Effectiveness and Safety of Generic Fixed-Dose Combination of Tenofovir/Emtricitabine/Efavirenz in HIV-1-Infected Patients in Western India

Abstract: Objective To assess effectiveness and safety of a generic fixed-dose combination of tenofovir (TDF)/emtricitabine (FTC)/efavirenz (EFV) among HIV-1-infected patients in Western India. Methods Antiretroviral (ARV)-naive and experienced (thymidine analog nucleoside reverse transcriptase inhibitor [tNRTI] replaced by TDF) patients were started on a regimen of 1 TDF/FTC/EFV pill once a day. They were followed clinically on a periodic basis, and viral loads and CD4 counts were measured at 6 and 12 months. Creatinine clearance was calculated at baseline and at 6 months and/or as clinically indicated. Effectiveness was defined as not having to discontinue the regimen due to failure or toxicity. Results One hundred forty-one patients who started TDF/FTC/EFV before 1 June 2007 were eligible. Of these, 130 (92.2%) and 44 (31.2%) had 6- and 12-months follow-up, respectively. Thirty-five percent of the patients were ARV-naive. Eleven patients discontinued treatment (4 for virologic failure, 1 for grade 3–4 central nervous system disturbances, 4 for grade 3–4 renal toxicity, and 2 for cost). Ninety-six percent of patients were virologically suppressed at 6 months. Frequency of TDF-associated grade 3–4 renal toxicity was 2.8%; however, 3 of these patients had comorbid conditions associated with renal dysfunction. Conclusion A fixed-dose combination of generic TDF/FTC/EFV is effective in ARV-naive and experienced patients. Although frequency of severe renal toxicity was higher than has been reported in the literature, it was safe in patients with no comorbid renal conditions.
Chimeras of Escherichia coli and Mycobacterium tuberculosis Single-Stranded DNA Binding Proteins: Characterization and Function in Escherichia coli

Abstract: Single stranded DNA binding proteins (SSBs) are vital for the survival of organisms. Studies on SSBs from the prototype, Escherichia coli (EcoSSB) and, an important human pathogen, Mycobacterium tuberculosis (MtuSSB) had shown that despite significant variations in their quaternary structures, the DNA binding and oligomerization properties of the two are similar. Here, we used the X-ray crystal structure data of the two SSBs to design a series of chimeric proteins (mβ1, mβ1′β2, mβ1–β5, mβ1–β6 and mβ4–β5) by transplanting β1, β1′β2, β1–β5, β1–β6 and β4–β5 regions, respectively of the N-terminal (DNA binding) domain of MtuSSB for the corresponding sequences in EcoSSB. In addition, mβ1′β2ESWR SSB was generated by mutating the MtuSSB specific ‘PRIY’ sequence in the β2 strand of mβ1′β2 SSB to EcoSSB specific ‘ESWR’ sequence. Biochemical characterization revealed that except for mβ1 SSB, all chimeras and a control construct lacking the C-terminal domain (ΔC SSB) bound DNA in modes corresponding to limited and unlimited modes of binding. However, the DNA on MtuSSB may follow a different path than the EcoSSB. Structural probing by protease digestion revealed that unlike other SSBs used, mβ1 SSB was also hypersensitive to chymotrypsin treatment. Further, to check for their biological activities, we developed a sensitive assay, and observed that mβ1–β6, MtuSSB, mβ1′β2 and mβ1–β5 SSBs complemented E. coli Δssb in a dose dependent manner. Complementation by the mβ1–β5 SSB was poor. In contrast, mβ1′β2ESWR SSB complemented E. coli as well as EcoSSB. The inefficiently functioning SSBs resulted in an elongated cell/filamentation phenotype of E. coli. Taken together, our observations suggest that specific interactions within the DNA binding domain of the homotetrameric SSBs are crucial for their biological function.

Abstract: Assistive and intelligent learning system is one of the best alternatives to provide education to handicapped and disabled persons. This kind of learning system may work well because handicapped and disabled students will not be encountered with the limitations and problems of the conventional education systems. This assistive learning system is mainly a multi-agent system based scheme. Those agents are autonomous agents and they have the cognitive behaviour and adaptability. This scheme is based on the centralized as well as distributed multi-agent planning for agent communication, collaboration and negotiation.

Abstract: This paper describes about the performance analysis of different data mining classifiers before and after feature selection on binomial data set. Three data mining classifiers Logistic Regression, SVM and Neural Network classifiers are considered in this paper for classification. The Congressional Voting Records data set is a binomial data set investigated in this study is taken from UCI machine learning repository. The classification performance of all classifiers is presented by using statistical performance measures like accuracy, specificity and sensitivity. Gain chart and R.O.C (Receiver Operating Characteristics) chart are also used to measure the performances of the classifiers. A comparative study is carried out among the data mining classifiers. Experimental result showed that without feature selection Logistic Regression and SVM classifiers provides 100% accuracy and neural network provides 98.13 % accuracy on test data set. With feature selection SVM classifier provides 100% accuracy. The performance of SVM classifier is found to be the best among all the classifiers with reduced number of features.
Extracting Quasiparticle Lifetimes from STM experiments

Abstract: Based on Quasiparticle Interference(QPI) around a point impurity, we demonstrate an analysis scheme that extracts the lifetime of a quasiparticle by using the Local Density of States(LDOS) data around the impurity in a Scanning Tunneling Microscopy(STM) experiment. This data analysis scheme would augment the Fourier-Transform Scanning Tunneling Spectroscopic method which provides us with the quasiparticle dispersion. Thus, point impurities can be used as probes to extract quasiparticle lifetimes from STM experiments and this would complement other experimental methods such as Angle Resolved Photo-emission Spectrocopy(ARPES). We detail how the scheme would apply to conventional metals/Fermi Liquids and Superconductors.

Abstract: The present study analyses the hydro-chemical data of groundwater samples of three different limestone mine sites, which are in close proximity and covers a tract along the Gujarat Coast of Indian peninsula. Higher TDS (> 4000 mg/L) and Cl values (> 2000 mg/L) as well as the Chloride: Bicarbonate (HCO3) ratio of more than one clearly support that the sea water intrusion is present in the coastal aquifer at all the three sites. In view of the economic importance of the area and the vulnerability of the coastal aquifer to sea water intrusion recommendations are made for sustainable use of groundwater by the mining companies and other stake holders.

Modeling of Multi-Agent Oriented learning System for Impaired Students with JADE

Abstract: In this research paper we presented a model of multi-agent system based learning environment for physically impaired students. The learning system is modeled on the basis of both centralized as well as distributed multi-agent planning. The entire learning system keeps track of the type of impairment the student has and mode of interaction of the environment is set depending on the type of impairment(s). The system consists of agents which are developed using JADE agent technology that helps the students with disabilities to continue studies from their own places.

Abstract: Analysis and proper assessment of multi-agent system properties are very much important. In this paper, we discussed about methodologies for modeling, analysis and design of multi-agent oriented system with the help of Petri net. A Multi-agent system can be considered as a discrete-event dynamic system and Petri nets are used as a modeling tool to assess the structural properties of the multi-agent system. Petri net provides an assessment of the interaction properties of the multi-agent.
