Durability of viral response after off-treatment in HBeAg positive chronic hepatitis B

Abstract: AIM: To evaluate the durability in hepatitis B e antigen (HBeAg) positive chronic hepatitis B patients who discontinued antiviral treatment. METHODS: A total of 48 HBeAg positive chronic hepatitis B patients who were administered nucleoside analogues and maintained virological response for ≥ 6 mo [hepatitis B virus (HBV) DNA < 300 copies/mL and HBeAg seroconversion] before cessation of treatment were enrolled between February 2007 and January 2010. The criteria for the cessation of the antiviral treatment were defined as follows: (1) achievement of virological response; and (2) duration of consolidation therapy (≥ 6 mo). After treatment cessation, the patients were followed up at 3-6 mo intervals. The primary endpoint was serologic and virologic recurrence rates after withdrawal of antiviral treatment. Serologic recurrence was defined as reappearance of HBeAg positivity after HBeAg seroconversion. Virologic recurrence was defined as an increase in HBV-DNA level > 104 copies/mL after HBeAg seroconversion with previously undetectable HBV-DNA level. RESULTS: During the median follow-up period of 18.2 mo (range: 5.1-47.5 mo) after cessation of antiviral treatment, the cumulative serological recurrence rate was 15 % at 12 mo. The median duration between the cessation of antiviral treatment and serologic recurrence was 7.2 mo (range: 1.2-10.9 mo). Of the 48 patients with HBeAg positive chronic hepatitis, 20 (41.6%) showed virological recurrence. The cumulative virologic recurrence rates at 12 mo after discontinuing the antiviral agent were 41%. The median duration between off-treatment and virologic recurrence was 7.6 mo (range: 4.3-27.1 mo). The mean age of the virological recurrence group was older than that of the non-recurrence group (46.7 ± 12.1 years vs 38.8 ± 12.7 years, respectively; P = 0.022). Age (> 40 years) and the duration of consolidation treatment (≥ 15 mo) were significant predictive factors for offtreatment durability in the multivariate analysis [P = 0.049, relative risk (RR) 0.31, 95% CI (0.096-0.998) and P = 0.005, RR 11.29, 95% CI (2.054-65.12), respectively]. Patients with age (≤ 40 years) who received consolidation treatment (≥ 15 mo) significantly showed durability in HBeAg positive chronic hepatitis B patients (P = 0.014). These results suggest that additional treatment for more than 15 mo after HBeAg seroconversion in patients who are ≤ 40 years old may be beneficial in providing a sustained virological response. Abstract: Taking Handan City (the south of Northern China) as an example, and making use of the environmentally friendly materials of Handan Environmental Monitor Station in 2013-2014, the surface observational data of Handan Meteorological Station and the sounding data of Xingtai station, combined with the integrated analysis of numerical prediction and simulation, this paper shows that, in 2014, the air quality improved; the emission-reduction effect was significant; the days of heavy pollution decreased; and the days up to the standard increased; the annual average concentration of pollutants PM2.5 and PM10 in the whole city showed a decreasing tendency compared to the same period of last year. In 2014, there were a total of 9 weather processes of heavy pollution lasting more than 3 days and the duration was significantly shorter than that in 2013, which indicated that effective emission-reduction measures significantly shortened the duration of weather processes of heavy pollution. The comprehensive analysis of meteorological conditions, such as the days of light wind, wind speed, wind frequency, PM2.5 concentration at different wind directions, the thickness and intensity of the inversion layer, and the height of the mixed layer, showed that, compared with the same period in 2013, the general meteorological conditions of air pollutant dispersion were basically flat and slightly disadvantageous in 2014. Handan municipal government increased the intensity of the prevention and control of air pollution in 2014, promulgated and adopted a series of air pollution control policies and emission-reduction control measures, and achieved some results, especially the emission-reduction measures during the APEC meeting which were obviously effective.

Abstract: Stroke is the second leading cause of death. Experimental animal models of cerebral ischemia are widely used for researching mechanisms of ischemic damage and developing new drugs for the prevention and treatment of stroke. The present study aimed to comparatively investigate neuroprotective effects of aspirin (ASA), decursinol (DA) and new synthetic aspirin-decursinol adduct (ASA-DA) against transient focal and global cerebral ischemic damage. We found that treatment with 20 mg/kg, not 10 mg/kg, ASA-DA protected against ischemia-induced neuronal death after transient focal and global ischemic damage, and its neuroprotective effect was much better than that of ASA or DA alone. In addition, 20 mg/kg ASA-DA treatment reduced the ischemia-induced gliosis and maintained antioxidants levels in the corresponding injury regions. In brief, ASA-DA, a new synthetic drug, dramatically protected neurons from ischemic damage, and neuroprotective effects of ASA-DA may be closely related to the attenuation of ischemia-induced gliosis and maintenance of antioxidants.

