Abstract: the gag gene of 5 caev samples, isolated from naturally infected goats from rio grande do sul, brazil, were analised by pcr and restriction endonuclease (ddei, haeiii e ndei) digestion. fragments of about 600 bp were amplified by pcr and submitted to enzymatic digestion. the patterns observed were compared with the corresponding gag sequences from 6 small ruminant lentiviruses. the results obtained allowed the separation of 3 distinct groups. the restriction fragment profiles observed were different from those previously described.

Abstract: small ruminant lentiviruses (srlv) are distributed worldwide and cause persistent infections in sheep and goats. the purpose of this work was to develop an indirect immunofluorescent antibody (ifa) test using brazilian srlv isolates for serological diagnosis of infection in goats. the ifa test was compared in its sensitivity and specificity to the agid test in which maedi-visna wlc-1 strain was used as antigen. ovine synovial secondary cell cultures infected with two goat srlv isolates (caev ufrgs/br-2 and caev ufrgs/br-5) were used for the ifa. goat serum samples (n = 239) were tested by the two tests. the agid test detected antibodies in 129 (53.9%) serum samples. a higher number of animals was considered positive for the ifa. however, different results were obtained depending on the isolate used for the antigen preparation. when caev ufrgs/br-2 was used as antigen, 216 (90.3%) serum samples tested positive, against 213 (89.1%) with caev ufrgs/br-5. there was no significant statistical difference between the antigens prepared with these two isolates. the ifa had sensitivity of 94.6% and 96.9%, and specificity of 14.5 % and 20%, when caev ufrgs/br-2 and caev ufrgs/br-5 were used as antigens, respectively. these results demonstrate that the ifa with antigens prepared with brazilian srlv isolates may play an important a role as a complementary serological test for the diagnosis of infections by these agents.

Abstract: Realizou-se a análise de parte do gene gag, que codifica para as proteínas do capsídeo viral, de 5 amostras de CAEV isolados de animais naturalmente infectados do Rio Grande do Sul, Brasil. As amostras foram analisadas por PCR e clivagem com enzimas de restri o (DdeI, HaeIII e NdeI). Fragmentos de aproximadamente 600 pb foram amplificados na PCR e submetidos à digest o enzimática. Os perfis obtidos foram comparados com as seqüências gag de 6 lentivírus de pequenos ruminantes.Os resultados obtidos permitiram separar as amostras em 3 grupos distintos. Os fragmentos observados foram diferentes dos descritos previamente.

Abstract: Os lentivírus de pequenos ruminantes (SRLV) têm distribui o mundial e causam infec es persistentes em ovinos e caprinos. O objetivo deste trabalho foi desenvolver um teste de imunofluorescência indireta (IFA), utilizando isolados brasileiros de SRLV, para o diagnóstico sorológico de infec o por estes agentes em caprinos. A técnica de IFA foi comparada, quanto à sensibilidade e à especificidade, ao teste de AGID com antígeno do vírus Maedi-Visna WLC-1. Cultivos celulares secundários de membrana sinovial ovina infectadas com dois isolados de SRLV de origem caprina (CAEV Br/UFRGS-2 e CAEV Br/UFRGS-5) foram utilizados para o teste de IFA. Duzentas e trinta e nove amostras de soro caprino foram submetidas aos dois testes. O teste de AGID detectou 129 (53.9%) amostras de soro caprino com anticorpos para SRLV. O teste de IFA detectou mais amostras reagentes, sendo que resultados diferentes foram observados de acordo com o isolado de SRLV empregado. Quando o isolado CAEV Br/UFRGS-2 foi utilizado como antígeno, 216 (90.3%) amostras de soro caprino foram reagentes, enquanto que o isolado CAEV Br/UFRGS-5 detectou 213 (89.1%) amostras de soro positivas. N o houve diferen a estatisticamente significativa entre esses dois isolados. O teste de IFA desenvolvido teve sensibilidade de 94.6% e 96.9% e especificidade 14.5% e 20%, quando os isolados CAEV Br/UFRGS-2 e CAEV Br/UFRGS-5 foram usados como antígeno, respectivamente. O aprimoramento da técnica, assim como sua compara o com um teste mais sensível, ainda se fazem necessários. No entanto, os resultados demonstraram que a técnica de IFA, utilizando isolados brasileiros de SRLV como antígeno, apresenta potencial como um teste alternativo e complementar para o diagnóstico sorológico de infec o por estes agentes.

Abstract: feline immunodeficiency virus (fiv) infection of domestic cats is one of the most promising animal models for the infection by the human immunodeficiency virus (hiv) which causes acquired immunodeficiency syndrome (aids). infected cats may develop a disease similar to that observed in aids patients, with increased susceptibility to opportunistic infections. in this study we used the polymerase chain reaction (pcr) to detect proviral dna of feline immunodeficiency virus on the blood and tissue samples from cats with a clinical diagnosis of immunodeficiency. the pcr primers were used to amplify the gag gene, which is conserved among different isolates. from 40 samples analyzed, 15 were positive and 4 of them were submitted to hybridization to confirm the specificity of the amplified fragments. these results confirm the presence of fiv in domestic cats in rio grande do sul, brazil.

