Estradiol Stimulates Vasodilatory and Metabolic Pathways in Cultured Human Endothelial Cells

Abstract: Vascular effects of estradiol are being investigated because there are controversies among clinical and experimental studies. DNA microarrays were used to investigate global gene expression patterns in cultured human umbilical vein endothelial cells (HUVEC) exposed to 1 nmol/L estradiol for 24 hours. When compared to control, 187 genes were identified as differentially expressed with 1.9-fold change threshold. Supervised principal component analysis and hierarchical cluster analysis revealed the differences between control and estradiol-treated samples. Physiological concentrations of estradiol are sufficient to elicit significant changes in HUVEC gene expression. Notch signaling, actin cytoskeleton signaling, pentose phosphate pathway, axonal guidance signaling and integrin signaling were the top-five canonical pathways significantly regulated by estrogen. A total of 26 regulatory networks were identified as estrogen responsive. Microarray data were confirmed by quantitative RT-PCR in cardiovascular meaning genes; cyclooxigenase (COX)1, dimethylarginine dimethylaminohydrolase (DDAH)2, phospholipase A2 group IV (PLA2G4) B, and 7-dehydrocholesterol reductase were up-regulated by estradiol in a dose-dependent and estrogen receptor-dependent way, whereas COX2, DDAH1 and PLA2G4A remained unaltered. Moreover, estradiol-induced COX1 gene expression resulted in increased COX1 protein content and enhanced prostacyclin production. DDAH2 protein content was also increased, which in turn decreased asymmetric dimethylarginine concentration and increased NO release. All stimulated effects of estradiol on gene and protein expression were estrogen receptor-dependent, since were abolished in the presence of the estrogen receptor antagonist ICI 182780. This study identifies new vascular mechanisms of action by which estradiol may contribute to a wide range of biological processes.
Computed Tomography Angiography before Intravenous Thrombolysis Does Not Increase the Risk of Renal Dysfunction

Abstract: Our aim is to determine whether computed tomography angiography (CTA) before intravenous thrombolysis (IVT) affects renal function in acute ischemic stroke (AIS) patients. We performed an observational analysis of AIS patients treated with IVT for three years. Patients were classified into 2 groups: those who underwent CTA (CTA-group) and those who did not (control-group). Differences in creatinine levels between baseline and 24–72 hours after IVT were calculated. Acute renal dysfunction (ARD) was defined as an increase in serum creatinine level of ≥0.5?mg/dL and/or ≥25% above baseline within 24–72 hours after IVT. 190 patients were treated with IVT. Renal function (before and after IVT) was assessed in 162 (115 in control-group; 47 in CTA-group). Nine patients (5.5%) developed ARD (2 (4.2%) in CTA-group and 7 (6.1%) in control-group; ). CTA was not associated with a higher risk of ARD and did not affect the efficacy or safety of IVT. Previous chronic renal insufficiency, baseline creatinine levels, and previous use of nonsteroidal anti-inflammatory drugs were associated with a significant increase in creatinine levels, independently of contrast use. In conclusion, CTA does not seem to increase the risk of renal dysfunction. This technique may be used safely without knowledge of baseline creatinine levels. 1. Introduction Computed tomography angiography (CTA) is commonly used in patients with acute ischemic stroke (AIS) to diagnose cerebral arterial occlusion before thrombolysis. This technique makes it possible to identify intracranial and extracranial vascular stenoses or occlusions, which are a relevant prognostic factor indicating intra-arterial interventions such as thrombolysis or mechanical thrombectomy [2–4]. Computed tomography perfusion (CTP) has proven useful for assessing the extent of brain ischemia and for identifying “tissue at risk” that is potentially salvageable with recanalization. CTP also enables clinicians to optimize acute stroke therapy and to predict clinical outcome [5, 6]. The advantages of CTA and CTP over magnetic resonance-based techniques are that they are accessible in the emergency department, inexpensive, fast, and well tolerated. In addition, the spatial resolution of both techniques is high [6]. However, administration of nonionic contrast agents can lead to elevated serum creatinine levels, which are associated with poor outcome and increased mortality in patients with AIS [4, 7, 8], and contrast-induced nephropathy (CIN), which has been associated with higher morbidity and mortality in patients undergoing

Abstract: Cuando se trata de traducción científico-técnica o especializada, la investigación suele centrarse en un campo de especialidad y en la terminología que le es propia. Existe, sin embargo, un aspecto relacionado con esta modalidad de traducción poco considerado en general, pero de interés en el ámbito de la formación de traductores. Nos referimos a la información que proporciona el texto, a la clase textual como conjunto de niveles significativos. Este trabajo pretende mostrar que los datos textuales obtenidos en la fase interpretativa del texto de partida conforman el andamiaje sobre el que se estructura y ordena el sentido global del texto. ------------------------------------- The role of textual information in the process of reading specialized texts. Research on scientific-technical or specialized translation often centers on the specialist discipline and its terminology. However, one aspect related with this type of translation has generally received little attention, but is nonetheless of interest with regard to translator training: information provided by the text, i.e., the type of text as a set of levels of meaning. This study aims to show that information about the text obtained during interpretation of the source text constitutes the scaffolding upon which the overall meaning of the text is structured and ordered.
