Computational Analysis of Physicochemical, Pharmacokinetic and Toxicological Properties of Deoxyhypusine Synthase Inhibitors with Antimalarial Activity  [PDF]

Abstract: Malaria is a parasitic disease which has as etiological agents protozoa of the genus Plasmodium prevalent in tropical countries. The appearance of Plasmodium strains resistant to artemisinin has become necessary the development of new drugs using computational tools to combat this epidemic. Diverse transporter proteins can act as antimalarials targets, thereby being the enzyme deoxyhypusine synthase a promising antimalarial target. The present study aimed to investigate 15 most active inhibitors of deoxyhypusine synthase target, deposited in databases Binding DB, in order to trace a pattern of physicochemical, pharmacokinetic and toxicological properties of the inhibitors for this enzyme and propose new inhibitors of deoxyhypusine synthase target. The physicochemical properties were obtained according to the Lipinski parameters to evaluate oral absorption. Based on the certain properties were proposed three new inhibitors (A, B and C). The ADME/Tox properties were calculated for new inhibitors compared with results of the selected compounds. The fifteen inhibitors for oral administration showed satisfactory results, because they have adapted to the Lipinski parameters. In relation to the penetration of the blood-brain barrier the inhibitors analyzed showed penetration values less than 1, and ranged from 0.0411815 to 0.481764, being that the compound 1 showed value of CBrain/CBlood = 0.135467. Compound B showed a higher strength in plasma protein binding in relation to the compound 1, having a variation be-tween them of ±1.489344. Therefore, the compound B would present a longer halflife compared with compound 1. The proposed compounds showed positive and satisfactory results, being able to reach less adverse effects related to the central nervous system depending of administered dose.

Abstract: introduction: end-stage chronic kidney disease (ckd) requiring dialysis affects the quality of life sometimes more severely than other chronic diseases, such as rheumatoid arthritis, heart failure, coronary artery disease, and chronic obstructive pulmonary disease, exerting a negative effect on the energy and vitality levels, limiting social interactions, and hindering psychic health. objective: to evaluate the quality of life of patients with ckd on hemodialysis in a public brazilian amazonian hospital. methods: data were collected through interview based on the brazilian version of the sf-36 questionnaire. the study was conducted on 50 patients (mean age, 48 ± 16 years; mean hemodialysis time, 3 ± 2.9 years). results: the most affected domain was role limitations due to physical health, with a mean score of 36 ± 36, and 58% of the patients in the lowest quartile, while mental health and social functioning were relatively preserved, with most patients in the highest quartile. men obtained poorer scores than women did for role limitations due to physical health and vitality. age correlated negatively with physical functioning. patients on hemodialysis for more than one year had better scores in the social functioning domain, with a positive correlation between dialysis time and physical functioning. conclusions: the domains assessed were globally impaired in the population studied, especially regarding role limitations due to physical health, suggesting that chronic disease with prolonged treatment has a negative influence on those domains.

Abstract: aujeszky' s disease (ad) is an infectious disease causing important economic losses to the swine industry worldwide. the disease is caused by an alpha-herpesvirus, aujeszky' s disease virus (adv), an enveloped virus with a double stranded linear dna genome. the adv genome encodes 11 glycoproteins, which are major targets for the immune system of the host in response to the infection. the glycoprotein e (ge) is a non-essential protein and deletion of the ge gene has been used for the production of marker vaccines. aiming to develop molecular reagents for the production of a ge specific elisa test, the ge gene was amplified by pcr, cloned and expressed into a baculovirus expression system. the recombinant dna vector pfastbac-ge-adv was used for site-specific transposition into the recombinant baculovirus (bacmid). colonies with recombinant bacmid-pfastbac-ge-adv were selected by antibiotic and color selection and the presence of the ge gene was confirmed by pcr. the recombinant bacmid-pfastbac-ge-adv was cotransfected in insect trichoplusia ni and the presence of the recombinant dna and ge protein were detected by pcr, sds-page and western blotting, respectively.
Allelopathic interaction of Jatropha curcas L. with Helianthus annuus L. and Zea mays L.

Abstract: The goals of this work were to evaluate the effect of the allelopathic interaction of Jatropha curcas with Helianthus annuus and Zea mays from root exudates, as well as to evaluate how these exudates influenced the seeds of these species. Four replicates were made with seeds that were germinated in germiteste paper, which was previously autoclaved. Dormancy of the H. annuus seeds was broken by pre-cooling them in a freezer for seven days at 5°C. Jatropha curcas seeds were cultivated for eight days in a germination chamber under a controlled temperature and photoperiod. These seedlings were then discarded and the same substrate was used to germinate 50 H. annuus seeds; only distilled water was used as the control. The root exudates of H. annuus tested on J. curcas and the allelopathic interaction between J. curcas and Z. mays were carried out using the same methodology. Statistical analysis indicated that J. curcas has an inhibitory effect on the time and average speed of germination of H. annuus seedlings and Z. mays has an inhibitory effect on the germination of the seeds of J. curcas.

