Short-Term Effect of Pitavastatin Treatment on Glucose and Lipid Metabolism and Oxidative Stress in Fasting and Postprandial State Using a Test Meal in Japanese Men

Abstract: Introduction. The objective of this study was to clarify how pitavastatin affects glucose and lipid metabolism, renal function, and oxidative stress. Methods. Ten Japanese men (average age of 33.9 years) were orally administered 2?mg of pitavastatin for 4 weeks. Postprandial glucose, lipoprotein metabolism, and oxidative stress markers were evaluated at 0 and 4 weeks of pitavastatin treatment (2?mg once daily) with a test meal consisting of total calories: 460?kcal, carbohydrates: 56.5?g (226?kcal), protein: 18?g (72?kcal), lipids: 18?g (162?kcal), and NaCl: 1.6?g. Metabolic parameters were measured at 0, 60, and 120 minutes after test meal ingestion. Results. After administration of pitavastatin, serum total cholesterol, low-density lipoprotein cholesterol, apolipoprotein B, arachidonic acid, insulin, and adjusted urinary excretion of uric acid decreased, whereas creatinine clearance ( ) and uric acid clearance ( ) increased. And postprandial versus fasting urine 8-hydroxydeoxyguanosine remained unchanged, while postprandial versus fasting isoprostane decreased after pitavastatin treatment. Next, we compared postprandial glucose and lipid metabolism after test meal ingestion before and after pitavastatin administration. Incremental areas under the curve significantly decreased for triglycerides ( ) and remnant-like particle cholesterol ( ), while those for apolipoprotein E (apoE), glucose, insulin, and high-sensitivity C-reactive protein remained unchanged. Conclusion. Pitavastatin improves postprandial oxidative stress along with hyperlipidemia. 1. Introduction It has been generally recognized that postprandial hyperglycemia and hyperlipidemia are highly related to the development of atherosclerosis [1–5]. Hyperglycemia is known to damage vascular endothelial cells, increase oxidative stress, promote the expression of adhesion molecules, and inhibit Nitric Oxide (NO) production [6]. Remnant lipoprotein, an important component of postprandial hyperlipidemia, promotes foam cell formation of macrophages and proliferation of smooth muscle cells [7]. A very recent study on a large number of subjects demonstrated that remnant cholesterol was a causal risk factor for ischemic heart disease [8]. Lipid-lowering drugs, such as statins, fibrates, and ezetimibe are considered to be useful for the treatment of postprandial hyperlipidemia [9–15]. Angiotensin II type 2 receptor signaling significantly attenuates growth of murine pancreatic carcinoma grafts in syngeneic mice

Abstract: The role of AT2 receptor-signaling in stromal cells on the growth of murine pancreatic carcinoma cells (PAN02) was studied using various in vitro and in vivo assays. In vivo cell proliferation, apoptosis, and vasculature in tumors were monitored by Ki-67 immunostaining, TUNEL assay, and von Willebrand factor immunostaining, respectively. In the co-culture study, cell proliferation was measured by MTT cell viability assay. All the data were analyzed using t-test and data were treated as significant when p < 0.05.Our results show that the growth of subcutaneously transplanted syngeneic xenografts of PAN02 cells, mouse pancreatic ductal carcinoma cells derived from the C57/BL6 strain, was significantly faster in AT2-KO mice compared to control wild type mice. Immunohistochemical analysis of tumor tissue revealed significantly more Ki-67 positive cells in xenografts grown in AT2-KO mice than in wild type mice. The index of apoptosis is slightly higher in wild type mice than in AT2-KO mice as evaluated by TUNEL assay. Tumor vasculature number was significantly higher in AT2-KO mice than in wild type mice. In vitro co-culture studies revealed that the growth of PAN02 cells was significantly decreased when grown with AT2 receptor gene transfected wild type and AT2-KO mouse-derived fibroblasts. Faster tumor growth in AT2-KO mice may be associated with higher VEGF production in stromal cells.These results suggest that Ang II regulates the growth of pancreatic carcinoma cells through modulating functions of host stromal cells; Moreover, Ang II AT2 receptor signaling is a negative regulator in the growth of pancreatic carcinoma cells. Prostate Carcinogenesis with Diabetes and Androgen-Deprivation-Therapy-Related Diabetes: An Update