Abstract: A infec o de gatos domésticos pelo Vírus da Imunodeficiência Felina (FIV) é um dos modelos mais promissores para o estudo da infec o pelo vírus da imunodeficiência humana (HIV) que causa a Síndrome de Imunodeficiência Adquirida (AIDS). O FIV causa, em gatos, uma enfermidade similar àquela observada em pacientes com AIDS, sobretudo no que diz respeito ao aumento da susceptibilidade a infec es oportunistas. No presente estudo, utilizou-se a Rea o em Cadeia da Polimerase (PCR), com o objetivo de detectar o provírus do FIV em gatos com sinais clínicos de imunodeficiência. O fragmento de DNA escolhido como alvo para amplifica o situa-se no gene gag do lentivírus felino, o qual é conservado entre as diferentes amostras do vírus. O DNA utilizado foi extraído a partir de amostras de sangue e de tecidos de animais com suspeita clínica de imunodeficiência. Das 40 amostras analisadas, 15 foram positivas, das quais 4 foram submetidas à hibridiza o, confirmando a especificidade dos fragmentos amplificados. Esses resultados demonstram a presen a do FIV na popula o de gatos domésticos do Rio Grande do Sul, Brasil.
Amphiphile Adsorption on Rigid Polyelectrolytes

Abstract: A theory is presented which quantitatively accounts for the cooperative adsorption of cationic surfactants to anionic polyelectrolytes. For high salt concentration we find that the critical adsorption concentration (CAC) is a bilinear function of the polyion monomer and salt concentrations, with the coefficients dependent only on the type of surfactant used. The results presented in the paper might be useful for designing more efficient gene delivery systems.
Co-infection by porcine circovirus type 2 and porcine parvovirus in aborted fetuses and stillborn piglets in southern Brazil

Abstract: porcine circovirus types 1 and 2 (pcv1, pcv2) and porcine parvovirus (ppv) are widespread in pig populations around the world. nevertheless, only pcv2 has been associated with different clinical syndromes, thus representing a major problem to the pig industry. the association of cases of swine abortions and stillborns with pcv1 and pcv2 and ppv was studied retrospectively (2005-2007). additional pathogens were also investigated in lesioned fetuses. the studied litters included stillborn piglets and several mummified fetuses of varied sizes. ventricular dilatation, myocardial pale areas, and mesocolic edema were the gross lesions. escherichia coli was detected as co-infecting with pcv2 the cases in which mesocolic edema was seen. microscopic lesions included non-suppurative myocarditis, myocardial necrosis and fibrosis, mineralization foci and intranuclear inclusion bodies in cardiomyocytes, and interstitial mononuclear pneumonia. samples from 7 (5.78 per cent) of 121 aborted fetuses and stillborn piglets had lesions consistent with a viral cause and showed both positive anti-pcv2 immunostaining as well as pcv2-pcr. in samples from 3 (2.47 per cent) of these 7 fetuses, co-infection with ppv was confirmed by nested-pcr. both viruses were detected in fetuses at different stages of gestation. viral antigens of pcv2 were detected by immunohistochemistry mainly in macrophages and myocytes. pcv1 individually was not detected in any of these affected fetuses, but it was associated with pcv2 and/or ppv in some of them. these findings indicate that pcv2 alone or in association with ppv should be kept in mind when investigating causes of infectious abortion in pigs in brazil.
Teaching with an Attitude: Finding Ways to the Conundrum of a Postmodern Curriculum  [PDF]

Abstract: Several scholars in the field of education have questioned the constituent aspects of a curriculum that would better respond to the rising demands in contemporary societies. The relevance of such enquiry finds its place in the very transformations in today’s societies marked by significant changes in the ways of knowing, being, and acting primarily due to the advent of new digital media in more recent globalizing processes. As Burke (2009) has stated, if schools wish to maintain their relevance in society, they must take into account such changes by first and foremost acknowledging the new and complex ways of making meanings in out-of-school literacy practices as equally legitimating those happening within school contexts. Along with Burke (ibid.), this paper advocates the need of rethinking the notion of curriculum in the light of the new ontologies and epistemologies of postmodernity. Such task proves itself to be a conundrum inasmuch as the very notion of curriculum has been traditionally founded on modern principles such as linearity, stability, and universality (Silva, 2009). In view of this, how can educators respond to the challenge of redefining what should be taught in schools in postmodern times so that students would better perform in relation to the self and the other within their social practices? This paper aims to analyse the relationship between postmodern philosophical concepts, curriculum theory and educational practice by presenting the notion of curricular attitude (Duboc, 2012) as a local redesign of teaching practices within a Brazilian educational context. Despite being situated in the field of foreign language teaching, the notion of a curricular attitude might be of interest of other areas of knowledge since it seeks to revisit teaching practices in the light of wider philosophical concerns.
Electro-Optical Properties of Carbon Nanotubes Obtained by High Density Plasma Chemical Vapor Deposition  [PDF]

Abstract: In this work, we studied the electro-optical properties of high-aligned carbon nanotubes deposited at room temperature. For this, we used the High Density Plasma Chemical Vapor Deposition system. This system uses a new concept of plasma generation: a planar coil is coupled to an RF system for plasma generation. This was used together with an electrostatic shield, for plasma densification, thereby obtaining high-density plasmas. The carbon nanotubes were deposited using pure methane plasmas. Three methods were used for the surface modification of the sample: reference substrate (silicon wafer only submitted to a chemical cleaning), silicon wafer with surface roughness generated by plasma etching, silicon wafer with a thin iron film and silicon wafer with diamond nano powder used as precursor materials. For each kind of silicon wafer surface, the carbon nanotubes were deposited with two different deposition times (two and three hours). The carbon nanotubes structural characteristics were analyzed by Atomic Force Microscope and Scanning Electronic Microscope. The carbon nanotubes electrical characteristics were observed by Raman Spectroscopy and the carbon nanotubes electro-optical properties were analyzed by current vs voltage electrical measurements and photo-luminescence spectroscopy measurements. The photoelectric effect in the carbon nanotubes were determined by photo-induced current measurements. In this work, we obtained carbon nanotubes with semiconductor properties and carbon nanotubes with metallic properties. The electro-optical effects depend strongly on the substrate preparation and the deposition parameters of the carbon nanotubes. The carbon nanotubes are high aligned and show singular properties that can be used for many applications.
