Interaction Between Mevalonate Pathway and Retinoic Acid-Induced Differentiation

Abstract: All trans retinoic acid (ATRA) is a potent inducer of differentiation of HL-60 cell line. The pretreatment of the cells by compactin, a competitive inhibitor of 3-hydroxy-3-methylglutaryl (HMG) CoA reductase, during 24 hours, enhances the ATRA-induced cell differentiation. At 50 nM, the percentage of cell differentiation is 34.9% ± 2 and 73% ± 2.96 in the control and compactin-treated cells, respectively. The removal of compactin boosts the level of HMG-CoA reductase and therefore the biosynthesis of sterol and nonsterol isoprenoid compounds. The participation of sterol and nonsterol pathway was then investigated. The supply of an excess of cholesterol (up to 80 μg/ml of LDL) leads to a significant decrease of cell differentiation by ATRA from 78% ±0.1 to 54% ±2.8. A concomitant decrease of cell growth (51% ± 6.4) was observed. The pretreatment of cells by the geranylgeranyltransferase inhibitor (GGTI-298) has no effect on the cell differentiation process. By contrast, the farnesyltransferase inhibitors (FTI-II and FTI-277) completely abolish theATRA-induced differentiation, thus confirming the involvement of farnesylated proteins in the differentiation mechanism.

Long-Term Risperidone Treatment Induces Visceral Adiposity Associated with Hepatic Steatosis in Mice: A Magnetic Resonance Approach

Abstract: Although atypical antipsychotic drugs (APDs) have led to significant advances in the treatment of psychotic disorders, they still induce metabolic disturbances. We aimed at characterizing the metabolic consequences of a risperidone treatment and at establishing a link with noninvasive MR markers, in order to develop a tool for predicting symptoms of the metabolic syndrome. Fat deposition and liver morphometry were assessed by T1-weighted imaging. Fatty acid composition and fat accumulations in tissues were determined using MR spectroscopy with and without water suppression, respectively. Risperidone treatment induced a weight gain accompanied with metabolic disturbances such as hyperglycemic status, an increase in visceral adipose tissue (VAT), and liver fat depositions. Correlations using Methylene-Water Ratio (MWR) and Polyunsaturated Index (PUI) demonstrated a concomitant increase in the weight gain, VAT and liver fat depositions, and a decrease in the quantity of polyunsaturated fatty acids. These results were consistent with a hepatic steatosis state. We evaluated the ability of MR techniques to detect subtle metabolic disorders induced by APDs. Thus, our model and methodology offer the possibility to investigate APDs side effects in order to improve the health conditions of schizophrenic patients. 1. Introduction Antipsychotic drugs (APDs) are widely used in current psychiatric practice and are commonly classified as typical (conventional) or atypical (second generation). Atypical APDs have been introduced in clinical practice after 1990, including clozapine, olanzapine, quetiapine, and risperidone. Atypical APDs cause less extrapyramidal symptoms (tremors) than typical APDs. However, both of them produce a weight gain [1–3], which increases the risk to develop a metabolic syndrome [4] associating several disorders such as diabetes mellitus, hypertension, hyperglycemia, dyslipidemia, and abdominal fat deposition [4, 5]. The excess of visceral adipose tissue (VAT) mass is particularly correlated to the prevalence of metabolic syndrome and insulin resistance [6]. Indeed, abnormal VAT depositions lead to the storage of lipids in undesired organs such as pancreas, skeletal muscle, heart, and liver. This so-called “ectopic fat deposition” contributes to the development of metabolic syndrome [6–12]. VAT accumulation could thus represent a biomarker of metabolic disturbances, as used in clinic through the measurement of waist circumference in replacement of the body mass index [5]. Bicaudal Is a Conserved Substrate for Drosophila and Mammalian Caspases and Is Essential for Cell Survival

Abstract: Members of the caspase family of cysteine proteases coordinate cell death through restricted proteolysis of diverse protein substrates and play a conserved role in apoptosis from nematodes to man. However, while numerous substrates for the mammalian cell death-associated caspases have now been described, few caspase substrates have been identified in other organisms. Here, we have utilized a proteomics-based approach to identify proteins that are cleaved by caspases during apoptosis in Drosophila D-Mel2 cells, a subline of the Schneider S2 cell line. This approach identified multiple novel substrates for the fly caspases and revealed that bicaudal/βNAC is a conserved substrate for Drosophila and mammalian caspases. RNAi-mediated silencing of bicaudal expression in Drosophila D-Mel2 cells resulted in a block to proliferation, followed by spontaneous apoptosis. Similarly, silencing of expression of the mammalian bicaudal homologue, βNAC, in HeLa, HEK293T, MCF-7 and MRC5 cells also resulted in spontaneous apoptosis. These data suggest that bicaudal/βNAC is essential for cell survival and is a conserved target of caspases from flies to man.
A Case Crossover Analysis of Out-of-Hospital Cardiac Arrest and Particulate Matter Air Pollution: Investigation of Specific Subgroups  [PDF]

Abstract:
There is increasing evidence of association between particulate matter air pollution and cardiovascular mortality and morbidity. However, the association with the out-of-hospital cardiac arrest (OHCA) event is less clear. We investigated the effects of short-term particulate matter exposure on OHCA especially among specific subgroups. The study included OHCA that occurred in the Nord-Pas-de-Calais region, France, in 2015. A time-stratified case-crossover study design coupled with a conditional logistic regression was used to evaluate the association between OHCA and particle levels of 10 or 2.5 micrometers or less (PM10 or PM2.5 respectively) measured within the hour of the arrest up to 5 days before. Susceptible subgroups by sex, age, diabetes status among OHCA that occurred during non-holiday periods were investigated. In all, 1039 cases were included. Significant associations were found between OHCA during non-holiday periods and PM2.5 and PM10 exposure four days before the arrest and on the day of the arrest. The largest OR were found for the cumulative average twelve hours before the arrest of PM2.5 (OR = 1.17, p = 0.016) and PM10 (OR = 1.33, p < 0.001). With PM2.5, larger OR with smaller p-values were generally obtained within the subgroups of men, age 50 to 75 years old and cases with diabetes. The findings show a significant link between short-term exposure to particulate matter and OHCA during non-holiday periods, with susceptible subgroups to PM2.5 (men, age 50 to 75 years old and diabetics).