Abstract: This paper will build up a model about the dynamic relationship between scientific innovation and economic growth in Beijing, with VAR model (Vector Autoregressive Model), and choose the quantity of patent applications, sum of business transactions in the market of technologies and the foreign direct investment as its indicators of science and technology output, GDP as its indicator of economic growth, which finally uses variance decomposition analysis. The research shows that there is a long-term equilibrium relationship between scientific innovation and economic growth, and both have interactivity which means that scientific innovation promotes economic growth and economic growth boosts the scientific innovation.
Human endometrial cell coculture reduces the endocrine disruptor toxicity on mouse embryo development

Abstract: Bisphenol A (BPA) and Aroclor 1254 (polychlorinated biphenyls) were used as endocrine disruptors in this study. Mouse 2-cell embryos were cultured in medium alone or vehicle or co-cultured with human endometrial epithelial layers in increasing ED concentrations.At 72 hours the percentage of normal blastocyst were decreased by ED in a dose-dependent manner while the co-culture system significantly enhanced the rate and reduced the toxicity of endocrine disruptors on the embryonic development in vitro.In conclusion, although EDs have the toxic effect on embryo development, the co-culture with human endometrial cell reduced the preimplantation embryo from it thereby making human reproductive environment protective to preimplantation embryo from the toxicity of endocrine disruptors.
Inferring biological functions and associated transcriptional regulators using gene set expression coherence analysis

Abstract: In this study, we propose an algorithm for discovery of molecular functions and elucidation of transcriptional logics using two kinds of gene information, functional and regulatory motif gene sets. The algorithm, termed gene set expression coherence analysis first selects functional gene sets with significantly high expression coherences. Those candidate gene sets are further processed into a number of functionally related themes or functional clusters according to the expression similarities. Each functional cluster is then, investigated for the enrichment of transcriptional regulatory motifs using modified gene set enrichment analysis and regulatory motif gene sets. The method was tested for two publicly available expression profiles representing murine myogenesis and erythropoiesis. For respective profiles, our algorithm identified myocyte- and erythrocyte-related molecular functions, along with the putative transcriptional regulators for the corresponding molecular functions.As an integrative and comprehensive method for the analysis of large-scaled gene expression profiles, our method is able to generate a set of testable hypotheses: the transcriptional regulator X regulates function Y under cellular condition Z. Moxibustion for hypertension: a systematic review

Abstract: We searched 15 databases without language restrictions from their respective dates of inception until March 2010. We included randomized controlled trials (RCTs) comparing moxibustion to either antihypertensive drugs or no treatment. The risk of bias was assessed for each RCT.During the course of our search, we identified 519 relevant articles. A total of 4 RCTs met all the inclusion criteria, two of which failed to report favorable effects of moxibustion on blood pressure (BP) compared to the control (antihypertensive drug treatment alone). However, a third RCT showed significant effects of moxibustion as an adjunct treatment to antihypertensive drug therapy for lowering BP compared to antihypertensive drug therapy alone. The fourth RCT included in this review addressed the immediate BP-lowering effects of moxibustion compared to no treatment. None of the included RCTs reported the sequence generation, allocation concealment and evaluator blinding.There is insufficient evidence to suggest that moxibustion is an effective treatment for hypertension. Rigorously designed trials are warranted to answer the many remaining questions.By 2025, the number of adults with hypertension is predicted to be 1.56 billion worldwide [1]. Rate-Based Active Queue Management for TCP Flows over Wired and Wireless Networks

Abstract: Current active queue management (AQM) and TCP protocol are designed and tuned to work well on wired networks where packet loss is mainly due to network congestion. In wireless networks, however, communication links suffer from significant transmission bit errors and handoff failures. As a result, the performance of TCP flows is significantly degraded. To mitigate this problem, we analyze existing AQM schemes and propose a rate-based exponential AQM (REAQM) scheme. The proposed REAQM scheme uses the input rate as a primary metric and queue length as the secondary metric. The objectives of REAQM are to stabilize networks with low packet loss, low packet delay, and high link utilization regardless the dynamic of network conditions. We prove the global asymptotic stability of the equilibrium based on Lyapunov theory. Simulation results suggest that REAQM is capable of performing well for TCP flows over both wired and wireless networks, and has comparable implementation complexity as other AQM schemes.
