Abstract: Royal palm tree peroxidase (RPTP) has been isolated to homogeneity from leaves of Roystonea regia palm trees. The enzyme purification steps included homogenization, (NH4)SO4 precipitation, extraction of palm leaf colored compounds and consecutive chromatography on Phenyl-Sepharose, TSK-Gel DEAE-5PW and Superdex-200. The novel peroxidase was characterized as having a molecular weight of 48.2 ± 3.0 kDa and an isoelectric point pI 5.4 ± 0.1. The enzyme forms dimers in solution with approximate molecular weight of 92 ± 2 kDa. Here we investigated the steady-state kinetic mechanism of the H2O2-supported oxidation of different organic substrates by RPTP. The results of the analysis of the initial rates vs. H2O2 and reducing substrate concentrations were seen to be consistent with a substrate-inhibited Ping-Pong Bi-Bi reaction mechanism. The phenomenological approach used expresses the peroxidase Ping-Pong mechanism in the form of the Michaelis-Menten equation and affords an interpretation of the effects in terms of the kinetic parameters KmH2O2, KmAH2, kcat, KSIH2O2, KSIAH2 and of the microscopic rate constants k1 and k3 of the shared three-step peroxidase catalytic cycle. Furthermore, the concentration and time-dependences and the mechanism of the suicide inactivation of RPTP by hydrogen peroxide were studied kinetically with guaiacol as co-substrate. The turnover number (r) of H2O2 required to complete the inactivation of the enzyme was 2154 ± 100 and the apparent rate constants of catalysis 185 s–1 and 18 s–1.

Fitosanidad , 2010,
Abstract: thirty five single spore isolates of guignardia citricarpa (gm, phyllosticta citricarpa) from black spot symptoms and four from g. mangiferae (gm; phyllosticta capitalensis) associated to black dots (non pathogenic) were obtained in the rind of citrus fruits. the growth and morphology of the colonies, teleomorph and anamorph structures morphology and dimensions in both fungi were studied in pda, oatmeal agar (aa) and v8 juice agar (v8). a principal components and cluster statistical analysis were performed to determine the characteristics that contribute the most to species differentiation. it were concluded that both species are concurrent in citrus fruits in cuba and that the most important characteristics to differentiate them were growth in pda, the perithecia development in vitro, the minimal size of the conidiostrome and the production and release of a yellow pigment in v8: gc is slow growing in pda, develop picnidia abundantly in media, the conidiostrome is frequently smaller than 145 μm, do not develop perithecia in pda (heterotallic) and release a yellow pigment in v8. gm es fast growing in pda, frequently has a conidiostrome higher than 145 μm, develops perithecia in single spore cultures (homotallic) and do not produce and release a yellow pigment in v8. those characteristic allow the differentiation of both species in citrus plants.

Revista de la Facultad de Ciencias Veterinarias , 2006,
Abstract: the aim of this study was to standardize and evaluate the efficacy of the indirect immunofluorescent technique (iift) for early detection of venezuelan equine encephalitis virus (veev) antigen in the brain of suckling mice, using the complement fixation test (cft) as a comparison. viral antigen tc-83 was detected at 10, 12 and 26 hours post-inoculation (hpi), for p-676, lara-95 and tc-83 strains, respectively. the sensitivity of iift for lara-95 strain, was 89% at 24 hpi (non symptomatic period), when compared to 32% for cft, indicating that the low concentration of viral antigen during the early phases of the disease, can be detected with iift. the sensitivity of both techniques compared to the biological assay was 86% and 29% for iift and cft, respectively. at 36 hpi in mice with evident symptomatology, the sensitivity and specificity of both test was 100%. these results indicate that the iift is a valid and efficient test to evaluate out breaks of venezuelan equine encephalitis, in order to contributing to epidemiological surveillance of the disease.

The aim of this study was to standardize and evaluate the efficacy of the indirect immunofluorescent technique (IIFT) for early detection of Venezuelan Equine Encephalitis Virus (VEEV) antigen in the brain of suckling mice, using the Complement Fixation Test (CFT) as a comparison. Nutrición Hospitalaria , 2011,
Abstract: Background: In 2007, the Hospital Infantil Los ángeles (HILA) in Colombia implemented a slightly-modified version of the WHO guidelines for the diagnosis and management of malnutrition during childhood. Objective: To evaluate the efficacy of the WHO-HILA protocol in children hospitalized with severe, chronic marasmus and kwashiorkor malnutrition (MS-KWK) in 2007 and 2008. Material and methods: In this descriptive retrospective study the records of 100 children hospitalized with MSKWK were initially evaluated. Of these, 30 fulfilled the inclusion criteria: children of both sexes with a primary diagnosis of MS-KWK. Patients with any chronic disease liable to cause malnutrition were excluded. Anthropometric parameters, clinical signs and biochemical indicators of malnutrition were assessed upon admission and again at discharge following application of the WHO guidelines. Univariate analysis was performed for each study variable; serum hemoglobin and albumin levels on admission and at discharge were compared, and data were subjected to bivariate analysis. Results: Marasmus was diagnosed in 23.3% of children, kwashiorkor in 73.3% and marasmic kwashiorkor in 3.3%. The major clinical findings were: edema (70%), emaciation (40%), "flag sign" hair (42.86%), low serum albumin (93%) and anemia (80%). Thirteen children following the WHO-HILA protocol showed a significant nutritional status improvement (p < 0.05), whereas no improvement was noted in the 17 children not treated according to the protocol. Conclusions: Application of the WHO-HILA protocol was associated with reduced morbimortality in children with marasmus-kwashiorkor malnutrition. Implementation of this protocol should therefore be considered in all children′s hospitals in countries where this disease is prevalent.
