Preparation of monospecific anti-PAG antibodies for cattle pregnancy detection: use of synthetic peptides to improve specificity  [PDF]

Abstract: Immunological methods involving pregnancy associ-ated glycoproteins (PAG) are used for cattle preg-nancy detection. The faults of these methods could be overcomed by using antibodies specific for each member of the PAG family. In order to differentiate between very similar proteins, preparation of anti-bodies specific for peptides is a method of choice. In this work, we summarize a series of considerations regarding peptide design and choose free access NCBI, Antigenicity Plot, EMBOSS Antigenic and Expasy tools to apply them. We design peptides spe-cific for different reported PAG members and obtain the corresponding polyclonal antibodies for five of them.

Abstract: Los catéteres tunelizados (CT) constituyen el acceso vascular de hemodiálisis (HD) en un porcentaje elevadísimo de pacientes. La utilización de CT se asocia a dos problemas fundamentales: disfunción e infecciones. Según las guías de la Sociedad Espa ola de Nefrología (SEN), en los pacientes en HD se aconseja el empleo de tapones de un solo uso con rosca de seguridad (luer-lock). En el mercado existen también bioconectores, que son dispositivos mecánica y microbiológicamente cerrados. La ventaja que ofrecen es la menor manipulación de la luz del catéter al cambiarse una vez por semana. Se realizó un estudio en nuestro centro analizando en una muestra de 14 pacientes portadores de CT, dos periodos consecutivos, un primer periodo siguiendo el protocolo de conexión convencional y el segundo utilizando los bioconectores TEGO. Se analizaron presiones arterial y venosa, flujos sanguíneos, Kt, Kt/v y tasa de bacteriemia. No se obtuvieron diferencias significativas en estos parámetros con el uso de dispositivos TEGO frente al uso convencional. Podemos concluir que el uso de tapones TEGO no afecta a los parámetros dialíticos. No obstante, debemos considerar el manejo de cualquier tipo de dispositivo como secundario en la prevención de bacteriemias, ya que se ha demostrado que son las medidas universales de asepsia las que logran una tasa de bacteriemias muy baja. Tunnelled catheters (TC) are the vascular access used in haemodialysis (HD) for a very high percentage of patients. The use of TC is associated to two fundamental problems: dysfunction and infections. According to the guidelines established by the Spanish Nephrology Society (SEN), the use of luer-lock catheters is recommended in HD patients. On the market there are also bioconnectors, which are mechanically and microbiologically sealed devices. The advantage they offer is better handling of the catheter lumen when changed once a week. A study was carried out at our centre analysing two consecutive periods in a sample of 14 patients with CT. In the first period the conventional connection protocol was followed and in the second TEGO bioconnectors were used. Arterial and venous blood pressure was measured, blood flows, Kt, Kt/v and bacteraemia rate. No significant differences were obtained in these parameters with the use of TEGO devices compared to conventional use. We may conclude that the use of TEGO locks does not affect the dialytic parameters. Abstract: the objective of this work was to investigate the insulin sensitivity (si) and the beta cell function (bcf) in different glucose tolerance statuses in a comparative cross-sectional study. eighty-four patients with different glucose tolerance statues were classified as normoglicemic-control group (nc) patients without family history of type 2 diabetes (dm2) or hypertension (htn); normoglicemic-patients with familiar history of dm2 or htn (fpdm2); patients with glucose intolerance (igt group) and patients with diagnostic of dm2 < 10 years (dm2 group). in each patient was obtained a clinical history and an oral glucose tolerance test (75g) was performed. glucose (god-pap) and insulin (ria) were quantified. insulin sensitivity (is) (whole body insulin sensitivity index), insulin resistance (ir) (homa) and bcf (insulinogenic index) were evaluated. the statistical analysis was realized in spss v. 10.0. it was found that glucose was similar among nc, fpdm2 and igt groups. insulin was higher in the fpdm2 (26.71 ± 22.25 μu/ml), and igt (34.10 ± 34.98 μu/ml) in comparison with nc (22.83 ± 21.26 μu/ml) (p < 0.01). is was different among nc and igt group (0.74 ± 0.43 to 0.29 ± 0.18, p < 0.05) and among igt and dm2 (0.29 ± 0.18 to 0.86 ± 0.55, p < 0.001). bcf was different among the dm2 and nc (0.051 ± 0.05 to 1.28 ± 1.99 μu.ml/mg.dl, p < 0.05), dm2 and fpdm2 group (0.051 ± 0.05 to 1.23 ± 1.51μu.ml/mg.dl, p < 0.01), dm2 and igt (0.051 ± 0.05 to 2.64 ± 2.22 μu.ml/mg.dl, p < 0.001). si was inversely related to corporal weight and imc (r = -0.38, p = 0.001 and r = -0.33, p = 0.004, respectively). bcf was related to age (r = -0.27, p = 0.016), the area under curve of glucose (r = -0.30, p = 0.010) and body weight (r = 0.34, p = 0.002). in conclusion, the decrease of glucose tolerance was associated with the lowering of insulin sensitivity (is). the beta cell function (fcb) was lower in the diabetic patients group in comparison with the tga and fpdm2 groups. these measurements need t

