Amlodipine Ameliorates Up-Regulation of ET-1 in Left Ventricle of Hypercholesterolemia Rabbits

Abstract: Objective(s)In addition to antihypertensive effects, amlodipine may exhibit cardiovascular protective effects in heart tissue. The aim of this study was to evaluate the effects of amlodipine and/or high cholesterol diet on blood, heart tissue concentration and mRNA expression of endothelin-1 (ET-1) in male New Zealand white rabbits.Materials and MethodsA total of 40 male New Zealand rabbits were divided into four groups: the normal control group, normal group receiving amlodipine, high-cholesterol diet group and high-cholesterol diet with amlodipine group. After 8 weeks, all the animals anesthetized and blood or tissues samples were collected.ResultsAfter 8 weeks of a high cholesterol diet, the group with such a diet had a significantly higher ratio of left ventricle (LV) weight to body weight than the control group (P= 0.0001). After treatment with amlodipine for 8 weeks, ET-1 level was reduced considerably in comparison with the control (P= 0.01) and high-cholesterol diet groupes (P= 0.01). Amlodipine consumption caused significant reduction (P= 0.01) in the level of ET-1 in heart tissues of high-cholesterol diet group but it had no remarkable effect on the reduction of heart tissue ET-1 in amlodipine group compared with the control group. ConclusionThe present study demonstrates that ventricular prepro-ET-1 mRNA quantitatively increases in the high-cholesterol diet rabbits which results in development of ventricular hypertrophy. It seems that the treatment with amlodipine retards the progression of LV hypertrophy through attenuation of ET-1 levels independent of lipid changes.
The Pattern of Brain-Derived Neurotrophic Factor Gene Expression in the Hippocampus of Diabetic Rats

Abstract: Objective(s)The aim of this study was to evaluate the effects of regular exercise in preventing diabetes complication in the hippocampus of streptozotocin (STZ)-induced diabetic rat.Materials and MethodsA total of 48 male wistar rats were divided into four groups (control, control exercise, diabetic and diabetic exercise). Diabetes was induced by injection of single dose of STZ. Exercise was performed for one hr every day, over a period of 8 weeks. The antioxidant enzymes (SOD, GPX, CAT and GR) and oxidant indexes with brain-derived neurotrophic factor (BDNF) protein and its mRNA and apoptosis were measured in hippocampus of rats. ResultsA significant decrease in antioxidant enzymes activities and increased malondialdehyde (MDA) level were observed in diabetic rats (P= 0.004). In response to exercise, antioxidant enzymes activities increased (P= 0.004). In contrast, MDA level decreased in diabetic rats (P= 0.004). Induction of diabetes caused an increase of BDNF protein and its mRNA expression. In response to exercise, BDNF protein and its mRNA expression reduced in hippocampus of diabetic rats. ConclusionDiabetes induced oxidative stress and increased BDNF gene expression. Exercise ameliorated oxidative stress and decreased BDNF gene expression.
INFLUENCE OF CHRONIC EXERCISE ON RED CELL ANTIOXIDANT DEFENSE, PLASMA MALONDIALDEHYDE AND TOTAL ANTIOXIDANT CAPACITY IN HYPERCHOLESTEROLEMIC RABBITS

Abstract: Despite the knowledge on the antiatherogenic effects of exercise, the mechanism by which exercise reduces atherogenic risk remains unknown. In this study, we investigated the hypothesis that chronic exercise-induced oxidative stress may increase plasma total antioxidant capacity and antioxidant defense in the red cells. For 8 weeks, 60 male Dutch rabbits were fed rabbit chow with or without the addition of 2% cholesterol. The animals were further divided into rest and exercise groups (n = 15 for each group). Animals in exercise groups ran on a rodent treadmill at 15 m/min for 10 to 60 minutes gradually for 5 days per week for a total of 8 weeks. At the end of experiments, blood samples were collected and glutathione peroxidase (GPX), superoxide dismutase (SOD), and catalase (CAT) activities were determined in red blood cells. Total antioxidant capacity (TAC), malondialdehyde (MDA) and total thiol (T-SH) levels were measured in plasma. Thoracic aorta and carotid arteries were isolated for histological examination to evaluate atherosclerosis. Eight weeks of chronic exercise reduced atherogenic diet-induced atherosclerotic lesions in all the arteries studied, along with positive changes in cholesterol profile, especially increase of serum HDL-C level. Plasma MDA, TAC and T-SH concentrations were enhanced by exercise in both control and hypercholesterolemic diet groups. Erythrocyte catalase activity was significantly increased by chronic exercise (p < 0.05), whereas total SOD activity rose with exercise only in the control group. Surprisingly, GPX activity was significantly reduced (P < 0.05) in response to exercise in the control group and also in the high cholesterol diet group. Exercise is a useful tool for the prevention and regression of atherosclerosis which is evident by our findings of the enhancement of plasma TAC and positive change in serum cholesterol profile. However, the effect of exercise on red cell antioxidant activities is limited in the hypercholesterolemic animals compared to control animals, possibly in part because of alterations in the ability to adapt to exercise-induced oxidative stress in high cholesterol diet
Effect of endurance training and cinnamon supplementation on post-exercise oxidative responses in rats

