Development and application of a specific SCAR marker for detection of a released Metarhizium acridum strain M189 in the field

Abstract: To monitor the effect of a Metarhizium acridum strain spread in the field, a DNA molecular marker to differentiate a released strain from other strains or native isolates was developed. Sixteen arbitrary primers to amplify the genomic DNA of 51 Metarhizium strains were used and 81 polymorphic sites were obtained. Among them, 30 polymorphic bands were obtained from the strain M189. One band ascertained to be specificity to the strain M189 was sequenced and transformed as a specific SCAR (sequence characterized amplified regions) marker for this strain. The marker sensitivity was tested and confirmed by identifying strain M189 from 51examined strains. The SCAR marker was used to distinguish 3 isolates from the soil samples collected from the field and confirmed one of them being the released strain M189.

Abstract: The liquid fermentation kinetics of Metarhizium anisopliae IMI330189 was observed. Kinetic models of biomass growth and substrate consumption were established based on Sigmoid model and the parameters of the models were determined by Origin7.5 software. The results showed that the models fitted well with the liquid fermentation process of M. anisopliae IMI330189. The maximum specific growth rate was found to be 0.084h-1 at 22.8h, specific total sugar consumption rate 0.246h-1 at 9.6h and specific total nitrogen consumption rate 0.007h-1 at 10.3h. The maximum yield coefficient of biomass from total sugar was 0.861g/g at 39.8h. The model simulation matched well with the experimental observations, which made it possible to elucidate the kinetic characteristics of M. anisopliae IMI330189 during liquid fermentation. These models thus can be employed for the development and optimization of M. anisopliae IMI330189 liquid fermentation processes.

Abstract: This paper provides an overview of recently developed two dimensional (2D) fragment-based QSAR methods as well as other multi-dimensional approaches. In particular, we present recent fragment-based QSAR methods such as fragment-similarity-based QSAR (FS-QSAR), fragment-based QSAR (FB-QSAR), Hologram QSAR (HQSAR), and top priority fragment QSAR in addition to 3D- and nD-QSAR methods such as comparative molecular field analysis (CoMFA), comparative molecular similarity analysis (CoMSIA), Topomer CoMFA, self-organizing molecular field analysis (SOMFA), comparative molecular moment analysis (COMMA), autocorrelation of molecular surfaces properties (AMSP), weighted holistic invariant molecular (WHIM) descriptor-based QSAR (WHIM), grid-independent descriptors (GRIND)-based QSAR, 4D-QSAR, 5D-QSAR and 6D-QSAR methods.
Effect of aerospace mutation on entomopathogenic fungi

Abstract: Five strains of Metarhizium anisopliae and three strains of Beauveria bassiana were boarded on a spacecraft aviating at the heigh of 200~350km for 18d. Then the strains were inoculated on PDA medium supplement with 1% pepton. There were more than 20% survival spores for strain M189, but only 10-7 survival for strain M2029 and MR8. The spores of other five strains were died completely after the aerospace boarding. Characteristics of isolates from the colonies of survival strains were tested. The results showed that extensive varieties had happened to the isolates on modality of colonies, growth rate, sporular quantity and virulence to insects. The varieties could be positive or negative in contrast with corresponding wild strains. Four isolates from strain M189 were obviously superior to their wild strain on growth rate, sporulation and virulence. Aerospace mutation is an effective approach in breeding of entomopathogenic fungi.
Optimization of the key factors for processing the granule formulation of Metarhizium anisopliae

Abstract: The factors of carriers,accelerant additives and water addition were the key factors in granule formulation processing of Metarhizium anisopliae.After study the influence of these factors on conidial viability,we found that ten examined carriers had no significantly negative effects on conidial germination.Of these,on the contrary,the calcium bentonite,kaolin,pulvistalci,floridin and light calcium carbonate could improve the shelf-life of fungal conidia.Two types of accelerant agents,i.e.,trehalose and tyrosine,could significantly improve the conidia germination with varying rates at 15.12%-24.88% and 15.06%-29.67%,respectively.The effects of water addition on granulation ratio and granulation temperature were obviously correlated.By regression analyses,the suitable addition of water was optimized for reaching more than 90% granulation ratio and obtaining corresponding granulation temperature.The suitable additional quantity of water and suitable granulation temperature for calcium bentonite were 19.1%-25.0% and 50.2-54.5℃,for kaolin 35.0%40.0% and 55.1-56.2℃,for pulvistalci 27.2%-40.0% and 50.4-51.1℃,for floridin 18.3%-25.0% and 51.3-53.6℃ and for light calcium carbonate 40.8%-45.0% and 49.7-56.1℃,respectively.The shelf-life of formulated conidia could be effectively prolonged during storage.

Research on Aspect Oriented Operating Systems

Abstract: Aspect-Oriented software development (AOSD) is a new type of software design idea and technique. This paper analyzes recent research results in three fields which are: (1) the crosscutting concerns and Aspect concept in operating systems; (2) component reconstruction, system evolution and design, and system security; (3) performance monitoring and fault tolerance. The paper points out that some positive research results of Aspect-Oriented operating systems have been acquired. However, the current research results lack necessary deepness and extent, the operating systems that adopt the Aspect idea during design stage do not exist yet, and there is no whole engineering and standardization for Aspect extraction procedure in the existing operating systems code. The solutions for these problems rely on further Aspect-Oriented research results. Final, the paper describes the respective of Aspect oriented operating systems research and indicates that the research of AOSD may possibly bring a significant impact to the research and development of future operating systems.
Chemogenomics Knowledgebased Polypharmacology Analyses of Drug Abuse Related G-Protein Coupled Receptors and Their Ligands

Abstract: Drug abuse (DA) and addiction is a complex illness, broadly viewed as a neurobiological impairment with genetic and environmental factors that influence its development and manifestation. Abused substances can disrupt the activity of neurons by interacting with many proteins, particularly G-protein coupled receptors (GPCRs). A few medicines that target the central nervous system (CNS) can also modulate DA related proteins, such as GPCRs, which can act in conjunction with the controlled psychoactive substance(s) and increase side effects. To fully explore the molecular interaction networks that underlie DA and to effectively modulate the GPCRs in these networks with small molecules for DA treatment, we built a drug-abuse domain specific chemogenomics knowledgebase (DA-KB) to centralize the reported chemogenomics research information related to DA and CNS disorders in an effort to benefit researchers across a broad range of disciplines. We then focus on the analysis of GPCRs as many of them are closely related with drug abuse. Their distribution in human tissues was also analyzed for the study of side effects caused by abused drugs. We further implement our computational algorithms/tools to explore DA targets, DA mechanisms and pathways involved in polydrug addiction and to explore polypharmacological effects of the GPCR ligands. Finally, the polypharmacology effects of GPCR-targeted medicines for drug abuse treatment were investigated and such effects can be exploited for the development of drugs with polypharmacophore for drug abuse intervention. The chemogenomics database and the analysis tools will help us better understand the mechanism of drugs abuse and facilitate to design new medications for system pharmacotherapy of drug abuse.
