Impaired Skeletal Muscle Repair after Ischemia-Reperfusion Injury in Mice

Abstract: Ischemia/reperfusion (IR) injury can induce skeletal muscle fibre death and subsequent regeneration. By 14 days, absolute and specific maximal forces and fatigue resistance in ischemic/reperfused soleus muscles were still reduced (−89%, −81%, and −75%, resp.) as compared to control muscles (<.05). The decrease of these parameters in ischemic/reperfused muscle was much greater than that of myotoxic injured muscles (−12%, −11%, and −19%; <.05). In addition, at 14 days ischemic/reperfused muscle structure was still abnormal, showing small muscle fibres expressing neonatal myosin heavy chain and large necrotic muscle fibres that were not observed in myotoxin treated muscles. By 56 days, in contrast to myotoxin treated muscles, specific maximal force and muscle weight of the ischemic/reperfused muscles did not fully recover (<.05). This differential recovery between ischemic/reperfused and myotoxin treated muscles was not related to the differences in the initial cell death, loss of satellite cells after injury, expression of growth factors (IGF1, IGF2..), or capillary density in regenerating muscles. In conclusion, our results demonstrate that IR injury in mice induces long term detrimental effects in skeletal muscles and that the recovery following IR injury was delayed for yet unknown reasons as compared to myotoxic injury.
Epo Is Relevant Neither for Microvascular Formation Nor for the New Formation and Maintenance of Mice Skeletal Muscle Fibres in Both Normoxia and Hypoxia

Abstract: Erythropoietin (Epo) and vascular growth factor (VEGF) are known to be involved in the regulation of cellular activity when oxygen transport is reduced as in anaemia or hypoxic conditions. Because it has been suggested that Epo could play a role in skeletal muscle development, regeneration, and angiogenesis, we aimed to assess Epo deficiency in both normoxia and hypoxia by using an Epo-deficient transgenic mouse model (Epo-TAgh). Histoimmunology, ELISA and real time RT-PCR did not show any muscle fiber atrophy or accumulation of active HIF-1 but an improvement of microvessel network and an upregulation of VEGFR2 mRNA in Epo-deficient gastrocnemius compared with Wild-Type one. In hypoxia, both models exhibit an upregulation of VEGF120 and VEGFR2 mRNA but no accumulation of Epo protein. EpoR mRNA is not up-regulated in both Epo-deficient and hypoxic gastrocnemius. These results suggest that muscle deconditioning observed in patients suffering from renal failure is not due to Epo deficiency.
Dynamic expression of synemin isoforms in mouse embryonic stem cells and neural derivatives

Abstract: We have examined the expression profile of synemin isoforms in mouse pluripotent ES cells and during their neural differentiation induced by retinoic acid. Using RT-PCR, Western blotting and immunostaining, we show that synemin M is present at both mRNA and protein levels in undifferentiated ES cells as early as pluripotency factor Oct-3/4 and IF keratin 8. Synemin H was produced only in neural precursors when neural differentiation started, concurrently with synemin M, nestin and glial fibrillary acidic protein. However, both synemin H and M were restricted to the progenitor line during the neural differentiation program. Our in vivo analysis also confirmed the expression of synemins H/M in multipotent neural stem cells in the subventricular zone of the adult brain, a neurogenic germinal niche of the mice. Knocking down synemin in ES cells by shRNA lentiviral particles transduction has no influence on expression of Oct4, Nanog and SOX2, but decreased keratin 8 expression.Our study shows a developmental stage specific regulation of synemin isoforms in ES cells and its neural derivatives. These findings represent the first evidence that synemins could potentially be useful markers for distinguishing multipotent ES cells from undifferentiated neural stem cells and more committed progenitor cells.Synemin belongs to the intermediate filament (IF) protein family [1,2]. In mammals, the synemin gene is one of the rare IF genes encoding three isoforms (180, 150 and 41 kDa) achieved by alternative mRNA splicing, exon skipping and a shift in the open reading frame [3,4]. The two larger isoforms of synemin (H and M) harbor extended C-terminal tails that project from the surface of the filament and provide connecting arms that associate with neighboring proteins [5-7]. Abstract: Dystrophin contributes to force transmission and has a protein-scaffolding role for a variety of signaling complexes in skeletal muscle. In the present study, we tested the hypothesis that the muscle adaptive response following mechanical overloading (ML) would be decreased in MDX dystrophic muscle lacking dystrophin. We found that the gains in muscle maximal force production and fatigue resistance in response to ML were both reduced in MDX mice as compared to healthy mice. MDX muscle also exhibited decreased cellular and molecular muscle remodeling (hypertrophy and promotion of slower/oxidative fiber type) in response to ML, and altered intracellular signalings involved in muscle growth and maintenance (mTOR, myostatin, follistatin, AMPKα1, REDD1, atrogin-1, Bnip3). Moreover, dystrophin rescue via exon skipping restored the adaptive response to ML. Therefore our results demonstrate that the adaptive response in response to ML is impaired in dystrophic MDX muscle, most likely because of the dystrophin crucial role.
Stress Loops Effect in Ductile Failure of Mild Steel  [PDF]

Abstract: A simulation study of effect of stress on mild steel microstructure has revealed stress loops at areas of stress application which are believed to be the root cause of the ductile failure morphology (cup and cone) in ductile alloys under plane strain conditions. The areas of concentration of these stress loops were observed to be that of subsequent instability (or necking) observed in mild steel and other materials of low friction (flow) stress. Shear stress loops with angular bearing of 450 to the tensile axis were observed to instigate both the failure site and shear morphology in these materials. In plane stress conditions, shear was seen to be that of a wave shape running obliquely to area of stress application resulting in oblique necking observed in thin mild steel sheets.

Abstract: The paper is devoted to obtaining the necessary and sufficient conditions of the solvability of weakly perturbed boundary-value problems for the nonlinear operator-differential Riccati equation in the Hilbert space on the interval and whole line with parameter ?. We find the solution of the given boundary value problem which for ε = 0 turns in one of the solutions of generating boundary value problem. Solution of the generating problem is constructed with the using generalized operator in analytical form. Iterative process for finding of solutions of weakly nonlinear equation with quadratic error is constructed.

Abstract: We examine FVT (free, vertex transitive) actions of wallpaper groups on semiregular tilings. By taking quotients by lattices we then obtain various families of FVT maps on a torus, and describe the presentations of groups acting on the torus. Altogether there are 29 families, 5 arising from the orientation preserving wallpaper groups and 2 from each of the remaining wallpaper groups. We prove that all vertex-transitive maps on torus admit an FVT map structure.

One of ways rise and control after a level of safety flights is integration of all facilities and forms in the complex automated systems. Acceptance of decision about the choice of the best forms management of mean safety flights level in such terms is erected to satisfaction a number of indexes, and in spite of their possible contradiction is a problem which it is possible to decide by means the complex automated use application of mathematical models, methodological approaches to implementation initial stages of design, conceptual design, formalization and algoritmizationi of models and bases management mean of safety flights level.
