Assessment of proteolysis and sensory characteristics of prato cheese with adjunct culture Avalia o da proteólise e das características sensoriais de queijo prato com cultura adjunta

Abstract: Influence of adjunct cultures on the chemical and sensory characteristics, and proteolysis of Prato cheese was investigated. Cheeses were manufactured using a commercial starter culture and Lactobacillus strains (Lactobacillus plantarum or Lactobacillus helveticus) as adjunct cultures. Control cheeses lacked the adjunct culture. The chemical composition was analyzed at day 5 after manufacture and the proteolysis at days 5, 25, 45 and 65 of ripening. The sensory acceptance was assessed at 60 days. A split-plot design was used and the complete experiment was carried out in triplicate. The results were evaluated by ANOVA and Tukey’s test test at 5% significance level. There were no significant differences in chemical composition among the cheeses. A significant increase in proteolysis occurred during ripening period for the cheeses with adjunct culture when compared to cheeses without adjunct culture. Cheese with Lactobacillus helveticus showed higher scores for flavor, texture and purchase intent compared with the others treatments. Use of adjunct Lactobacillus suggests that the proteolysis of Prato cheese should be accelerated in order to reduce ripening period. A influência de culturas adjuntas sobre as características químicas e sensoriais, e sobre a proteólise do queijo Prato foi avaliada. Os queijos foram fabricados com cultura starter comercial e cepas de Lactobacillus (Lactobacillus plantarum ou Lactobacillus helveticus) como culturas adjuntas. Os queijos controle n o foram adicionados de cultura adjunta. A composi o química foi analisada no dia 5 após a fabrica o e a proteólise nos dias 5, 25, 45 e 65 de matura o. A aceita o sensorial foi avaliada em 60 dias. Um delineamento de parcelas subdivididas foi utilizado e o experimento completo foi realizado em triplicata. Os resultados foram avaliados pela análise de variancia e teste de Tukey no nível de 5% de probabilidade. Os queijos n o apresentaram diferen as significativas em rela o à composi o química. Um aumento significativo na proteólise ocorreu durante o período de matura o para os queijos com cultura adjunta, quando comparado aos queijos sem adi o desse tipo de cultura. Os queijos com Lactobacillus helveticus apresentaram médias das notas mais altas para os atributos sabor, textura e inten o de compra em compara o aos demais tratamentos. A utiliza o de Lactobacillus como cultura adjunta indica que a proteólise do queijo prato pode ser acelerada a fim de reduzir o seu tempo de matura o.

Abstract: The objective of this work was the production of probiotic “requeij o cremoso” that keeps the physicalchemical and sensorial characteristics of the tradicional product, linked to the functional properties. Control (without probiotic addition) and probiotic “requeij o cremoso” added with Lactobacillus casei after fusion, at 50±1oC, were produced, packed and stored at 4±1oC. Control and probiotic “requeij o cremoso” were evaluated to physical-chemical composition 1 day after processing. After 1, 7, 14 and 28 days of storage, the pH were evaluated for both products and Lactobacillus casei count only for probiotic “requeij o cremoso”. Sensory evaluation was accomplished on the 4th day of storage for appearance, taste, flavor, texture, general acceptance and purchasing intention. The results were evaluated by analysis of variance and Tukey’s test. There was no difference on physical-chemical composition of control and probiotic “requeij o cremoso”. Lactobacillus casei count was 2,2x107 UFC/g after 1 day of processing and not showed significant variations during the 28 days of storage. The probiotic “requeij o cremoso” presented good sensory acceptance and 82% of the consumers certainly or probably buy the probiotic product. The results showed that “requeij o cremoso” may regard as a good alternative of probiotic product. O objetivo deste trabalho foi a elabora o de um requeij o cremoso probiótico, que mantenha as características físico-químicas e sensoriais do produto tradicional, agregando propriedades funcionais. Foi produzido um requeij o cremoso controle (sem a adi o de probiótico) e outro adicionado de Lactobacillus casei, após a fus o da massa, à temperatura de 50±1oC. Os requeij es controle e probiótico foram envasados, armazenados a 4±1oC e submetidos às análises de composi o centesimal após 1 dia de processamento. O pH dos requeij es, assim como a contagem de Lactobacillus casei do requeij o probiótico foram avaliados após 1, 7, 14 e 28 dias de fabrica o. A avalia o sensorial foi realizada no 4o dia de armazenamento e os atributos analisados foram: aparência, sabor, aroma, textura, aceita o geral e inten o de compra. Os resultados foram avaliados por análise de variancia e teste de médias de Tukey. N o houve diferen a quanto à composi o centesimal entre o requeij o controle e o requeij o probiótico. Abstract: neem (azadirachta indica) is an indian tree well known for its several pharmacological activities, including antimicrobial activity. more than 300 composites have already been isolated and azadirachtin (aza) is its main active component. in the present work, neem leaves hydroalcoholic extracts were prepared by percolation in 96% ethanol different concentrations (50%, 60%, 70%, 80% and 90% (v/v)). the presence of aza was tested by tlc by eluting the extracts and a standard solution of aza through a chromatographic plate developed with anisaldehyde/sulfuric acid solution followed by heating. by hplc, extracts elution took place on a c18 column, water:acetonitrile (60:40) as mobile phase, 1.0 ml/min flow rate and detection at λ217 nm. the extracts did not display aza spots or peaks, however, they were tested against gram-positive and gram-negative bacteria, yeasts and a mold fungus. the extracts were tested in different increasing concentrations, in order to detect a dose-dependent relationship of the activity. despite the absence of aza, the 70% and 80% (v/v) ethanol extracts showed activity against staphylococcus aureus. however, this activity was not dose-dependent according to tukey's test (q0,05;3;7).

