c-Jun N-terminal kinase (JNK) cooperates with Gsk3β to regulate Dishevelled-mediated microtubule stability

Abstract: Here we show that the Wnt pathway also activates c-Jun N-terminal kinase (JNK) to regulate MT stabilization. Although in the Wnt planar cell polarity (PCP) pathway, JNK lays downstream of Rho GTPases, these GTPases are not required for Wnt-mediated MTs stability. Epistatic analyses and pharmacological studies suggest that the Wnt-Dvl signalling regulates the dynamic of the cytoskeleton through two different pathways that lead to inhibition of Gsk3β and activation of JNK in the same cell.We demonstrate a novel role for JNK in Wnt-mediated MT stability. Wnt-Dvl pathway increases MT stability through a transcription independent mechanism that requires the concomitant inhibition of Gsk3β and activation of JNK. These studies demonstrate that Wnts can simultaneously activate different signalling pathways to modulate cytoskeleton dynamics.Regulation of the MT cytoskeleton is crucial for diverse cellular functions such as cell division, cell polarity, migration and morphogenesis. In the nervous system, regulation of the dynamic and organization of MTs is essential for the initiation, extension and maintenance of neuronal processes. MT arrays are also used for the transport of organelles and molecules along axons and dendrites. MTs are dynamic polymers that can polymerize and depolymerise very rapidly and this dynamic instability allows cells to respond quickly to extracellular signals. Although great progress has been made in understanding how intracellular molecules regulate the dynamics of MTs [1-3], very little is known about the mechanisms by which extracellular signals and their pathways modulate MT behaviour.The Wnt signalling pathway has been shown to directly regulate the cytoskeletal network by regulating both the dynamics and organization of MTs [4,5]. Wnts can function as molecules that guide axons to their appropriate targets [6-8] but they can also function as target-derived signals to regulate the terminal arborisation of axons [9,10]. Abstract: in order to stablish the current prevalence by eggs of toxocara canis, 110 squares of five zones of the metropolitan region of santiago city were examined by a flotation in saturated zinc solution modified method. samples were taken from different kinds of soil in the four seasons of the year and they were preserved under anaerobic conditions over 2 years. 18,2% of samples resulted positive to toxocara canis eggs and 26.4% were positive to nematodes larvae. no viable eggs were found. western zone is the most contaminated of the city and autumn is the season in which we obtained higher recovery of eggs. silty clay sample soil seems to be better than sandy soils on maintenance of studied eggs.

Abstract: studies by renowned investigators on the problematic of teaching and learning calculus enable us to recognize the traditional teaching paradigm that has been practiced in the educative institution to which we belong. the revision of diverse articles on teaching alternatives suggests tendencies in the way changes are proposed; some only affect the teaching method, others consider the content to be taught. we observe that resorting to the history of the genesis of knowledge has permitted the identification of a variable in the mathematical content of the curriculum which influences the acquisition of the notions and procedures of calculus. at some point, the what to teach becomes part of how to teach and from there mathematics activity takes on a didactic sense in the classroom. in the framework of the socio-epistemological approach we find the emergence of a proposal for the teaching of calculus, which implementation in the educative institution in question is based on research.

Abstract: Introducción: en la práctica resulta complejo determinar la prevalencia de anticuerpos antivirus de hepatitis B (VHB), C (VHC) y VIH en la población general, dado que la relativa baja presencia de anticuerpos en algunos países exigiría tama os de muestra muy grandes. Por ello, los estudios de seroprevalencia realizados en bancos de sangre, como este de la ciudad de Corrientes, aportan para el conocimiento de la afectación sintomática o no de estas enfermedades. Datos obtenidos del Sistema Nacional de Vigilancia en el Boletín Epidemiológico Nacional revelan que Argentina, en 2001 registraba una incidencia de 2,1 casos de VHB notificados por 100 000 habitantes, 1,3 x 100 000 para el VHC y 2,9 casos de VIH cada 100 000 habitantes. Objetivos: determinar la prevalencia en el Banco de Sangre Central de Corrientes, mediante pruebas serológicas, de anticuerpos antivirus de hepatitis B, C y virus de inmunodeficiencia humana en donantes o potenciales donantes de sangre. Métodos: para conocer la prevalencia de anticuerpos contra los agentes de transmisión parenteral mencionados, en el Banco de Sangre Central de Corrientes, se examinaron 41 503 fichas de donantes o potenciales donantes. Se realizaron estudios serológicos completos entre los que se incluyó: detección de HBsAg (ELISA), Anti-VHC (ELISA) y VIH (ELISA Anti-VIH confirmación con Western Blot). Resultados: la prevalencia de hepatitis B fue de 0,38 %, de hepatitis C de 0,41 % y la de VIH de 0,03 %. Conclusiones: las cifras obtenidas resultaron sustancialmente mayores que las reportadas en el Boletín Epidemiológico Nacional, pues aunque este refleja tasas de incidencia, no detecta a los afectados asintomáticos y en Argentina todavía se adolece de incompleta notificación. Background: It is quite complex to determine in practice the prevalence of antibodies to hepatitis B, C and HIV in the general population, given that the relative low presence of antibodies in some countries would demand very large samples. Therefore, the seroprevalence studies performed in blood banks like that of Corrientes city, contribute to knowing about the symptomatic effect or not of these diseases. Data taken from the National Surveillance System that appeared in the National Epidemiological Bulletin reveal that Argentina had in 2001 an incidence rate of 2,1 cases of notified HBV cases, 1,3 HCV cases and 2.9 HIV cases per 100 000 pop. respectively. Abstract: an elisa test for trichinosis using as antigen a larvae soluble fraction from trichinella spiralis was carried out for the detection of igmand iga specific antibodies in 45 serum samples from patients confirmed or suspected to have trichinosis by strong clinical and epidemiological evidences. all the patients had positive serology detected by precipitin test, bentonite floculation test, indirect hemagglutination test and elisa igg test. the cut-off value was determined using two criteria. criterion a was determined in each plate, using three positive controls and two negative ones; the average of the negative controls and the weakest positive control, muliplied by a 1.2 factor was, considered the cut-off value. criterion b was determinated using the average plus three standard deviations from 64 apparently healthy persons serum samples. in both cases, three serum dilutions (1:10, 1:100 and 1:500) were used. the sensitivity of elisa igm was 100.0, 93.3 and 82.2% using serum dilutions of 1:10, 1:100 and 1:500 respectively (criterion a) and 100.0, 97.8 and 95.6% for the same dilutions (criterion b), whereas the values for elisa iga were: 100.0, 91.1 and 86.7% (criterion a) and 100.0, 100.0 and 91.1%(criterion b). in order to find out the specificity of elisa igm and elisa iga, additional118 serum samples from individuals with other parasitoses, such as cysticercosis (18) hydatidosis (39), fascioliasis (12), toxocariasis (30), chagas' disease(12) and individuals with non-specif eosinophilia (7), were also tested. elisa igm presented a specificity of 92.3, 93.4 and 97.3% (criterion a) and 96.2, 97.8 and 97.8% (criterion b) whereas the results for elisa iga were 97.8, 98.9 and 99.4% (criterion a) and 98.4% for the 1:10 and 1:100 dilutions and 100.0% for the 1:500 dilution (criterion b). the positive predictive values of elisa igm were 76.3, 77.8 and 88.1% (criterion a) and 86.5, 91.7 and 91.5% (criterion b) whereas the negative ones were 100.0, 98.3 and 95.7% (criterion a
