Effects of salvianolic acid B on expressions of TGF-β1 and its receptors in liver of rats with dimethylnitrosamine-induced hepatic fibrosis

Abstract: Objective: To study the therapeutic mechanism of salvianolic acid B (SA-B) in treating hepatic fibrosis. Methods: Twenty-four Wistar rats were randomly divided into 3 groups: normal control group, untreated group and SA-B-treated group. Rats in the untreated and SA-B-treated groups were injected intraperitoneally with 0.5% dimethylnitrosamine (DMN) for 4 weeks, daily for 3 days each week at a dose of 10 μg/kg, to induce hepatic fibrosis. Then, rats in the SA-B-treated group were given SA-B orally for another 4 weeks. Hydroxyproline (Hyp) contents in liver tissue of the rats in 3 groups were determined with HCl hydrolysis, and collagen typeⅠ, transforming growth factor-beta 1 (TGF-β1), transforming growth factor-beta receptor typeⅠ(TβRⅠ) and transforming growth factor-beta receptor typeⅡ(TβRⅡ) were detected by Western blotting. Results: The Hyp content and the expressions of collagen typeⅠ, TGF-β1, TβRⅠ and TβRⅡ in the liver tissue of rats in the untreated group increased significantly as compared with those in the normal control group, while those in SA-B-treated group decreased significantly as compared with those in the untreated group. Conclusion: The therapeutic mechanism of SA-B in treating hepatic fibrosis may be related to inhibiting the expressions of TGF-β1 and its receptors.

Discovery and validation of potential drug targets based on the phylogenetic evolution of GPCRs  [PDF]

Target identification is a critical step following the discovery of small molecules that elicit a biological phenotype. G-protein coupled recaptors (GPCRs) are among the most important drug targets for the pharmaceutical industry. The present work seeks to provide an in silico model of known GPCR protein fishing technologies in order to rapidly fish out potential drug targets on the basis of amino acid sequences and seven transmembrane regions (TMs) of GPCRs. Some scoring matrices were trained on 22 groups of GPCRs in the GPCRDB database. These models were employed to predict the GPCR proteins in two groups of test sets. On average, the mean correct rate of each TM of 38 GPCRs from two test sets (ST23 and ST24) was found 62% and 57.5%, respectively, using training set 18 (SLD18); the mean hit rate of each TM of 38 GPCRs from ST23 and ST24 was found 68.1% and 64.7%, respectively. Based on the scoring matrices of PreMod, the mean correct rate of each TM of GPCRs from ST23 and ST24 was found 62% and 62.04%, respectively; the mean hit rate of each TM of GPCRs from ST23 and ST24 was found 67.7% and 68.0%, respecttively. The means of GPCRs in ST23 based on SLD18 is close to those based on PreMod; whereas the means of GPCRs in ST24 based on SLD18 is less than those based on PreMod. Moreover, the accuracy (“2”) and validity (“2 + 1”) rates of prediction all seven TMs of 38 GPCRs by the scoring matrices of PreMod are more than those by SLD18, SLA14 and SLA3; whereas the hit rates (94.74% and 97.37%) by PreMod are less than those of SLA3 but bigger than those of SLD18 and SLA14, respectively. This is the reason that we choose PreMod to predict some potential drug targets. 22 GPCR proteins in the sense chain of chromosome 19 constructing validation set were predicted and validated by PreMod whose hit rate is up to 90.91%. Further evaluation is under investigation.

Accurate Modeling of Monopole Antennas in Shielded Enclosures with Apertures

Abstract: Monopole antennas are usually used for measurement of the shielding effectiveness of metallic enclosures. This paper presents an accurate numerical modeling for monopole antennas attached in shielded enclosures with apertures. The electric field integral equation (EFIE) is formulated for the induced currents on both the monopoles and enclosure. The precorrected-FFT accelerated method of moments (pFFT-MoM) is used to solve the integral equation and the incomplete LU (ILU) preconditioner is applied to speed up the convergence of the equation. Compared with existing methods, the model presented in this paper considers the mutual coupling between the monopoles and shielded enclosure. Therefore, it is a better approximation to the actual measurement geometry.
Effects of Wenyang Chubi Decoction on connective tissue growth factor and collagen-Ⅰ in a mouse model of scleroderma

Abstract: Objective: To study the effects of Wenyang Chubi Decoction (WYCBD), a compound traditional Chinese herbal medicine, on connective tissue growth factor (CTGF) and collagen-Ⅰ (COL-Ⅰ) in a mouse model of scleroderma.Methods: Scleroderma was induced in BALB/c mice by daily local injection of bleomycin for three weeks and the mice were randomly divided into untreated, WYCBD-treated and normal saline (NS) treated groups, with another group of BALB/c mice as normal control. WYCBD and NS were given orally for one month respectively. Histopathology in the skin and lungs of the mice were examined. The CTGF and COL-Ⅰ expressions in the skin or skin lesions were detected by immunohistochemical Elivision assay.Results: The expression levels of CTGF and COL-Ⅰ in the untreated group were significantly higher than those in the normal control group (P<0.05). Compared with the NS-treated group, the WYCBD-treated group had significant improvement in the skin and lung histopathology and remarkably decreased expression levels of CTGF and COL-Ⅰ (P<0.05).Conclusion: Scleroderma mice showed high expressions of CTGF and COL-Ⅰin the skin. WYCBD had the effects of decreasing the CTGF and COL-Ⅰ expressions and improving the skin fibrosis.
Extremal Isolated Horizon/CFT Correspondence

Abstract: The near-horizon limit of the extremal (weakly) isolated horizon is obtained under the Bondi-like coordinates. For the vacuum case, explicit coordinate transformation relating the near-horizon metric under the Bondi-like coordinates and the standard Poincar\'e-type or global near-horizon metric of the extremal Kerr black hole is found, which shows that the two geometries are the same. Combined with the known thermodynamics of the (weakly) isolated horizon, it is argued that the Kerr/CFT correspondence can be generalized to the case of a large class of non-stationary extremal black holes.
