In P. chabaudi and P. falciparum, melatonin induces calcium release from internal stores, and this response is abolished by U73122, a phospholipase C inhibitor, and luzindole, a melatoninreceptor competitive antagonist. Here we show that, in vitro, melatonin is not able to modulate cell cycle, nor to elicit an elevation in intracellular calcium concentration of the intraerythrocytic forms of P. berghei or P. yoelii, two rodent parasites that show an asynchrononous development in vivo. Interestingly, melatonin and its receptor do not seem to play a role during hepatic infection by P. berghei sporozoites either. These data strengthen the hypothesis that hostderived melatonin does not synchronize malaria infection caused by P. berghei and P. yoelii. Moreover, these data explain why infections by these parasites are asynchronous, contrary to what is observed in P. falciparum and P. chabaudi infections.Keywords: malaria, calcium, melatonin, cell cycle, rhythm, sporozoite

A Nature-Inspired Betalainic Probe for Live-Cell Imaging of Plasmodium-Infected Erythrocytes

Abstract: A model betalainic dye was semisynthesized from betanin, the magenta pigment of the red beet, and was effective for live-cell imaging of Plasmodium-infected red blood cells. This water-soluble fluorescent probe is photostable, excitable in the visible region and cell membrane-permeable, and its photophysical properties are not notably pH-sensitive. Fluorescence imaging microscopy of erythrocytes infected with Plasmodium falciparum, a causative agent of malaria in humans, showed that only the parasite was stained. Z-stacking analysis suggested that the probe accumulates proximal to the nucleus of the parasite. Indicaxanthin, one of the natural fluorescent betalains found in the petals of certain flowers, did not stain the parasite or the red blood cell.

Abstract: The article aims to analyze the phenomenon of "secular pilgrimage", or the number of trips and parades that during the first fifty years of Italian unity, saw the places of memory as the protagonists of the Risorgimento as ossuaries, battlefields, monuments and tombs of the fathers of the fatherland. Although the number of "pilgrims lay" is not never approached the numbers involved from the far more deep-rooted traditions and religious organizations, from many sources found emerges as the involvement of patriots, the number of sponsoring organizations, the "places of worship" and anniversaries (official or not) they have been, as a whole, particularly significant. Necessary starting point to introduce the topic will be the examination of the element vocabulary, borrowed from the religious. The rest of the text will have as its purpose the creation of a map - on a national scale - of the places of pilgrimage divided according to the "utilities". Correspond to different destinations, in fact, different types of pilgrimage, mirrors the realities of liberal policies.

Abstract: In the present work, it was demonstrated by mass spectrometry analysis that Plasmodium parasites (Plasmodium chabaudi and Plasmodium falciparum) internalize and process plasma kininogen, thereby releasing vasoactive kinins (Lys-BK, BK and des-Arg9-BK) that may mediate haemodynamic alterations during acute malaria. In addition, it was demonstrated that the P. falciparum cysteine proteases falcipain-2 and falcipain-3 generated kinins after incubation with human kininogen, suggesting that these enzymes have an important role in this process. The biologic activity of peptides released by Plasmodium parasites was observed by measuring ileum contraction and activation of kinin receptors (B1 and B2) in HUVEC cells; the peptides elicited an increase in intracellular calcium, measured by Fluo-3 AM fluorescence. This effect was suppressed by the specific receptor antagonists Des-Arg9[Leu8]-BK and HOE-140.In previously undescribed means of modulating host physiology, it was demonstrated that malaria parasites can generate active kinins by proteolysis of plasma kininogen.

Abstract:

Galileo introduced the scientific method, based on observation of natural phenomena, their interpretation and validation on the base of verified results. Albert Einstein broke the pattern of Galilean physics by postulating the invariance of the light speed and arguing that the laws of physics are valid independently of the inertial reference system used, introducing the concept of relativity and proposing a new physics based on four dimensions instead of the three previously known, with the addition of the variable time as the fourth dimension. In this article, we wonder if it is possible to find a methodology, necessarily not conventional, allowing to recognize additional new dimensions. The analysis of the quantization of nature has led us to hypothesize the use of irrational numbers in a ratio as a detector for any new dimension. A series of conjectures can bring us to imagine a scenario following the Big Bang with the creation of orthogonal matter, allowing us to find a seemingly plausible explanation to some unanswered questions of the contemporaneous physics. Under these hypotheses, it is possible to give two not conventional interpretations about gravity and light. The article prefers the logic explanation of the phenomena to their quantification through mathematics, reserving this last one only where it is necessary to clear the concept.

Does Life Need Water or Can It Be Generated by Other Fluids?  [PDF]

Abstract: In the present work the Stochastic generalization of the quantum hydrodynamic analogy (SQHA) is used to obtain the far-from-equilibrium kinetics for a real gas and its fluid phase. In gases and their liquids, interacting by Lennard-Jones potentials whose mean distance is bigger than the quantum correlation distance and the molecular interaction distance r0, it is possible to define a Fokker-Plank type equation of motion as a function of the mean phase space molecular volume that far-from-equilibrium shows maximizing the dissipation of a part of the generalized SQHA-free energy. In the case of a real gas with no chemical reactions with small temperature gradients, the principle disembogues into the maximum free energy dissipation confirming the experimental outputs of electro-convective instability. In this case, the model shows that the transition to stationary states with higher free energy can happen and that in incompressible fluids, the increase of free energy is almost given by a decrease of entropy leading to the appearance of self-ordered structures. The output of the theory showing that the generation of order via energy dissipation, is more efficient in fluids than in gases, because of their incompressibility, which leads to the reconciliation between physics and biology furnishing the explanation why the life was born in water. The theoretical output also suggests that the search for life out of the earth must consider the possibility to find it in presence of liquid phases different from water.
The Pillars of Learning, Understanding, Studying and Explaining  [PDF]

Abstract: This paper is based on the hypothesis that intelligent persons have a high capacity to understand and learn effectively, because processes involving study and explanation allow them to develop the capacity to construct a system of coherent models that can be quickly updated. The first part of the paper will highlight the logic and power of systemic understanding and learning that is founded in Systems Thinking. It is clear that understanding and learning represent different mental activities: We understand when we form a coherent model of a concept, but we learn only when that model becomes stable and permanent; when it is memorized so that we can repeat and apply it. There is no learning without studying. For this reason, the second part of the paper proposes ten steps for studying effectively (which permits understanding) and efficiently (using minimum effort). But how do we attain understanding? Often we reach understanding through personal experience; in most cases, however, we are helped by a professor, a teacher or a textbook that provides us with an explanation. The explanation process represents the topic of the third part of the paper, where the author considers the five most important structures of the explanation process—the common, classical, procedural, systemic and teleological structures. In this sense, Understanding, Studying and Explaining can be considered the three pillars on which are based learning and knowledge.