Abstract: Prostate cancer and the androgen deprivation therapy (ADT) thereof are involved in diabetes in terms of diabetes-associated carcinogenesis and ADT-related metabolic disorder, respectively. The aim of this study is to systematically review relevant literature. About 218,000 men are estimated to be newly diagnosed with prostate cancer every year in the United States. Approximately 10% of them are still found with metastasis, and in addition to them, about 30% of patients with nonmetastatic prostate cancer recently experience ADT. Population-based studies have shown that dissimilar to other malignancies, type 2 diabetes is associated with a lower incidence of prostate cancer, whereas recent large cohort studies have reported the association of diabetes with advanced high-grade prostate cancer. Although the reason for the lower prevalence of prostate cancer among diabetic men remains unknown, the lower serum testosterone and PSA levels in them can account for the increased risk of advanced disease at diagnosis. Meanwhile, insulin resistance already appears in 25–60% of the patients 3 months after the introduction of ADT, and long-term ADT leads to a higher incidence of diabetes (reported hazard ratio of 1.28–1.44). Although the possible relevance of cytokines such as Il-6 and TNF-α to ADT-related diabetes has been suggested, its mechanism is poorly understood.
A New Look at the Most Successful Prodrugs for Active Vitamin D (D Hormone): Alfacalcidol and Doxercalciferol

Abstract: Alfacalcidol (1α-hydroxyvitamin D3) has been widely used since 1981 as a prodrug for calcitriol (1α,25-dihydroxyvitamin D3) in the treatment of hypocalcemia, chronic renal failure, hypoparathyroidism and osteoporosis. More recently, doxercalciferol (1α-hydroxyvitamin D2) has been used since 1999 as a prodrug for 1α,25-dihydroxyvitamin D2 for the treatment of secondary hyperparathyroidism. Currently, six forms of vitamin D are known. They range from vitamin D2 to vitamin D7 and are distinguished by their differing side chains. Only vitamin D2 and vitamin D3 have been found to be biologically active based on the elucidation of activation pathways. Alfacalcidol and osteoporosis/doxercalciferol and secondary hyperparathyroidism are discussed, with a new look at old compounds including their practical syntheses.

Abstract: considerations are made on the role of htl.v-i in the etiopathogeny of ham/ tsp. neuroepidemiologic data reported in the literature are revisited for this purpose. among results of this evaluation it is pointed-out that the gkinawan community of brazil presents ethnographic and demographic characteristics which are ideal for designining new studies. for instance, analyses on htlv-i and on ham/tsp in face of the cohort of such community classified according to time and direction of the migration (japan-brazil and vice-versa), will ensure promising results for the understanding of etiopathogeny of ham/tsp. they can also be paths towards clarifying the simultaneous generating, of geographical foci of the disease distant one the other, as that of tumaco and that of south japan.

Introduction Prostate cancer and the hormonal therapy thereof (androgen deprivation therapy, ADT) have been associated with diabetes in terms of diabetes-associated carcinogenesis [1] and ADT-related metabolic disorder [2], respectively. The present paper systematically introduces prostate carcinogenesis with diabetes and ADT-related diabetes/insulin resistance both in epidemiological and etiological approaches. 2. Search Method PubMed and MEDLINE searches were performed for articles published between January 1991 and November 2011 based on the following key words for diabetes-associated prostate carcinogenesis: prostate cancer AND insulin resistance, hyperglycemia, cancer risk, and diabetes. Literature on ADT-related diabetes was searched using the following keywords: androgen deprivation therapy OR hormone therapy AND diabetes, insulin resistance, hyperglycemia, and metabolic syndrome. Relevant articles on growth hormone (GH)/insulin-like growth factor (IGF)-1 and androgen metabolism were searched with similar strategy. Except for studies concerning statistics, meta-analysis, or reanalysis, review articles were excluded. All full papers based on evidence level 1 and 2 and full papers on level 3

Abstract: Recent numerical results on the fractal structure of two-dimensional quantum gravity coupled to $c=-2$ matter are reviewed. Analytic derivation of the fractal dimensions based on the Liouville theory and diffusion equation is also discussed. Excellent agreements between the numerical and theoretical results are obtained. Non-String Pursuit towards Unified Model on the Lattice

Abstract: Non-standard overview on the possible formulation towards a unified model on the lattice is presented. It is based on the generalized gauge theory which is formulated by differential forms and thus expected to fit in a simplicial manifold. We first review suggestive known results towards this direction. As a small step of concrete realization of the program, we propose a lattice Chern-Simons gravity theory which leads to the Chern-Simons gravity in the continuum limit via Ponzano-Regge model. We then summarize the quantization procedure of the generalized gauge theory and apply the formulation to the generalized topological Yang-Mills action with instanton gauge fixing. We find N=2 super Yang-Mills theory with Dirac-K{\"a}hler fermions which are generated from ghosts via twisting mechanism. The Weinberg-Salam model is formulated by the generalized Yang-Mills action which includes Connes's non-commutative geometry formulation as a particular case. In the end a possible scenario to realize the program is proposed. The formulations given here are by far incomplete towards the final goal yet include hopeful evidences. This summary of the overview is the extended version of the talk given at Nishinomiya-Yukawa memorial symposium (Nishinomiya-city Japan, Nov. 1998).
