Anti-inflammatory activity of edible oyster mushroom is mediated through the inhibition of NF-κB and AP-1 signaling

Abstract: RAW264.7 murine macrophage cell line and murine splenocytes were incubated with the oyster mushroom concentrate (OMC, 0-100 μg/ml) in the absence or presence of lipopolysacharide (LPS) or concanavalin A (ConA), respectively. Cell proliferation was determined by MTT assay. Expression of cytokines and proteins was measured by ELISA assay and Western blot analysis, respectively. DNA-binding activity was assayed by the gel-shift analysis. Inflammation in mice was induced by intraperitoneal injection of LPS.OMC suppressed LPS-induced secretion of tumor necrosis factor-α (TNF-α, interleukin-6 (IL-6), and IL-12p40 from RAW264.7 macrophages. OMC inhibited LPS-induced production of prostaglandin E2 (PGE2) and nitric oxide (NO) through the down-regulation of expression of COX-2 and iNOS, respectively. OMC also inhibited LPS-dependent DNA-binding activity of AP-1 and NF-κB in RAW264.7 cells. Oral administration of OMC markedly suppressed secretion of TNF-α and IL-6 in mice challenged with LPS in vivo. Anti-inflammatory activity of OMC was confirmed by the inhibition of proliferation and secretion of interferon-γ (IFN-γ), IL-2, and IL-6 from concanavalin A (ConA)-stimulated mouse splenocytes.Our study suggests that oyster mushroom possesses anti-inflammatory activities and could be considered a dietary agent against inflammation. The health benefits of the oyster mushroom warrant further clinical studies.Systemic inflammation has been linked to the pathogenesis of a variety of diseases including endotoxemia and sepsis [1]. Abstract: Epidemiological studies suggest that mushroom intake is inversely correlated with gastric, gastrointestinal and breast cancers. We have recently demonstrated anticancer and anti-inflammatory activity of triterpene extract isolated from mushroom Ganoderma lucidum (GLT). The aim of the present study was to evaluate whether GLT prevents colitis-associated carcinogenesis in mice.

Abstract:

A. tortilis is commonly known as “Israeli babool”. The species is exposed to the world for its pods, gum and other aerial parts as fodder &feed for cattles. The plant was used as antidiabetic, antidiarrhoeal, antiasthmatic and also had several other medicinal benefits but the seed oil was not explored to the extent of its real potential. Hence the study was carried out to characterize the seed for oil and analysis of fatty acid composition through GC for quality control and maintenance. Commercial indices of oil were also estimated. Extracted oil is rich in Linolenic acid (50.43%) and Linoleic acid (36.74%). Beside this palmitic, stearic and oleic acid are other fatty acids identified. MUFA, PUFA, SFA and omega-6/ omega-3 ratio were also found to be promising for its potentiality in edible and clinical uses. The investigation thus scopes the utility of oil and prospects the lesser known plant seed as alternative oil source of nutritional and therapeutic importance.

Trends and Interannual Variability of Satellite-Based Wind and Sea Surface Temperature over the Southern Ocean in the Recent Decade  [PDF]

Abstract: Using satellite-based 10-m surface wind (SW), wind stress (WS) and sea surface temperature (SST) anomalies, trends and inter-annual variability during 1993 and 2008 over the Southern Ocean (SO) are addressed. The climatological mean (16 years average) indicates that negative wind stress curl diminished (enhanced) between 40°S and 60°S zonal strap region coincide with weak (strong) SW and warm (cold) SST anomaly during January (July). Annual climatology indicates that strong region of SW divides warmer waters northward with positive wind stress curl (WSCL) and colder waters southward with negative WSCL. The time series anomalies are smoothened with a 12-month running mean filter. The filtered area-mean time series anomalies of zonal and meridional component of SW and SST have linear trends of -0.0005 ± 0.0003 m/s/decade, 0.0012 ± 0.0002 m/s/decade and -0.00005°C ± 0.0001°C/decade, respectively. The SW anomalies show an increasing trend of 0.0013 ± 0.0002 m/s/decade, with the meridional (zonal) component exhibiting an increasing (decreasing) trend. The meridional component plays a critical role in heat transfer through atmospheric circulation. The WS and wind stress divergence exhibit increasing trends whereas wind stress curl shows a decreasing trend. The SST fluctuates close to zero with repeated high and low peaks at an interval of 2 - 3 years. We address the interannual variability by performing EOF analysis on SW, WS, WSCL and SST anomalies which have been passed through a 12-month running mean filter.

Abstract: History of breast cancer dates back to at least 1600 B.C. and treatment methods have undergone significant progress over the last hundred years. We are moving away from frighteningly radical, and towards increasingly more conservational breast cancer surgery. And while mastectomy is no longer a first-line choice for all breast cancers, it is still an important and, really, an essential procedure to discuss and research about. Different types and techniques exist and evidence regarding each is vast-with novel techniques appearing even nowadays. For example, robotic surgery is increasingly more common in many surgical specialties and procedures, and mastectomy is no exception. With several high-profile celebrities recently discussing their experiences of breast cancer and mastectomies, this article covers a multitude of essential aspects relevant to this topic, in turn, hopefully, helping patients and doctors deal with the diagnosis and plan the treatment accordingly. Current breast cancer care and mastectomy trends are also discussed here, giving the readers an up-to-date overview of how breast cancer can and should be managed.
Evaluation of rationality of promotional drug literature using World Health Organization guidelines