Abstract: El objetivo de este estudio fue investigar la sensibilidad a la insulina (SI) y la función de la célula beta (FCB) en los diferentes estados de tolerancia a la glucosa. Se trata de un estudio transversal comparativo. Se estudiaron 84 sujetos con diferentes estados de tolerancia a la glucosa y se clasificaron en cuatro grupos: Normoglicémico-Control (NC) que fueron individuos sin antecedentes de diabetes mellitus tipo 2 (DM2) o hipertensión (HTN); Normoglicémico-Familiares con DM-2 y/o HTN (FPDM2); el grupo con tolerancia a la glucosa alterada (TGA) y el grupo con DM2 diagnosticado con un tiempo < 10 a os. A cada sujeto se le realizó historia clínica y prueba de tolerancia oral a la glucosa (75g). Se cuantificaron glucosa (método GOD-PAP) e insulina séricas (técnica de radioinmunoanálisis). Se estimó la sensibilidad a la insulina (SI) (índice de sensibilidad corporal de la insulina, WBISI), resistencia a la insulina (RI), (HOMA) y la función de la célula beta (FCB, índice insulinogénico). Los coeficientes de variación intra e interensayo fueron menores al 5%. Los datos se analizaron en el SPSS v 10.0. Se observó que la glucosa fue similar entre el NC, el FPDM2 y el TGA. La insulina fue mayor en el FPDM2 (26,71 ± 22,25 μU/mL) y TGA (34,10 ± 34,98 μU/mL) en relación al grupo NC (22,83 ± 21.26 μU/mL) (p < 0,01). Se encontró diferencia en la SI entre el NC y el TGA (0,74 ± 0,43; 0,29 ± 0,18, respectivamente) (p < 0,05) y entre el TGA y el DM2 (0,29 ± 0,18; 0,86 ± 0,55, respectivamente) (p < 0,001). La FCB fue diferente entre el grupo DM2 y el NC (0,051 ± 0,05 vs 1,28 ± 1,99 μU.mL/mg.dL, p < 0,05), entre el DM2 y el FPDM2 (0,051 ± 0,05 vs 1,23 ± 1,51 μU.mL/mg.dL; p < 0,01) y entre el DM2 y el TGA (0,051 ± 0,05 vs 2,64 ± 2,22 μU.mL/mg.dL, p < 0,001). La SI se relacionó negativamente con el peso corporal y el IMC (r = -0,38, p = 0,001, r = -0,33, p = 0,004) y la FCB con: edad (r = -0,27, p = 0,016), area bajo de la curva (ABCG) (r = -0,30, p = 0,010) y peso (r = 0,34, p = 0,002). En conclusión, la disminución de la tolerancia a la glucosa se asoció con baja sensibilidad a la insulina (SI). La función de la célula beta (FCB) fue menor en el grupo de pacientes con diabetes comparados con el grupo TGA y con el de FPDM2. Abstract: This article describes the results of an investigation which have been gotten in the “Family and development human investigation Group” about the axis of “Socialization of the Families in a high vulnerability” marked in the investigation line “The family in the social economic and politic context: Changes and Permanence”. The main theoretical references have been authors like Aguirre, Zuleta and Daza, being the families and the children of the “Historical Center” from the city, the people who has been boarded. These results show us difficult in the relations and communication, as inside the familiar group as their neighbours. It doesn’t have any limits and fair rules, it doesn’t share activities between fathers and sons, dedicating each member of the family to their own activities, guide by particular interest.
Efficacy of adalimumab in patients with crohn's disease and failure to infliximab therapy: a clinical series