Abstract: Despite the preventative and therapeutic effects of regular exercise, exhaustive exercise may be harmful to health. The present study aimed to determine the protective effect of endurance training and cinnamon bark extract (CBE) supplementation on oxidative responses induced by an exhaustive exercise schedule in rats. The rats were randomly divided into the following five groups of 6; control sedentary (Con/Sed), control exercised (Con/Ex), trained exercised (Tr/Ex), supplemented exercised (Sup/Ex), and trained, supplemented and exercised (Tr/Sup/Ex). Animals in exercise groups ran on a rodent treadmill for an 8-week endurance training program. At the end of the experiment, blood samples were collected and (MDA) and total thiol (TT) levels were measured in plasma. Glutathione peroxidase (GPX), superoxide dismutase (SOD), and catalase (CAT) activities were determined in soleus muscles. Results showed significant increases in SOD activity and malondealdehyde (MDA) levels in the soleus muscles and serum of exercised rats fed with the normal diet. The exhaustive exercise also induced a decrease in serum total thiol level and GPX activity. Elevated levels of total thiol and total antioxidant capacity (TAC) and reduced serum MDA levels were found in the Sup/Ex and Tr/Sup/Ex groups. CAT and GPX activities increased by CBE treatment in trained rats. Regular training increased CAT and GPX activities in the Tr/Sup/Ex group. CAT, GPX and SOD activities were not affected by the CBE treatment in untrained rats. Results suggest that additional use of regular training and CBE supplementation increase TAC and protect healthy male rats against oxidative damage induced by exhaustive exercise.
Minimizing Products Rates Variation in Just-in-Time Mixed-Model Manufacturing  [PDF]

Abstract: Mixed-Model assembly lines are often used in manufacturing based on just-in-time techniques. The effective utilization of these lines requires a schedule for assembling the different models be determined. The objective is to minimize the total deviation of actual production rates from the desired production rates. Mathematical method with the optimization algorithm is proposed here to solve this problem. To prove the efficiency of the proposed algorithm, a number of test problems are solved. The results show that the proposed algorithm is an efficient and effective algorithm which gives better results with the large problem sizes. This paper presents a practical procedure to minimize total product variation rates, and easy to use by practitioner.
Exponential B-Spline Solution of Convection-Diffusion Equations  [PDF]

Abstract: We present an exponential B-spline collocation method for solving convection-diffusion equation with Dirichlet’s type boundary conditions. The method is based on the Crank-Nicolson formulation for time integration and exponential B-spline functions for space integration. Using the Von Neumann method, the proposed method is shown to be unconditionally stable. Numerical experiments have been conducted to demonstrate the accuracy of the current algorithm with relatively minimal computational effort. The results showed that use of the present approach in the simulation is very applicable for the solution of convection-diffusion equation. The current results are also seen to be more accurate than some results given in the literature. The proposed algorithm is seen to be very good alternatives to existing approaches for such physical applications.

Abstract: Applications of Electro-Rheological (ER) or Magneto-Rheological (MR) fluids as typical smart materials have been widely investigated over the past decades (since their introduction in 40s). The special applications of these materials as a means of noise suppression are not yet investigated. Constrained Layer Damping (CLD) sheets can be realized by incorporating EMR (ER/MR) materials. In this way, a multilayered damping sheet is obtained with adaptive (tunable) stiffness and damping characteristics. These properties are easily changed in proportion to the electric (magnetic) field applied upon the EMR layer. This notion has been introduced for semi-active vibration control problems. Experimental Evaluation of Transmission Loss of a Glass Cylinder Tube Containing a Fluid  [PDF]

Abstract: Transmission Loss (TL) of a glass cylinder tube containing a fluid is studied experimentally. This test specimen represents a typical double layer panel including a fluid. The tests are carried out by using a modified four-microphone standing-wave (impedance) tube for specimens with different lengths, 15 and 30 mm. Each cylinder tube is tested filled with one of the fluids at a time. The fluids are air, water, motor oil and a nanoparticle fluid (in absence of magnetic field). The effects of the cylinder length (thickness), impedance tube terminations, and the containing fluid are discussed. The increasing of the thickness led to an increase of the TL values and a decrease in resonance frequencies. Also, the addition of liquid middle layer led to considerable increase of the TL.

Abstract: The present paper is concerned with two novel approximate analytic solutions of the undamped Duffing equation. Instead of the traditional perturbation or asymptotic methods, a homotopy technique is employed, which does not require a small perturbation parameter or a large parameter for an asymptotic expansion. It is shown that proper choices of an auxiliary linear operator and also an initial approximation during the implementation of the homotopy analysis method can yield uniformly valid and accurate solutions. The obtained explicit analytical expressions for the solution predict the displacement, frequency and period of the oscillations much more accurate than the previously known asymptotic or perturbation formulas.