Abstract: Objectives : The study was aimed to evaluate collected drug promotional brochures for accuracy, consistency, and validity of the information presented in it, using World Health Organization (WHO) criteria for ethical medicinal drug promotion. Drug promotional brochures were evaluated for the type of claims and pictorial content presented in it and for references cited in support of these claims. Material and Methods : This observational, cross-sectional study was conducted in the outpatient department of Government Medical College and Hospital, Nagpur, India. In addition to the fulfillment of "WHO criteria, 1988," we examined 513 promotional brochures for the type of claims and pictorial content presented in it and references quoted in support of claims to check their retrievability, type, and authenticity. Results : None of the promotional literature fulfilled all WHO criteria. Majority (92%) brochures claimed about the efficacy of product, and a few about safety (37.8%). Out of 1003 references given in support of various claims, 84.4% were from journals and only 28.5% were validly presented researches. Brochures presenting irrelevant pictures were 41.3%, whereas brief prescription information (BPI) of the promoted drug was given only by 8.8% brochures. Conclusion : Pharmaceutical industries did not follow the WHO guidelines while promoting their products, thus aiming to satisfying their commercial motive rather than fulfill the educational aspect of promotion.
Anti-hyperalgesic effect of paracetamol in rat model of thermal hyperalgesia: implications for the treatment of neuropathic pain

Abstract: Background: Neuropathic pain conditions are prevalent in general population and difficult to treat. Most currently available therapies do not provide adequate benefit in all treated patients. Though Paracetamol is available for more than 100 years and have huge safety data, it is still not included in any treatment guidelines for neuropathic pain, as very few studies have evaluated its effects in neuropathic pain conditions. The aim of this study was to evaluate antihyperalgesic activity of paracetamol and to compare it with antihyperalgesic effect of amitriptyline and gabapentin in rat model of thermal hyperalgesia. Methods: Male albino wistar rats weighing 200-250 g of body weight and 4-6 months old were used. After anaesthetizing rats with 2% halothane, mild thermal injury was induced using hot plate analgesiometer. Drugs were administered intraperitoneally 30 minutes after thermal injury. Paw withdrawal latencies were measured at 30, 60, 90 and 120 minutes after drug administration. Statistical analysis done using GraphPad Prism version 5.01 and one way ANOVA followed by post hoc Dunnett's test or Tukey's test were used. Results: Paracetamol showed both dose as well as time dependent antihyperalgesic activity. Gabapentin demonstrated significantly more antihyperalgesic activity compared to amitriptyline (p< 0.05) and paracetamol (p< 0.01). Conclusions: Paracetamol showed antihyperalgesic activity however, it was less as compared to drugs like gabapentin and amitriptyline. Considering the excellent safety profile of paracetamol, it might be useful adjuvant drug for treatment of neuropathic pain conditions. [Int J Basic Clin Pharmacol 2013; 2(3.000): 290-297]
Promoting the proper use of medicines in rural school children of India

Abstract: Background: The problem of irrational use of medicines is prevalent in rural India. For safe and proper use of medicines there is a need to understand the proper way of taking medicines. Teaching proper use of medicines is the most neglected domain of the school health curriculum. So this study targeted school going children in rural area of India as the earlier the intervention, the more firmly longer lasting health related behavior can be successfully inculcated. The objectives of the study were (1) To study base level knowledge about use of medicines and (2) To implement and see the effect of an intervention to increase medicine knowledge in the form of information, education and communication (IEC) in rural school children. Methods: This was an interventional, questionnaire based (quantitative) study in which an intervention in the form of lectures which was given to school children. Rural school going children in age group of13-15 years were selected from 3 different schools of rural areas of Nagpur district, India. 500 school children were included after obtaining permission from school authorities and informed consent from their parents. Pretesting done by administering a questionnaire containing 20 questions to assess children's base level knowledge about medicine use. The same questionnaire was repeated on 16th and 45th day of intervention i.e. daily lectures of 25-30 minutes for 15 days on proper use of medicines, to judge the improvement. Results: It was observed that the interventions brought about a positive change in the knowledge of the students as well as increased awareness about proper use of medicines. Conclusions: This study showed that properly timed and meticulously implemented intervention can bring about a positive change in the attitude and knowledge of school children. Anomaly Cancellation in 2+1 dimensions in the presence of a domainwall mass

Abstract: A Fermion in 2+1 dimensions, with a mass function which depends on one spatial coordinate and passes through a zero ( a domain wall mass), is considered. In this model, originally proposed by Callan and Harvey, the gauge variation of the effective gauge action mainly consists of two terms. One comes from the induced Chern-Simons term and the other from the chiral fermions, bound to the 1+1 dimensional wall, and they are expected to cancel each other. Though there exist arguments in favour of this, based on the possible forms of the effective action valid far from the wall and some facts about theories of chiral fermions in 1+1 dimensions, a complete calculation is lacking. In this paper we present an explicit calculation of this cancellation at one loop valid even close to the wall. We show that, integrating out the ``massive'' modes of the theory does produce the Chern-Simons term, as appreciated previously. In addition we show that it generates a term that softens the high energy behaviour of the 1+1 dimensional effective chiral theory thereby resolving an ambiguity present in a general 1+1 dimensional theory.
A Large N Chiral Transition on a Plaquette

Abstract: We construct a model of a chiral transition using the well known large N transition in two dimensional U(N) lattice gauge theory. Restricting the model to a single plaquette, we introduce Grassmann variables on the corners of the plaquette with the natural phase factors of staggered fermions and couple them to the U(N) link variables. The classical theory has a continuous chiral symmetry which is broken at strong couplings, but is restored for weak couplings in the $N \to \infty$ limit.