Abstract: background: adalimumab, a human anti-tnf, is an effective induction and maintenance therapy for patients with moderate to severe crohn's disease. it seems to be effective in patients with resistance to infliximab, too, though the experience is more limited. aim: to evaluate the efficacy of adalimumab, in patients with crohn's disease (cd) and failure to previous treatment with infliximab. b twenty-five patients with cd and failure to previous treatment with infliximab were enrolled; they were treated with 160/80 (24 patients) and 80/40 (1 patient) induction doses. we analyze clinical response to treatment with adalimumab by the crohn's disease activity index (cdai) and plasma concentration of c-reactive protein (crp), steroid sparing and complete fistula closure at week 48. results: eighteen out of twenty-five patients (72%) achieved clinical remission (cdai score < 150) at week 24 and 15/25 (60%) patients at week 48. there was a statistically significant difference (p < 0.01) in crp serum levels from 21 to 8 mg/dl at week 48. nine out of fifteen patients (60%) treated with corticosteroids were able to discontinue steroids. three out of eleven patients (27%) with fistulizing crohn's disease had complete fistula closure after the treatment. seventy two percent of the patients (18/25) needed to increase adalimumab to weekly dose, in order to maintain clinical response. five out of twenty-five patients (20%) had adverse events; two of them (8%) with serious adverse events (tuberculous meningitis and abdominal abscess) that forced the withdrawal of treatment. conclusions: according to these data, adalimumab provides a clinical and analytical improvement in patients with cd and failure to previous therapy with infliximab.

Abstract: this descriptive study was carried out using questionnaires to analyze the attitudes toward mathematics by engineering students at autonomous venezuelan univer - sities. the population under study consisted of students studying in engineering faculties in the first semester of 2004, giving a total of 5,931 students. to select the sample, a stratified sampling by conglomerates was applied accepting a level of reliability of 95%. the sample was thus reduced to 613 participants whose ages ranged between 15 and 30. the results obtained through factorial analysis of the main components of an exploratory nature with varimax rotation pointed to a coherent and parsimonious structure made of three components: likes, difficulty and utility.

Abstract: Neurospora crassa es un modelo de ritmicidad circadiana ampliamente estudiado. El metabolismo de este hongo es controlado por múltiples factores que incluyen el desarrollo, las características del medio y el reloj circadiano. El estudio de la regulación circadiana del metabolismo en este hongo podría estar enmascarado por el uso de medios restrictivos que suprimen su crecimiento y desarrollo. En el presente trabajo se estudió la variación en el tiempo de la actividad de la enzima Succinato Deshidrogenasa durante 24 horas en medio completo sin restricción para la conidiación en cepas de Neurospora crassa rítmica y arrítmica. Se encontró una variación circadiana en la actividad enzimática de la cepa rítmica con valores altos en las horas correspondientes a la noche y bajos durante el día. Este hallazgo resalta la importancia de profundizar en el estudio del control circadiano sobre el metabolismo de este hongo, con base en la existencia de vías de regulación múltiples sobre las enzimas del metabolismo y de mecanismos post-transcripcionales y transcripcionales de control circadiano. Neurospora crassa is a widely studied model of circadian rhythmicity. In this fungus, metabolism is controlled by multiple factors which include development, medium characteristics and the circadian clock. The study of the circadian control of metabolism in this fungus could be masked by the use of restrictive media that inhibit growth and development. In this report, the presence of a circadian rhythm in the activity of the enzyme Succinate Dehydrogenase in Neurospora crassa is demonstrated. Rhythmic and arrhythmic Neurospora strains were grown in complete medium without conidiation restriction. A circadian change in the enzymatic activity was found with high values in hours corresponding to the night and a low level during the day. This finding highlights the importance of deeper studies in the circadian control of metabolism in this fungus, given the existence of multiple pathways of regulation of metabolic enzymes and a circadian clock control at the transcriptional and post-transcriptional levels.
