Garcilaso de la Vega traductor
Ricardo Silva-Santisteban
Fragment-Based Screening Maps Inhibitor Interactions in the ATP-Binding Site of Checkpoint Kinase 2

Abstract: Checkpoint kinase 2 (CHK2) is an important serine/threonine kinase in the cellular response to DNA damage. A fragment-based screening campaign using a combination of a high-concentration AlphaScreen? kinase assay and a biophysical thermal shift assay, followed by X-ray crystallography, identified a number of chemically different ligand-efficient CHK2 hinge-binding scaffolds that have not been exploited in known CHK2 inhibitors. In addition, it showed that the use of these orthogonal techniques allowed efficient discrimination between genuine hit matter and false positives from each individual assay technology. Furthermore, the CHK2 crystal structures with a quinoxaline-based fragment and its follow-up compound highlight a hydrophobic area above the hinge region not previously explored in rational CHK2 inhibitor design, but which might be exploited to enhance both potency and selectivity of CHK2 inhibitors.

Between 1980 and 2000, Peru experienced an internal armed conflict that resulted in victims in numbers unprecedented in the history of Latin America. The protagonists of the armed struggle included the subversive collectives (PCP-SL and MRTA), as much as the army, the police and the rural militias ( ronde paysanne ). The consequences were dramatic in that the sheer human losses were compounded by a tragic unraveling of the social fabric. Women who participated directly or indirectly in the conflict – if they still live – bear the stigmata of their experience: scars, rape, post-traumatic stress. Here, we consider the victims of both camps, but also those women who became justice agents making claims on behalf of their own near relations, their spouses and children – thus forming the first human rights collectives. However, the women who have benefited from media recognition undoubtedly include those who committed crimes: women who took up arms as militants, combatants, or as members of the forces of order.
Silva, Ricardo;
Lua Nova: Revista de Cultura e Política , 2008, DOI: 10.1590/S0102-64452008000200007
Abstract: the article aims to analyze the interventions of quentin skinner and philip pettit in the current disputes on the meaning of the concept of liberty. it argues that despite differences of method and style between the authors, both skinner and pettit converge to the advocacy of a republican ideal of "freedom as the absence of domination" which is neither reduced to the ideal of self-government of active citizens (positive liberty), nor to the ideal of simple absence of coercion on agents (negative liberty). although the republican liberty à la skinner and pettit is also characterized by the absence of interference, it differentiates itself from the dominant liberal view in that it repudiates only the arbitrary forms of interference - phenomena of dependence and domination - which corrupt the relations amongst citizens as well as the relations between the citizens and the state. non-arbitrary forms of interference may be beneficial to - or even constitutive of - liberty.
O contextualismo linguístico na história do pensamento político: Quentin Skinner e o debate metodológico contemporaneo
Silva, Ricardo;
Dados , 2010, DOI: 10.1590/S0011-52582010000200002
Abstract: this article aims to map and investigate the methodological debate spearheaded by the cambridge school of history of political thought in the last four decades, focusing on the formulations of quentin skinner and the main objections raised by his critics. after a brief presentation of the prescriptions in skinner's methods, the article discusses how his linguistic contextualism has been criticized for: a) his epistemological commitments (denounced alternately as relativist and positivist); b) his adherence to intentionalism; and c) his tendency towards "antiquarianism". the article concludes that of these three modalities of objections, the attribution of "antiquarianism" was the one that most compromised skinner's original methodological formulations, recently leading him to virtually abandon his original antipresentist thrust.
Silva, Ricardo;
Revista Brasileira de Ciências Sociais , 2010, DOI: 10.1590/S0102-69092010000100004
Abstract: machiavelli's ideas have been radically re-interpreted in the scope of the current republican revival both in intellectual history and political theory. the "advisor to tyrants" and master of disguise has been replaced by the selfless patriot and the philosopher dedicated to the cause of freedom and to the defense of the commonwealth. nevertheless, the republican machiavelli that emerges from the criticism to the traditional interpretation still doesn't have stable and well defined features. what does the nucleus of the machiavellian republicanism consist of? by mobilizing a series of recent interpreters of machiavelli, this article examines, in a comparative manner, three interpretation models of the florentine's political ideas.
Republicanismo neo-romano e democracia contestatória
Silva, Ricardo;
Revista de Sociologia e Política , 2011, DOI: 10.1590/S0104-44782011000200004
Abstract: this article examines the concept of democracy that is derived from the "neo-roman" formula of freedom as absence of domination. for those authors linked to the neo-roman tradition, the fulfillment of the republican principle of the people as the "guardian of freedom" depends upon the constitution of a model of contestatory democracy presented as surpassing the models of electoral and participatory democracy _ both of which are considered to be tributaries of a positive conception of freedom. after an initial focus on the concept of contestatory democracy, this article goes on to analyze the major criticisms that have been made of this concept. two objections are given salience: first, that the rationalism of the contestatory model leads to the de-politization of democratic deliberation and second, that excessive neo-roman zeal against populism and the "tirany of the majority" reproduces the "elitist" characteristics of the classic republican tradition. we conclude with a brief evaluation of the pertinence of these criticisms.
Silva, Ricardo;
Revista de Sociologia e Política , 2000, DOI: 10.1590/S0104-44782000000100005
Abstract: this article has the aim of reconstructing the trajectory of programs of economic stabilization elaborated in the context of the crisis of the early 1960's in brazil. between the late 1950's and the fall of jo?o goulart's government, in april of 1964, at least three ambitious monetary stabilization programs were attempted by the governments of the period, all of which with no success. the article focus on the political reaction of the different actors in that critical conjuncture in relation to kubitschek's government "plano de estabiliza??o monetária" (1958-1959), quadros' government "reforma cambial" (1961) and, in detail, goulart's government "plano trienal" (1963-1965). it will be seen that those reactions follow from the break up of ideological consensus around development policies and the negative answer of the main socio-economic groups to the submission to "sacrifices" imposed by the planners for the achievement of monetary stability.
História intelectual e teoria política
Silva, Ricardo;
Revista de Sociologia e Política , 2009, DOI: 10.1590/S0104-44782009000300020
Abstract: this essay attempts to present and analyze three theoretical endeavors within the field of intellectualhistory that have contributed to enriching the prospects for a historically informed political theory.regardless of the fact that kosselleck, skinner and bevir disagree regarding the specific object ofintellectual history (concepts, linguistic acts and expressed beliefs, respectively), these three authorscan be seen as providing formulations of a type of political theorization that is sensitive to time and tothe contingencies of political life.
