SLE – Hematopoietic stem cell transplantation for systemic lupus erythematosus

Abstract: The prognosis of systemic lupus erythematosus (SLE) markedly improved following the introduction of monthly intravenous pulse cyclophosphamide (500–1000 mg/m2). Nevertheless, despite pulse cyclophosphamide and advances in supportive care such as new antihypertensive medications, patients with active SLE involving visceral organs have 2-year and 5-year mortalities of approximately 20% and 35%, respectively. Since SLE is predominately a disease of young women, improvements in disease-related morbidity and mortality were desperately needed.Autologous hematopoietic stem cell transplant regimens for patients with cancer are based on dose escalation of chemotherapeutic drugs that demonstrate effectiveness at standard dosing. Borrowing this concept from the field of oncology, we dose escalated cyclophosphamide, the most effective antilupus medication, to 200 mg/kg. Also borrowing from experience with a hematopoietic stem cell transplantation (HSCT) conditioning regimen for aplastic anemia, where T lymphocytes help to sustain the disease manifestations, a regimen of 200 mg/kg cyclophosphamide (divided as 50 mg/kg over 4 days) and of 90 mg/kg equine antithymocyte globulin (ATG) (divided as 30 mg/kg over 3 days) was selected.To collect hematopoietic stem cells, they need to be mobilized into the peripheral blood with either a hematopoietic colony stimulating factor such as granulocyte-colony stimulating factor or a chemotherapeutic drug such as cyclophosphamide, or both. Since granulocyte-colony stimulating factor is a proinflammatory cytokine that by itself may exacerbate disease, the stem cells were mobilized into the peripheral blood using 2.0 g/m2 cyclophosphamide, granulocyte-colony stimulating factor beginning 72 hours later, and harvest beginning upon white blood cell rebound (usually 10 days after cyclophosphamide). Abstract: Ridley Scott’s Kingdom of Heaven (2005) certainly invites a reading along the lines of historicist cultural criticism. The final intertitle (and final shot of the film) refers to an “uneasy truce,” which implicitly extended to the film and its more critical reviewers. Despite statements to the contrary, Bush has been widely viewed as advancing U.S. imperialism in more naked form. A number of reviewers and Scott himself have connected the dots between Bush’s reference to a “crusade” against terrorism just after 9/11 and Scott’s film. But understanding the film and its paratexts as relayed between two uncannily double DVDs means that historicism and film and media theory in its cultural studies version cannot escape the trauma which the film and the historicist film critic want to make symbolic and narratable. The film’s uncanny epitexts and delayed delivery, potentially accelerating the speed of reviewing, and collapsing old and new, mechanical and human, oscillate without a telos between cinematic (re)inscriptions and their framing ex-scriptions.
Whole genome comparative studies between chicken and turkey and their implications for avian genome evolution

Abstract: We present a detailed comparative cytogenetic map between chicken and turkey based on reciprocal chromosome painting and mapping of 338 chicken BACs to turkey metaphases. Two inter-chromosomal changes (both involving centromeres) and three pericentric inversions have been identified between chicken and turkey; and array CGH identified 16 inter-specific CNVs.This is the first study to combine the modalities of zoo-FISH and array CGH between different avian species. The first insight into the conservation of microchromosomes, the first comparative cytogenetic map of any bird and the first appraisal of CNVs between birds is provided. Results suggest that avian genomes have remained relatively stable during evolution compared to mammalian equivalents.Comparative genomics is a powerful means for establishing relationships between gene function and location in a range of organisms. Moreover it allows insight into large-scale genomic rearrangements, conservation of functional elements and tracing of evolutionary phylogenies through the examination of both closely and distantly related species. The completion of the chicken (Gallus gallus, GGA) genome [1] and its associated resources provide the basis for the rapid development of detailed genomic information, potentially in all other birds. The most powerful strategies combine in-silico and experimental approaches, e.g. sequence comparison, cross-species fluorescent in-situ hybridization (zoo-FISH) [2,3] and, more recently, the use of whole-genome tiling path microarrays for cross-species array comparative genomic hybridization (array CGH) [4-8]. Sediment-water column fluxes of carbon, oxygen and nutrients in Bedford Basin, Nova Scotia, inferred from 224Ra measurements

