Reordenamiento del gen MLL en pacientes pediátricos con leucemia aguda MLL gene rearrangement in pediatric patients with acute leukemia

Abstract: Los reordenamientos del gen MLL están presentes en la mayoría de los casos de leucemia aguda (LA) infantil y están asociados con variables biológicas específicas y con pobres resultados clínicos. Con el objetivo de analizar su incidencia y relevancia pronóstica realizamos un estudio molecular mediante la técnica de Southern blot, en 29 ni os de nuestra institución con LA, los cuales se encontraban al inicio o en recaída de la enfermedad. Para el estudio empleamos la sonda FA4, que es un inserto genómico del gen MLL. Pudimos detectar reordenamiento genético en 3 pacientes menores de 1 a o que tenían una LMA de tipo morfológico M5, con altos conteos leucocitarios y con visceromegalia. Estos 3 pacientes fallecieron sin alcanzar la remisión, lo que confirmó el valor del reordenamiento del gen MLL como indicador de leucemia de alto riesgo, y por tanto, como una herramienta apropiada para la administración oportuna de terapia adaptada al riesgo MLL gene rearrangements are present in most of the cases with infant acute leukemia (AL) and are associated with specific biological variables and with poor clinical outcomes. These 3 patients died without attaining remission, which confirmed the value of the MLL gene rearrangement as an indicator of high risk leukemia and, therefore, as an appropiate tool for the timely administration of therapy adapted to risk

Abstract: Los reordenamientos del gen MLL en la banda cromosómica 11q23 son frecuentes en leucemias agudas (LA) en ni os y en las LA secundarias desarrolladas después de la terapia con inhibidores de la enzima topoisomerasa II. En menor medida también se aprecia en adultos con LA. La presencia de estos reordenamientos se considera un indicador de mal pronóstico asociado con resultados clínicos desfavorables, por ello es muy importante realizar su determinación en las LA. En este trabajo mostramos los resultados preliminares de la introducción del estudio del gen MLL en nuestro país mediante la técnica de Southern. Analizamos ADN de 30 pacientes con LA, incluidos ni os y adultos, que en el momento del estudio se encontraban al debut o en recaída. El estudio molecular se realizó con la sonda FA4, que es un inserto genómico del gen MLL. Sólo uno de los 30 pacientes mostró bandas de reordenamiento con 2 enzimas de restricción diferentes, el resto mostró el gen MLL en configuración germinal. Es interesante destacar que el paciente con el reordenamiento era un ni o con leucemia mieloblástica aguda subtipo M5b, lo cual concuerda con la literatura, donde se describe que estos reordenamientos están estrechamente correlacionados con los subtipos mielomonocítico (M4) y monocítico (M5) de leucemia mieloide aguda (LMA) Rearrangements of MLL gen in llq23 chromosomal band are frequents in childhood type of acute leukemia (AL) and in secondary AL, developed after therapy with II topoisomerase enzyme. Only one out of 30 patients, presented rearrangement band with 2 distinct restriction enzymes, remainder showed MLL gen in germinal form. It is interesting to underline that patient with rearrangement was a child presenting subtype M5b acute myeloblastic leukemia, which agree with literature showing that these rearrangements are closely correlated to subtype myelomonocytic (M4) and monocytic (M5) of acute myeloid leukemia (AML)

Tissue Homeostasis in the Wing Disc of Drosophila melanogaster: Immediate Response to Massive Damage during Development

Abstract: All organisms have developed mechanisms to respond to organ or tissue damage that may appear during development or during the adult life. This process of regeneration is a major long-standing problem in Developmental Biology. We are using the Drosophila melanogaster wing imaginal disc to study the response to major damage inflicted during development. Using the Gal4/UAS/Gal80TS conditional system, we have induced massive cell killing by forcing activity of the pro-apoptotic gene hid in two major regions of the disc as defined by Gal4 inserts in the genes rotund (rn) and spalt (sal). The procedure ensures that at the end of a 40–48 hrs of ablation period the great majority of the cells of the original Rn or Sal domains have been eliminated. The results indicate that the damage provokes an immediate response aimed to keep the integrity of the epithelium and to repair the region under ablation. This includes an increase in cell proliferation to compensate for the cell loss and the replacement of the dead cells by others from outside of the damaged area. The response is almost contemporaneous with the damage, so that at the end of the ablation period the targeted region is already reconstructed. We find that the proliferative response is largely systemic, as the number of cells in division increases all over the disc. Furthermore, our results indicate that the Dpp and Wg pathways are not specifically involved in the regenerative response, but that activity of the JNK pathway is necessary both inside and outside the ablated domain for its reconstruction.

Abstract: congenital pathologies are those existing at or dating from birth. occurrence of congenital cystic lesions in the oral cavity is uncommon in neonates. eruption cyst (ec) is listed among these unusual lesions. it occurs within the mucosa overlying teeth that are about to erupt and, according to the current world health organization (who) classification of epithelial cysts of the jaws, ec is a separate entity. this paper presents a case of congenital ec successfully managed by close monitoring of the lesion, without any surgical procedure or tooth extraction. eruption of the teeth involved, primary central incisors, occurred at the fourth month of age. during this time neither the child nor mother had any complication such as pain on sucking, refusal to feed, airway obstruction, or aspiration of fluids or teeth.

Abstract: Cada vez más en distintos ámbitos de nuestra sociedad circula la noción de responsabilidad asociada a lo sucedido en una historia reciente que lleva inscrito el sino de la tragedia. Como se argumentará, se trata de una noción densa, polisémica y en permanente resignificación y, por tanto, su empleo genera ciertos problemas en los debates públicos y en la historiografía sobre el pasado cercano. En este artículo me propongo, entonces, realizar un recorrido conceptual sobre distintos usos y ámbitos de aplicación de la noción de responsabilidad para volver a pensar su utilidad y sus múltiples significados en relación con el pasado reciente argentino The notion of responsibility, associated to and inscribed tragically in our recent past, is increasingly used in different spheres of society. I will argue that this is a dense and polysemic notion, permanently subjected to re-signification and, therefore, its use generates certain problems in public debates and history studies of the recent past. In the present article I shall cover the different meanings and application schemes for the notion of responsibility in order to rethink its utility and multiple significances regarding the recent Argentinean past
On the quality of random number generators with taps

Abstract: Recent exact analytical results developed for the random number generators with taps are reported. These results are applicable to a wide class of algorithms, including random walks, cluster algorithms, Ising models. Practical considerations on the improvement of the quality of random numbers are discussed as well.
