) is difficult to manage due to the high frequency of post-surgical recurrence. Early detection of the HCC recurrence after liver resection is important in making further therapeutic options, such as salvage liver transplantation. In this study, we utilized microRNA expression profiling to assess the risk of HCC recurrence after liver resection.
Suppression of Adiponectin by Aberrantly Glycosylated IgA1 in Glomerular Mesangial Cells In Vitro and In Vivo

Abstract: The pathogenesis of IgA nephropathy (IgAN) may be associated with the mesangial deposition of aberrantly glycosylated IgA1. To identify mediators affected by aberrantly glycosylated IgA1 in cultured human mesangial cells (HMCs), we generated enzymatically modified desialylated and degalactosylated (deSial/deGal) IgA1. The state of deglycosylated IgA1 was confirmed by lectin binding to Helix aspersa (HAA) and Sambucus nigra (SNA). In the cytokine array analysis, 52 proteins were upregulated and 34 were downregulated in HMCs after stimulation with deSial/deGal IgA1. Among them, the secretion of adiponectin was suppressed in HMCs after stimulation with deSial/deGal IgA1. HMCs expressed mRNAs for adiponectin and its type 1 receptor, but not the type 2 receptor. Moreover, we revealed a downregulation of adiponectin expression in the glomeruli of renal biopsy specimens from patients with IgAN compared to those with lupus nephritis. We also demonstrated that aberrantly glycosylated IgA1 was deposited in the mesangium of patients with IgAN by dual staining of HAA and IgA. Moreover, the urinary HAA/SNA ratio of lectin binding was significantly higher in IgAN compared to other kidney diseases. Since adiponectin has anti-inflammatory effects, including the inhibition of adhesion molecules and cytokines, these data suggest that the local suppression of this adipokine by aberrantly glycosylated IgA1 could be involved in the regulation of glomerular inflammation and sclerosis in IgAN.
A case of focal segmental glomerulosclerosis in an adult patient with hypogammaglobulinemia superimposed on membranoproliferative glomerulonephritis in childhood

Abstract: We report a 36-year-old man who had suffered membranoproliferative glomerulonephritis (MPGN) in his childhood, later diagnosed with CVID at 35?years of age. He presented at our hospital with signs of proteinuria. A renal biopsy revealed he suffered from focal segmental glomerulosclerosis (FSGS), possibly due to obesity and hypertension, not CVID - associated MPGN.This is the first case report of FSGS in a CVID patient. In this case, we have to pay attention not only to the treatment of obesity and hypertension for FSGS but also to the recurrence of immune-complex glomerulonephritis such as MPGN, in case of the restoration of hypogammaglobulinemia.

Abstract: Background Male germ cell RacGTPase activating protein (MgcRacGAP) is an important regulator of the Rho family GTPases — RhoA, Rac1, and Cdc42 — and is indispensable in cytokinesis and cell cycle progression. Inactivation of RhoA by phosphorylated MgcRacGAP is an essential step in cytokinesis. MgcRacGAP is also involved in G1-S transition and nuclear transport of signal transducer and activator of transcription 3/5 (STAT3/5). Expression of MgcRacGAP is strictly controlled in a cell cycle-dependent manner. However, the underlying mechanisms have not been elucidated. Methodology/Principal Findings Using MgcRacGAP deletion mutants and the fusion proteins of full-length or partial fragments of MgcRacGAP to mVenus fluorescent protein, we demonstrated that MgcRacGAP is degraded by the ubiquitin-proteasome pathway in the late M to G1 phase via APCCDH1. We also identified the critical region for destruction located in the C-terminus of MgcRacGAP, AA537–570, which is necessary and sufficient for CDH1-mediated MgcRacGAP destruction. In addition, we identified a PEST domain-like structure with charged residues in MgcRacGAP and implicate it in effective ubiquitination of MgcRacGAP. Conclusions/Significance Our findings not only reveal a novel mechanism for controlling the expression level of MgcRacGAP but also identify a new target of APCCDH1. Moreover our results identify a C-terminal region AA537–570 of MgcRacGAP as its degron.
Solvability of the Economic Input-Output Equation by Time Irreversibility  [PDF]

Abstract:
This paper reinterprets the economic input-output equation as a description of a realized situation without considering decision making. This paper uses the equation that the self-sufficiency rate is added to the Leontief type, and discusses its solvability. The equation has a unique solution if and only if each part of the relevant society satisfies the space-time openness condition. This condition means that commodities which a part of the relevant society possesses are not all inputted to its inside. Moreover, if the process of input and output is time irreversible, each part of the relevant society satisfies the space-time openness condition. Therefore, the solvability of the equation is guaranteed by time irreversibility. This proposition seems to be relevant to the grandfather paradox which is a type of time paradox.

Abstract:
A real square matrix whose non-diagonal elements are non-positive is called a Z-matrix. This paper shows a necessary and sufficient condition for non-singularity of two types of Z-matrices. The first is for the Z-matrix whose row sums are all non-negative. The non-singularity condition for this matrix is that at least one positive row sum exists in any principal submatrix of the matrix. The second is for the Z-matrix \"\"which satisfies \"\"where \"\" . Let \"\" be the ith row and the jth column element of \"\", and \"\" be the jth element of \"\". Let \"\"be a subset of \"\"which is not empty, and \"\" be the complement of \"\" if \"\" is a proper subset. The non-singularity condition for this matrix is \"\"such that \"\"or \"\"such that \"\" for \"\" . Robert Beauwens and Michael Neumann previously presented conditions similar to these conditions. In this paper, we present a different proof and show that these conditions can be also derived from theirs.
Money Circulation Equation Considering Time Irreversibility  [PDF]

Abstract: The essence of money circulation is that money continues to transfer among economic agents eternally. Based on this recognition, this paper shows a money circulation equation that calculates the quantities of expenditure, revenue, and the end money from the quantity of the beginning money. The beginning money consists of the possession at term beginning, production and being transferred from the outside of the relevant society. The end money consists of the possession at term end, disappearance and transferring to the outside of the relevant society. This equation has a unique solution if and only if each part of the relevant society satisfies the space-time openness condition. Moreover, if money is transferred time irreversibly, each part of the relevant society satisfies the space-time openness condition. Hence, the solvability of the equation is guaranteed by time irreversibility. These solvability conditions are similar to those of the economic input-output equation, but the details are different. An equation resembling our money circulation equation was already shown by Mária Augustinovics, a Hungarian economist. This paper examines the commonalities and differences between our equation and hers. This paper provides the basis for some intended papers by the author.
