Expression of Two N1 Clones with Single Amino Acid Dissimilarity of Avian Influenza H5N1 Virus

Abstract: Two clones of N1 gene derived from isolate A/Dk/Tangerang/Bbalitvet-ACIAR-TE11/2007 (H5N1) exhibit single mismatch of amino acid sequence at position 242 that is threonine and methionine for the clone #3 and #5, respectively. In order to evaluate the effect of the amino acid substitution, these clones were inserted into two different expression vectors that are pEGFP-C1 and pcDNA-3.3 TOPO TA cloning. Subsequently, the respective recombinant clones were transfected into eukaryotic cells, including CEF, RK13 and VERO using Lipofectamine ‘plus’ reagent. As a result, the clone #3 retaining atypical sequence showed lower expression level rather than the clone #15 in both vectors and all type of cells. The 3D conformational modelling revealed that the mutation occurs in the inner part of glycoprotein embedded within envelope or matrix. Therefore, the missense mutation seems has no effect on the antigenic properties of neuraminidase but this substitution by any means causes lethal mutagenesis in the individual gene expression by reducing level of protein transcript.
Reliability Analysis Using Fault Tree Analysis: A Review

Abstract: This paper reviews the literature published on the recent modifications made in the field of risk assessment using Fault Tree Analysis (FTA)in the last decade. This method was developed in 1960’s for the evaluation and estimation of system reliability and safety. In this paper we have presented the general procedure for FTA, its application in various fields and the modifications that have been made through the time to overcome the inadequacies of the method. In the last section some of the future wok is also discussed with a simplified methodology.
Evaluation of the Capabilities of Various Local Bamboo as the Places of Milk Fermentation without Inoculant of Lactic Acid Bacteria

Abstract: This research was conducted to evaluate the capabilities of the various local bamboos without the inoculation of lactic acid bacteria (LAB) in milk fermentation. The method used in this study was completely randomized design (CRD) with three treatments, namely D (delepung bamboo); A (ampel bamboo) and T (tali bamboo). The results of the study showed that fermentation in tali bamboo produced the lowest pH value (4.82) followed by ampel bamboo (4.87) and delepung bamboo (4.97). Low pH values were followed by a high total acidity with the results respectively 0.82%; 0.80% and 0.78%. Similarly, the highest level of product consistency was in tali bamboo treatment (1.08 min) followed by ampel bamboo (0.53 min) and delepung bamboo (0.38 minutes). Product moisture content was also the lowest in tali bamboo treatment (87.28%) followed by ampel bamboo (87.53%) and delepung bamboo (88.70%). Never the less the capability of tali bamboo as a natural incubator is not significantly different to delay delepung bamboo and ampel bamboo reviewed from protein percentage and fat product. While the total LAB found in the product of tali bamboo treatment (5.82 x 104 CFU/g) was significantly more than delepung bamboo (1.60 x 104 CFU/g) and ampel bamboo (2.37 x 104 CFU/g).