Ils exposent ensuite ce que ce résultat doit à une socialisation réciproque, entre coopération et conflits d’interprétation. Puis ils présentent diverses difficultés rencontrées et les solutions trouvées pour que cette coproduction reste effective à tous les stades du processus – de la définition des thèmes des journées d’étude à l’écriture des Cahiers – incluant l’organisation pratique et la fabrication des livres. Enfin, ils notent qu’une des conditions de réussite d’un tel partenariat interactif et contractuel est sans doute qu’il était exclusivement centré sur la production de connaissances historiques, sans visée transformatrice, quoique non sans effets sur la représentation des organisations actuelles de leur propre histoire. Misleading pustular plaques of the lower limbs during Crohn's disease: two case reports

Abstract: On both patients, differential diagnosis of neutrophilic dermatoses and infectious disease were evoked, and antimicrobial agents were introduced in one of them. Given the atypical presentation, the final diagnosis of cutaneous Crohn's disease could only be made with histological examination. In patient 1, the plaques decreased in size and infiltration by more than 75% after 3 weeks of treatment with bethametasone dipropionate 0.05% cream. In patient 2, the plaques decreased by more than 50% after 6 weeks of treatment with prednisolone (45 mg/day) and azathioprine (100 mg/day).Cutaneous Crohn's disease may present as dusky, erythematous, infiltrated, and ulcerated plaques and nodules. Female-to-male sex ratio is about 2, and the mean age at onset is 35. Recurrently, the hypothesis of a skin mycobacterial or fungal infection greatly delays proper treatment. Rarity of cutaneous Crohn's disease hampers therapeutic assessment in controlled trials. Thus, available literature is limited to case reports and sparse small series, with contradictory results. These reports are subject to publication bias, and no definite evidence-based recommendations can be made on the most adequate therapeutic strategy.Crohn's disease is characterized by a chronic relapsing transmural granulomatous inflammation that may involve any part of the digestive tract from mouth to anus, though mostly found in the ileum, the cecum, and the colon. Abstract: Le forage de puits profonds exige la mise au point de fluides de forage dont la formulation est adaptée aux températures élevées de formation : l'une des principales difficultés est la dégradation thermique des additifs chimiques utilisés dans les formulations à base d'eau qui se produit fréquemment lors du forage de puits à température élevée. Cette dégradation peut conduire à des variations importantes des caractéristiques rhéologiques et de filtration et à une perte de propriétés nécessaires à leur performance. De plus, même s'il n'y a pas de dégradation des composants la viscosité des solutions de polymères hydrosolubles couramment utilisés dans les formulations de fluides diminue fortement avec l'augmentation de la température au-dessus de 60°C. Une autre source de difficulté qui appara t à forte température avec les boues à base d'eau est la gélification des argiles utilisées dans la formulation. Ce phénomène provoque une forte augmentation de la viscosité de la formulation et des pertes de charges, en particulier en cas de reprise de forage. Diverses méthodes ont été mises au point pour étudier le comportement des boues à base d'eau dans des conditions de pression et de température élevées, entre autres : - des expériences de laboratoire, pour étudier par exemple la stabilité en conditions anaérobies des solutions de polymères et le comportement rhéologique des suspensions d'argile dans des conditions de pression et de température élevées afin de simuler les caractéristiques de gélification des argiles à haute température; - des essais sur boucle d'étude HP/HT en utilisant une boucle expérimentale con ue pour étudier les fluides de forage dans des conditions de fond réalistes, c'est-à-dire des températures atteignant 180°C, des pressions atteignant 500 bar et des taux de cisaillement atteignant 10 000 s puissance( -1) Cette méthodologie a permis d'étudier un certain nombre de boues à base d'eau. Les résultats font appara tre que la nature des composants de la formulation exerce une forte influence sur le comportement de la boue dans des conditions difficiles. Le choix approprié d'additifs, tels que viscosifiants et réducteurs de filtrat, peut limiter les risques inhérents aux conditions de forage difficiles. Abstract: Un des objectifs de la politique environnementale de l'Union européenne est de réduire les émissions des véhicules automobiles. Les normes Euro 96 et Euro 2000 s'intéressent en particulier aux émissions par évaporation dues à la perméation du carburant à travers les matériaux polymères. Dans ce papier, nous exposons la méthode d'essai des lignes de carburant développée à l'IFP avec PSA et Renault. Nous présentons également des résultats obtenus avec divers types de canalisations et plusieurs fluides d'essai. Au vu de ces résultats et de la sévérité des nouvelles normes, les équipementiers et les fournisseurs de matières doivent développer de nouvelles solutions afin d'améliorer la résistance à la perméation. The aim of the environmental policy of the European Union is to reduce the automotive hydrocarbon emissions. The Euro 96 and future Euro 2000 standards are particularly dealing with the emissions due to fuel permeation through polymeric materials. In this paper, the testing procedure for fuel lines developed at IFP, PSA, and Renault, is described. Results obtained with various types of fuel lines and several fuels are detailed. According to these results and to the new stringent standards, equipment and material suppliers will have to develop new solutions with improved permeation resistance.
