Endophytic Streptomyces spp. as Biocontrol Agents of Rice Bacterial Leaf Blight Pathogen (Xanthomonas oryzae pv. oryzae)

Abstract: Xanthomonas oryzae pv. oryzae (Xoo), a causal agent of bacterial leaf blight (BLB), is one of the most important pathogens of rice. The effectiveness of ten Streptomyces spp. isolates in suppressing Xoo disease was assessed in planta and in vitro. In planta experiments were carried out in a greenhouse and arranged in a randomized completely block design (RCBD) with three replications. Twenty treatments were tested which included plants inoculated with both Streptomyces spp. and Xoo, and plants inoculated with only Streptomyces spp. Plants inoculated with Xoo and sprayed with a chemical bactericide, and plants inoculated with only Xoo served as positive controls, whereas plants not inoculated with either Streptomyces spp. or Xoo were used as negative controls. The results showed that the effect of endophytic Streptomyces spp. on BLB disease expressed as area under disease progress curve (AUDPC) was not significantly different to that on control plants (P > 0.05). However, plants inoculated with endophytic Streptomyces spp. were significantly taller and produced higher tiller number than control plants (P < 0.05). Streptomyces spp. isolate AB131-1 gave the highest plant height. In vitro studies on biocontrol mechanisms of selected Streptomyces spp. isolates showed that isolate LBR02 gave the highest inhibition activity on Xoo growth, followed by AB131-1 and AB131-2. Two isolates (AB131-1 and LBR02) were able to produce chitinase, phosphatase, and siderophore which included biocontrol characteristics. Morphological and colonization studies under SEM and light microscopy confirmed that the three isolates were endophytic Streptomyces spp. from different species. These studies found that the paddy plant which was inoculated with endophytic Streptomyces spp. AB131-1 and infected by Xoo could increase the height of plant and number of tillers.
Prospective Use of 1-Aminocyclopropane-1-Carboxylate Deaminase-Producing Bacteria for Plant Growth Promotion and Defense against Biotic and Abiotic Stresses in Peat-Soil-Agriculture

Abstract: The 1-aminocyclopropane-1-carboxylate (ACC) deaminase (EC4.1.99.4) is an enzyme produced by some soil bacteria to degrade ACC (the immediate precursor of ethylene) to reduce ethylene biosynthesis in higher plants. Increased concentrations of ethylene in plant tissues, which are triggered by various biotic and abiotic stresses, inhibits plant growth and weakens the plant defense against the stressors. Various findings on the successful use of ACC deaminase producing bacteria for plant growth under unfavorable soil conditions are inspiring their use in tropical peat-soil-agriculture, which possesses bio-physical constraints. It has been proven that inoculation of plants with ACC deaminase producing bacteria decreased ethylene inhibition generated by unfavorable environmental conditions, such as nutrient shortage, flooding, drought, high salts, and the presence of heavy metals and organic pollutants. Understanding the mechanisms by which ACC deaminase-producing bacteria act to reduce plant stress and the fitness of bacterial traits with the properties and constraints of peat-soils becomes a key to utilize these bacteria in improving crop productivity. The bacteria may ameliorate plant stress as well as promote plant growth under seasonal bio-physical changes of peat-soils that are usually encountered in the field.

Abstract: The Depok Land Use Planning predicts that in 2010, urban land use will be 73 percent, while 63 percent is for settlement.Depok is allocated for catchment area, so the expansion of paving area should be monitor. In this study, there were four types of settlement lower and middle type in perumnas and in middle type (perumnas kecil dan besar serta perumahan sederhana kecil dan besar). Average of paved area in the perumnas kecil was 66 m2, in perumnas besar was 89.5 m2. In perumahan sederhana kecil was 54.4 m2 and in perumahan sederhana besar was 121.3 m2. Land square meter and length of stay were the variables that infl uence the expansion of paved area. They did that because they have more space to expand their house, and now, there was no open space left

Abstract: The tobacco plantation in the area of Klaten Regency has many huge barns for drying tobacco since the year of 1850s, named los. The los(es) can be found in remote area in the villages; such as in Bendo Gantungan, within Gayamprit tobacco plantation. In other area, some of the los(es) can be seen from the main road or from the train within Yogyakarta - Surakarta; but they are not as many as in the year of 1970s – 1980s. The finding of this research confirms that los has high vernacular value. This can be shown not only from its organization of space; but also from its form, its local materials, and its local technique. Abstract in Bahasa Indonesia:
KONTROVERSI RUMAH DOME DI NGLEPEN, PRAMBANAN, D.I. YOGYAKARTA