Abstract: This paper presents a modeling of a high-impedance bus differential protection logic using the ATP (Alternative Transients Program) MODELS language. The model is validated using ATP simulations on an electrical system consisting of a sectionalized bus arrangement with four transmission lines (TLs) and two autotransformers. The obtained results validate the model and present some of the advantages of using this type of bus protection, such as fast and safe operation, even when under adverse conditions such as current transformers (CTs) magnetic core saturation upon the occurrence of external faults.

Abstract: the purpose of this article is to investigate how the repression against the drug retailer traffic is related to the notion of state racism as formulated by the french philosopher michel foucault. the repression against the traffic, specially directed to the retailer traffic with bases of support in slums, and not to the big traffickers and others facilitators, suggests that its principal function is to legitimate practices of violence and extermination directed to the poor population. this text discusses the periculosity stigma often vinculed to the poverty, with the objective of understand the reasons that made possible the legitimization and the trivialization of these practices.

Abstract: in this paper we focus on the application of two mathematical alternative tasks to the teaching and learning of functions with high school students. the tasks were elaborated according to the following methodological approach: (i) problem solving and/or mathematics investigation and (ii) a pedagogical proposal, which defends that mathematical knowledge is developed by means of a balance between logic and intuition. we employed a qualitative research approach (characterized as a case study) aimed at analyzing the didactic pedagogical potential of this type of methodology in high school. we found that tasks such as those presented and discussed in this paper provide a more significant learning for the students, allowing a better conceptual understanding, becoming still more powerful when one considers the social-cultural context of the students.

Abstract: lupus erythematosus is an autoimmune disease of unknown etiology with cutaneous and vascular lesions. both discoid lupus erythematosus (dle) and systemic lupus (sle) affect the skin. visible skin lesions in young women can cause loss of self esteem. in the present study we aimed to evaluate and compare the quality of life in sle and led through an observational study of 64 patients. these patients were divided into 2 groups: group 1: sle (n = 38); group 2: dle (n = 26) and then completed the quality of life questionnaire - dermatology life quality index or dlqi. it was found that patients with dle have a worse quality of life than patients with sle. it is believed that this fact is generated by the difference in the spectrum of injuries.
Comparison of pharmacopeial statistical methods applied in microbiological assay for antibiotics potency determination using parallel lines and three-dose level

Abstract: Pharmaceutical equivalence studies, useful for checking the interchangeability of generic medicines and their respective innovator medicines, have been carried out in Brazil since 1999, as a consequence of the establishment of the generic medicine policy. For medicines containing antibiotics, microbiological assays are often the most appropriate method. However, the statistical methods applied in these assays are not widely known due to the difficult access to official codes and/or little knowledge of the statistical tools of analysis. Thus, the aim of this work was to compare the statistical methods for determining the potency of antibiotics through the cylinder-plate method using parallel lines and a three-dose level model, as described in the Brazilian Pharmacopeia (4th edition, 1988), British Pharmacopoeia 2011, European Pharmacopoeia (7th edition), The International Pharmacopoeia (4th edition), and United States Pharmacopeia (34th edition). The assay is illustrated with the antibiotic ofloxacin, and details on orthogonal coefficients, normality test, homogeneity of variance test, and detection of outliers are discussed. The calculations obtained by statistical analyses from different pharmacopeias lead to the same final interpretation. In practice, with the availability of alternative pharmacopeial methods, the analyst can choose the most appropriate statistical calculation to be used. Os estudos de equivalência farmacêutica, úteis na verifica o da intercambialidade entre os medicamentos genéricos e respectivos medicamentos de referência, têm sido realizados no Brasil desde 1999, como consequência do estabelecimento da política de medicamentos genéricos. Para medicamentos contendo antibióticos, os ensaios microbiológicos s o, muitas vezes, o método mais adequado. Entretanto, os métodos estatísticos aplicados nesses ensaios n o s o amplamente conhecidos devido à dificuldade de acesso aos compêndios oficiais e/ou pouca compreens o das ferramentas estatísticas de análises. Abstract: saccharin, a synthetic sweetener constituent of several pharmaceutical and cosmetic preparations, is analyzed qualitatively and quantitatively in the presence of interferences like active ingredients and excipients. a derivative ultraviolet spectrophotometric method employing the second and the fourth orders was used. the calibration data obtained were satisfactory, with a relative precision range of 0.53%-0.72%, and linearity between 0.9998-0.9999. the recovery test results for the toothgel sample ranged between 97.80% and 104.95% in the second order of derivative and between 98.04% and 101.32% in the fourth derivative, respectively, using zero-peak (zp) and peak-peak (pp) methods. the suspension sample showed values between 97.35% and 104.72% in the second derivative and between 95.94% to 104.41% in the fourth derivative, respectively, employing the zero-peak (zp) and peak-peak (pp) methods. the proposed method showed precision, accuracy and simplicity.