Abstract: Exchanges between sediment pore waters and the overlying water column play a significant role in the chemical budgets of many important chemical constituents. Direct quantification of such benthic fluxes requires explicit knowledge of the sediment properties and biogeochemistry. Alternatively, changes in water-column properties near the sediment-water interface can be exploited to gain insight into the sediment biogeochemistry and benthic fluxes. Here, we apply a 1-D diffusive mixing model to near-bottom water-column profiles of 224Ra activity in order to yield vertical eddy diffusivities (KZ), based upon which we assess the diffusive exchange of dissolved inorganic carbon (DIC), nutrients and oxygen (O2), across the sediment-water interface in a coastal inlet, Bedford Basin, Nova Scotia, Canada. Numerical model results are consistent with the assumptions regarding a constant, single benthic source of 224Ra, the lack of mixing by advective processes, and a predominantly benthic source and sink of DIC and O2, respectively, with minimal water-column respiration in the deep waters of Bedford Basin. Near-bottom observations of DIC, O2 and nutrients provide flux ratios similar to Redfield values, suggesting that benthic respiration of primarily marine organic matter is the dominant driver. Furthermore, a relative deficit of nitrate in the observed flux ratios indicates that denitrification also plays a role in the oxidation of organic matter, although its occurrence was not strong enough to allow us to detect the corresponding AT fluxes out of the sediment. Finally, comparison with other carbon sources reveal the observed benthic DIC release as a significant contributor to the Bedford Basin carbon system.

Paclitaxel (PTX)-loaded micelles and nanospheres were evaluated for their cytotoxicity, cellular polymer uptake, and drug accumulation in drug-sensitive (Madin–Darby Canine Kidney [MDCK]II) and multidrug-resistant (MDR) P-glycoprotein (P-gp)-overexpressing (MDCKII-MDR1) cell lines. Both types of PTX-loaded nanoparticles were equally effective at inhibiting proliferation of MDCKII cells, but PTX-loaded micelles were more cytotoxic than nanospheres in MDCKII-MDR1 cells. The intracellular accumulation of both PTX and the diblock copolymers were similar for both nanoparticles, suggesting that the difference in cytotoxicity might be due to the different drug-release profiles. Furthermore, the cytotoxicity of these PTX-loaded nanoparticles was enhanced when these systems were subsequently or concurrently combined with a low-molecular-weight MePEG-b-PCL diblock copolymer, which we have previously demonstrated to be an effective P-gp inhibitor. These results suggest that the dual functionality of MePEG-b-PCL might be useful in delivering drug intracellularly and in modulating P-gp in order to optimize the cytotoxicity of PTX in multidrug-resistant cells.

Finally, comparison with other carbon sources reveal the observed benthic DIC release as a significant contributor to the Bedford Basin carbon system.

The Universe as a Domain Wall

Abstract: It is shown that the effective five-dimensional theory of the strongly coupled heterotic string is a gauged version of N=1 five-dimensional supergravity with four-dimensional boundaries. For the universal supermultiplets, this theory is explicitly constructed by a generalized dimensional reduction procedure on a Calabi-Yau manifold. A crucial ingredient in the reduction is the retention of a "non-zero mode" of the four-form field strength, leading to the gauging of the universal hypermultiplet by the graviphoton. We show that this theory has an exact three-brane domain wall solution which reduces to Witten's "deformed" Calabi-Yau background upon linearization. This solution consists of two parallel three-branes with sources provided by the four-dimensional boundary theories and constitutes the appropriate background for a reduction to four dimensions. Four-dimensional space-time is then identified with the three-brane worldvolume.