Abstract: Housing the victims of the earthquake that does not take into consideration the localities can be predicted rarely successful. One of the many dwelling reconstructions can be found in the village of Nglepen, Prambanan, Yogyakarta Special Province. It is a dome houses complex. The people there who have lost their dwelling-houses may readily accept any form of shelter. But their attitude to accept and live in the dome houses actually causes many problems for themselves. Not only that the dome houses reject localities - therefore for housing Javanese people in rural area - but there are also other problems, one of which relates to financial budget. Many controversies come up from the finding of this work which applying environment-behavior research. Abstract in Bahasa Indonesia : Rekonstruksi bangunan bagi korban gempa dengan tidak mengindahkan lokalitas kemungkinan besar akan menimbulkan masalah di kemudian hari. Berbagai rekonstruksi bangunan bagi korban gempa 27 Mei 2006 di Yogyakarta banyak dilakukan, salah satunya berada di dusun Nglepen, Prambanan, D.I. Yogyakarta. Ini merupakan bantuan berupa rumah-rumah dome dalam satu kompleks perumahan. Korban gempa yang kehilangan rumah pasti akan menerima bantuan apapun yang dapat dipakai sebagai a tempat berteduha mereka. Namun kesediaan mereka untuk mau tinggal di rumah dome ternyata menimbulkan berbagai masalah yang harus mereka alami. Tidak saja bahwa rumah dome tidak mengindahkan lokalitas - dalam hal ini rumah untuk masyarakat Jawa di pedesaan - namun ada masalah lain yang harus mereka hadapi, antara lain dari segi finansial. Berbagai kontroversi muncul dari temuan-temuan penelitian ini yang memakai metode penelitian lingkungan-perilaku. PENGARUH TATA RUANG BANGSAL RUMAH SAKIT JIWA TERHADAP KESELAMATAN DAN KEAMANAN PASIEN

Abstract: Patients of a mental hospital stay in mental hospital wards during their ill-treatment time. The hope is that they can be recovered or at least their "illness" can be minimal before they involve in the rehabilitation process. It has been found out, according to this study, that some aspects of the elements of the interior of the ward affect negatively to the safety and security of adult patients. For example, in the means of safety, the patients might injure or be injured by another patients; while in the means of security, the patients might run away from the ward. This study has been conducted at P3/2nd Class mental hospital ward of Prof. dr. Soeroyo Mental Hospital of Type A in Magelang, Central Java. Because there is no standardized design in the interior of mental hospital wards, the findings of this study can be recommended only for the hospital has been studied. But when the wards of any other mental hospitals have similar characteristics in the interior with the interior of the ward under studied, then it can also be recommended for them. Abstract in Bahasa Indonesia : Bangsal suatu Rumah Sakit Jiwa (RSJ) digunakan sebagai tempat tinggal untuk pasien mental rawat inap. Harapannya ialah bahwa pasien mental akan bisa disembuhkan, atau paling tidak diperkecil "sakit"nya sebelum mengikuti proses rehabilitasi. Namun dari penelitian ditemukan bahwa justru aspek-aspek pada elemen tata ruang dalam bangsal itu sendiri yang bisa memberikan pengaruh negatip terhadap keselamatan dan keamanan pasien mental dewasa. Dalam hal keselamatan pasien mental dewasa antara lain memudahkan pasien untuk terlukai atau melukai sesama pasien. Dalam hal keamanan pasien mental dewasa antara lain memudahan pasien untuk melarikan diri. Penelitian ini dilakukan pada bangsal P3/Klas 2 Rumah Sakit Jiwa Tipe A Prof. dr. Soeroyo di Magelang, Jawa Tengah. Temuan-temuan penelitian ini direkomendasikan untuk RSJ yang dipakai sebagai studi, namun tidak tertutup kemungkinan untuk direkomendasikan bagi RSJ yang lain bila terdapat kesamaan karakteristik pada tata ruang dalam bangsalnya, karena memang tidak ada standar baku untuk RSJ. Kata kunci: bangsal RSJ, aspek-aspek pada elemen tata ruang dalam, pasien mental dewasa, keselamatan, keamanan.
MODERNISATION ISSUES OF GENDER AND SPACE

Abstract: Jurgen Habermas proposes a theory of modernisation based on the notion of a progressive "uncoupling" or separation of some aspects of everyday life, and their take over or "colonisation" by an expanding state and formal economy. This model is criticised by Nancy Fraser on the basis that it ignores the place of gender in the various roles that the individual plays in society and which mediate between the world of everyday life and the "system world" of the state and the official economy. Habermas's argument is further criticised for ignoring the issue of space (the house, the neighbourhood) in that mediation. Further, Habermas and also Fraser arguments however yield to what they call symbolic reproduction and material reproduction, socially integrated action contexts and system integrated action contexts, and also private sphere and public sphere. The above terms will be elaborated more fully in this paper.

Optimal Electroporation Condition for Small Interfering RNA Transfection into MDA-MB-468 Cell Line

Abstract: Background: Electroporation is a valuable tool for small interfering RNA (siRNA) delivery into cells because it efficiently transforms a wide variety of cell types. Since electroporation condition for each cell type must be determined experimentally, this study presents an optimal electroporation strategy to reproducibly and efficiently transfect MDA-MB 468 human breast cancer cell with siRNA. Methods: To identify the best condition, the cells were firstly electroporated without siRNA and cell viability was determined by trypan blue and MTT assays. Then siRNA transfection in the best condition was performed. Western blot analysis was used for monitoring successful siRNA transfection. Results: The best condition for electroporation of this cell line was 220 volt and 975 μF in exponential decay using the Gene Pulser X cell electroporation system. Our data demonstrated that by using proper electroporation condition, DNA methyl transferase mRNA was silenced by 10 nmol DNMT1 siRNA in MDA-MB 468 cells when compared with negative control siRNA electroporation. Analysis of cell viability demonstrated that optimal electroporation condition resulted in 74% and 78% cell viability by trypan blue staining and MTT assay, respectively. Conclusion: Transfection of the MDA-MB-468 breast cancer cell line with siRNA in the obtained electroporation condition was successful and resulted in effective gene silencing and high cellular viability.
