Synthesis and evaluation of anti-leishmania activity of analogs isoxazoles derivatives of grandisin and veraguensin neolignans Síntese e avalia o da atividade anti-Leishmania de análogos isoxazólicos derivados das neolignanas grandisina e veraguensina

Abstract: Using the concept of bioisosterismo, the new analogs isoxazole were designed from the molecular modification of grandisin and veraguensin neolignans, which have a grouping that is a furan ring bioisóstero isoxazole. In order to obtain more potent compounds, and better physical and chemical characteristics, our research group synthesized six analogues isoxazole neolignans. These reactions were performed of 1,3-dipolar cycloaddition between oximes chlorine and terminal acetylenes previously synthesized. The reaction system CuSO4.5H2O/Ascorbate Sodium / KHCO3, CH2Cl2/THF was used, and under this condition were obtained isoxazole six analogues with yields ranging from 71% To 90%. The compounds were sent for testing anti-Leishmania activity and anti-trypanosome. Utilizando o conceito de bioisosterismo, novos análogos isoxazólicos foram planejados a partir da modifica o molecular das neolignanas grandisina e veraguensina as quais possuem um grupamento furanico que é um bioisóstero de anéis isoxazólicos. Visando a obten o de compostos mais potentes, e melhores características físico-químicas, nosso grupo de pesquisa sintetizou seis análogos isoxazólicos neolignanicos. Foram realizadas rea es de cicloadi o 1,3-dipolar entre cloro oximas e acetilenos terminais previamente sintetizados. O sistema reacional CuSO4.5H2O/Ascorbato de Sódio/ KHCO3, CH2Cl2/THF foi utilizado e, sob esta condi o foram obtidos seis análogos isoxazólicos com rendimentos que variaram de 71 % a 90 %. Os compostos foram encaminhados para teste de atividade anti-Leishmania e anti-tripanossoma.

Abstract:

Galectin-3 is a member of the lectin family that binds β-galactosides and plays an important role in several types of tumors. Melanoma is an invasive cancer responsible for 80% of deaths associated to skin cancers. There are some evidences that galectin-3 interacts with β-catenin, a molecule involved with Wnt signaling pathway. Here, we evaluate the role of galectin-3 intumor growth and metastasis, as well as its interaction with β-catenin. Murine melanoma cells (B16F10) were injected subcutaneously and intravenously in male C57BL/6 wild-type (WT) and galectin-3 knock-out (KO) mice. Tumor growth and lung melanoma colonies were assessed. The expression of galectin-3 and β-catenin was evaluated by immuno-histochemistry. We observed that tumor growth did not differ between the groups. However, to metastasis, the number of lung colonies in WT mice was significantly increased in comparison to that observed in KO mice. The cytoplasm expression of galectin-3 was observed in subcutaneous and metastatic tumors,w in both groups. We observed its nuclear expression in some of subcutaneous tumors of KO mice. The expression of β-catenin was detected in cell membrane of all subcutaneous tumors analyzed, whereas in the metastatic tumors we observed both cytoplasm and cell membrane staining. Altogether, our data suggest that galectin-3 favors the metastasis of melanoma cells and this process is not associated with β-catenin.

Abstract: African-brazilian women have been excluded from society and research projects. This study points out the social inequality survey in Rio de Janeiro and its impact on the health status of African-brazilian woman. A survey was conducted with a population of 1.172 people and the 297 (25.3%) women compound the sample. The results revealed that 46.5% are unsatisfied with her own appearance; the majority is married, has less than a high school degree, has an income inferior than R$ 151 (US$ 75,5). We concluded that the lower socioeconomic status has its roots on racial discrimination which is a chronic and persistent stressor to the health of African-brazilian women. We suggest the further research on the responses of African-women to the health problems could help clarify the racism as a etiologic factor of many conditions.

Abstract: stomach contents of simulium perflavum roubaud larvae were analyzed and compared with plankton and periphyton collected in five streams, in central amazonia (manaus and presidente figueiredo counties), in sep./oct.1996 (dry season) and feb./mar. 1997 (rainy season). a total of 1,400 last-instar larvae were dissected; the stomach contents were analyzed using different methods: fresh and after oxidation. a total of 87 taxa (algae, diatoms and rotifers) were found in the stomach contents. in each stream, qualitative samples of plankton and periphyton were collected; these were mounted between slides and cover slips. a total of 94 taxa of plankton and 54 taxa of periphyton were collected. one species of rotifera was present in the stomach contents, plankton and periphyton. cluster analysis based on species composition of the organisms present in the stomach contents grouped the streams into two major groups, each belonging to a different drainage area. correlations based on presence/absence of species of microalgae in the stomach contents, plankton and periphyton indicated significant associations (p<0.05) between stomach contents and plankton and between plankton and periphyton (z test); the sorensen coefficient and cluster analysis corroborate the same associations.

Abstract: the population biology of three populations of panstrongylus megistus was compared to determine possible influence on the behaviour and epidemiological importance of this species. the results demonstrated differences in terms of egg eclosion time, nymphal mortality and development rates, and feeding and defaecation rates. these differences appeared to follow a geographical cline, primarily reflecting different degrees of adaptation to domestic habitats.

Abstract: the emergency of the socio-environmental crisis points the environmental education as a way to cope this problem. the school seems by the society as a locus to the environmental education implementation is scenery of educators that movie, even in the minority, against-hegemonic initiatives to insert the environmental education in the school schedule. this crisis manifests the civilizatory crisis from a model society and its hegemonic rationality. the participation in environmental education nets is a strategy with potential of to break the isolation of these educators in their schools, making them participants of a collective movement; of to provide interconnection with the local reality place in a larger context; and of to experience an education environment for continuing formation. this article presents for these educators the nets as an environment for the praxis compromised with the critical environmental education.

Abstract: geographic information system (gis) and satellite image were tested to analyze the current land use and automatic localization of areas where conflicts between present land use and land capability occurence, in arroio do meio watershed. geoprocessing techniques map algebra, consultation database and image reclassification were used. the watershed object of this study was chosen because it is considered by many authors, the best unit for social and economic planning and development of population of rural areas. in the studied microbasin: 555ha was covered by forests (24% of the total area). cultivated crops occupy 1.314ha, 56% of the microbasin. glassland covers 184ha (8% of the total area). the flooded areas represent 11% of the area of the microbasin, with 265ha. the use of 31ha could not be detected with accuracy due to shadowing. in areas with steepness above 47%, 32ha were detected without forest cover, totalizing 1,4% of the area of the microbasin. with slopes above 30%, 71ha (3%), are used for agriculture. areas with phaeozems and leptosols (ciríaco-charrua mapping units) with slopes above 30% and without forest coverage performe 14ha (0,6%). areas with conflict use reached only 5% of the total area, indicating that in most of the microbasin, the land is being used in agreement with its capability.
