Glycoscience aids in biomarker discovery

Abstract: The glycome consists of all glycans (or carbohydrates) within abiological system, and modulates a wide range of important biologicalactivities, from protein folding to cellular communications.The mining of the glycome for disease markers representsa new paradigm for biomarker discovery; however, this effortis severely complicated by the vast complexity and structuraldiversity of glycans. This review summarizes recent developmentsin analytical technology and methodology as applied tothe fields of glycomics and glycoproteomics. Mass spectrometricstrategies for glycan compositional profiling are described, as arepotential refinements which allow structure-specific profiling.Analytical methods that can discern protein glycosylation at aspecific site of modification are also discussed in detail.Biomarker discovery applications are shown at each level ofanalysis, highlighting the key role that glycoscience can play inhelping scientists understand disease biology.
Transcriptome Profiling of Bovine Milk Oligosaccharide Metabolism Genes Using RNA-Sequencing

Abstract: This study examines the genes coding for enzymes involved in bovine milk oligosaccharide metabolism by comparing the oligosaccharide profiles with the expressions of glycosylation-related genes. Fresh milk samples (n = 32) were collected from four Holstein and Jersey cows at days 1, 15, 90 and 250 of lactation and free milk oligosaccharide profiles were analyzed. RNA was extracted from milk somatic cells at days 15 and 250 of lactation (n = 12) and gene expression analysis was conducted by RNA-Sequencing. A list was created of 121 glycosylation-related genes involved in oligosaccharide metabolism pathways in bovine by analyzing the oligosaccharide profiles and performing an extensive literature search. No significant differences were observed in either oligosaccharide profiles or expressions of glycosylation-related genes between Holstein and Jersey cows. The highest concentrations of free oligosaccharides were observed in the colostrum samples and a sharp decrease was observed in the concentration of free oligosaccharides on day 15, followed by progressive decrease on days 90 and 250. Ninety-two glycosylation-related genes were expressed in milk somatic cells. Most of these genes exhibited higher expression in day 250 samples indicating increases in net glycosylation-related metabolism in spite of decreases in free milk oligosaccharides in late lactation milk. Even though fucosylated free oligosaccharides were not identified, gene expression indicated the likely presence of fucosylated oligosaccharides in bovine milk. Fucosidase genes were expressed in milk and a possible explanation for not detecting fucosylated free oligosaccharides is the degradation of large fucosylated free oligosaccharides by the fucosidases. Detailed characterization of enzymes encoded by the 92 glycosylation-related genes identified in this study will provide the basic knowledge for metabolic network analysis of oligosaccharides in mammalian milk. These candidate genes will guide the design of a targeted breeding strategy to optimize the content of beneficial oligosaccharides in bovine milk.
Effects of Indigenous Innovation Policy on the S & T Outputs in China Evidence from the Higher Education System  [PDF]

Abstract: This paper evaluated the effects of indigenous innovation policies changes in 1996 and 2002 on the S&T outputs in the Chinese higher education system. I used interrupted time series design with yearly measures of number of SCI articles, average citation per item in 5 years, number of invention patent application accepted and invention patent granted from 1980 to 2008. The results showed statistically significant increases in the four output measures after the 1996 and 2002 innovation policies were adopted by the Chinese universities. In terms of effect size, the increases were 133% and 82% in number of SCI articles and 41% and 37% in citation impact for the two policy changes. The effect sizes are more noteworthy with 177% and 202% increases in patent applied 146% and 256% increase in patent granted for the two policy changes. The results supported all hypotheses and are basically consisted with prior research done by other scholars. However, small size sample and no control group are the two limitations of this research and future research could consider about the possibility to combine time series data and cross-sector data.

Abstract: The failure to mention the relationship between China’s WTO-plus obligation in Paragraph 11.3 and the GATT 1994 does not mean the negotiators had meant to deny China the right to invoke Article XX of the GATT 1994. If that was the case, the intention would have been recorded in the Protocol and the Working Party Report. Contexts provided by the Working Party Report and provisions of the GATT 1994 and other provisions of the WTO Agreement prove that China cannot be denied the right to invoke Article XX. WTO-plus obligations are still integral parts of the WTO Agreement and the decision of the Panel and the AB in this case will lead to absurd results.

Abstract:
In wireless networks, cooperative relaying could improve throughput by exploiting diversity. In order to reduce the amount of feedback for the channel gain, a semi-distributed scheme based on the relay feasible condition is proposed. Each relay node can measure its backward and forward channel gains. If both the channel gains are larger than a pre-defined threshold, this relay node is feasible. The final decision on the best relay selection is still given by the base station. Besides, the switch-and-examine relay selection scheme, which selects the first feasible relay node, is also investigated. Simulation results are presented to illustrate the advantage of two proposed schemes.

Abstract: With the deployment of more wireless applications, spectrum scarcity becomes an issue in many countries. Recent reports show that the reason for this spectrum shortage is the underutilization of some spectrum re-sources. Fortunately, the emergence of open spectrum and dynamic spectrum assess (DSA) technology in cognitive radio networks relieves this problem. In this paper, we propose a novel DSA-driven cognitive MAC protocol to achieve highly efficient spectrum usage and QoS provisioning. In the proposed protocol, secondary users are divided into several non-overlapping groups, and all leftover channels are allocated among groups taking the groups’ bandwidth requirements into consideration. Moreover, the allocation of vacant channels can be adjusted dynamically when members join/leave groups or primary users return/leave the current network. Simulations show that the proposed MAC protocol greatly improves the quality of ser-vice for secondary users and maximizes the utilization ratio of spectrum resources.

Abstract: Efficient radio resource management is essential in Quality-of-Service (QoS) provisioning for wireless communication networks. In this paper, we propose a novel priority-based packet scheduling algorithm for downlink OFDMA systems. The proposed algorithm is designed to support heterogeneous applications consisting of both real-time (RT) and non-real-time (NRT) traffics with the objective to increase the spectrum efficiency while satisfying diverse QoS requirements. It tightly couples the subchannel allocation and packet scheduling together through an integrated cross-layer approach in which each packet is assigned a priority value based on both the instantaneous channel conditions as well as the QoS constraints. An efficient suboptimal heuristic algorithm is proposed to reduce the computational complexity with marginal performance degradation compared to the optimal solution. Simulation results show that the proposed algorithm can significantly improve the system performance in terms of high spectral efficiency and low outage probability compared to conventional packet scheduling algorithms, thus is very suitable for the downlink of current OFDMA systems.