Abstract: objective: to evaluate the nutritional status of children from day care centers of the public educational system in the municipality of bezerros, pernambuco, brazil. methods: this cross-sectional study examined the anthropometric indicators of 735 children aged between one and seven years old, which accounted for 70% of the children enrolled at the educational system of bezerros municipality in 2008. the indicators used for classification of the nutritional status were height/age (h/a), weight/age (w/a) and weight/height (w/h), all expressed as z-score values. the software used for data analysis was the epi-info, version 6.04. results: in the studied group, 53.7% of the children were males and 72.7% were from day care centers located in the urban area. the nutritional assessment revealed a prevalence of nutritional deficit of 6.8 and 0.7% according to h/a and w/h, respectively. the prevalence of obesity according to the w/h was 6.1%. a nutritional risk for short stature was observed among males, and a higher prevalence of short stature was verified among female children. there were no differences between urban and rural day care centers in relation to children nutritional status. obesity was associated to h/a deficits in 10% of the children. the ratio between the prevalence of overweight and deficit of w/a was 1.7:1, being 1.3:1 for children from the urban areas and 5:1 for those from rural areas. conclusion: short stature and obesity were the most prevalent nutritional disorders in the studied population. the nutritional transition process was more evident in children of rural area.
Interaction between feeding rate and area for periphyton in culture of Nile tilapia juveniles

Abstract: the objective of the present work was to determine the effects of the interaction between feeding rate and area for periphyton in rearing aquaria for nile tilapia. twenty 25 l polyethylene outdoor aquaria were used to hold experimental fish. three tilapia fingerlings (2.56 ± 0.14 g) were stocked in each aquarium for five weeks (12 fish m-2). there were two different feeding rates (full and full minus 30%) and two submerged areas for periphyton development (59.4% and 96.5% of the aquarium surface area; n = 5). final body weight, specific growth rate and yield in the 96.5%-area aquaria of fish submitted to 30% less feed were significantly higher than in the 96.5%-area aquaria submitted to full feeding rates. fish growth in the restricted-feed aquaria was favored by their better water quality (higher ph and lower total ammonia nitrogen). there were negative effects on final body weight, sgr, yield and feed conversion ratio of fish in full-feed aquaria when the area for periphyton increased from 59.4 to 96.5%. it was concluded that the combination of restricted feeding rates with large substrate areas for periphyton growth results in better water quality for fish culture than the adoption of each management alone.

Abstract: The aims of this paper are to present two methods combined for teaching and learning processes based on the Categories Theory and Knowledge Maps, and to invite the academic community, in different areas of knowledge, particularly in education, to reproduce the experiment in order to support the results, contributing to its maturity and its continuous improvement. In the end, we present a case study for an undergraduation course and discuss the results presenting a model for a digital ecosystem with web and mobile applications.
Swelling Behavior of Polyacrylamide Hydrogels near Phase Transition  [PDF]

Abstract:
Hydrogels based on acrylamide (AA) and sodium methacrylate (NMA) as ionic monomer were prepared by solution polymerization using N,N’-methylenebis(acrylamide) (MBA) or ethylene glycol dimethacrylate (EGMA) as crosslinkers and Ammonium Persulfate (APS) and N,N,N’,N’-Te-tramethyl-ethylenediamine (TMEDA) as initiators. Swelling behavior was greatly affected by NMA content near phase transition. Increasing ionic monomer concentration compared to total monomer one led to high expansion in water, oscillating around 285 g/g for MBA and 325 g/g for EGMA crosslinker, above 20% of NMA. Dynamic Light Scattering experiments were performed and, for both crosslinkers, the dynamic correlation length (ξ) decreased with increasing NMA content, contributing to diminish hydrogels spatial inhomogeneities.

Abstract: Os currículos da área de saúde deixam uma lacuna na forma o teórica e prática, permitindo que a morte seja interpretada como fracasso. A pesquisa objetivou apreender as representa es sociais da morte e do processo de morrer para os profissionais de saúde. Trata-se de uma pesquisa qualitativa de natureza exploratória. Foram entrevistados cinco profissionais de saúde que prestam assistência a pacientes fora de possibilidades de cura. Utilizou-se o software Alceste para apreender o conteúdo das representa es sociais dos entrevistados. Da análise de conteúdo das entrevistas emergiram dois eixos distintos: o primeiro eixo agrupa classes que dizem respeito à dimens o do profissional frente à morte; o segundo eixo refere-se à dimens o subjetiva frente à morte. Os resultados ressaltam a necessidade de investir na capacita o dos alunos n o somente no desenvolvimento de habilidades técnicas, mas também no desenvolvimento de habilidades interpessoais, elementos fundamentais para o cuidado humano, sobretudo diante a morte. Los currículos del área dejan un vacío en la educación y formación, favoreciendo que la muerte sea interpretada como un fracaso. Lo objetivo es aprehender el significado de la muerte y el proceso de morir. Esta investigación cualitativa tiene carácter exploratorio. Los sujetos fueron cinco profesionales de la salud que prestan atención a los pacientes fuera de posibilidades terapéuticas. Para el análisis de datos, se utilizó el software Alceste, Del análisis del contenido de las entrevistas dos ejes distintos habían emergido: el primer eje agrupa las clases que dicen respecto a la dimensión del profesional frente a la muerte; al secundo eje dice respecto a la dimensión subjetiva ante la muerte. Los resultados subrayan la necesidad de investir en la formación de los estudiantes a desarrollar no sólo conocimientos técnicos sino, también, en el desarrollo de habilidades interpersonales, fundamentales para el cuidado humano, especialmente teniendo en cuenta la muerte. Abstract: in situ hybridization (ish) has been used to identify mrna (or dna) in fresh tissue samples of humans and animals. several protocols describing this technique are available, although its description is not usually detailed enough. the present work describes in situ hybridization reaction on fresh tissue in a way to make understandable this important technique, which allows verifying the cellular localization, and the spatial and temporal expression, of gene transcripts (mrna). results with pitx1, tgif, shh and wnt-5a riboprobes, in fresh tissue samples, are presented.
