MicroRNAs play critical roles during plant development and in response to abiotic stresses

Abstract: micrornas (mirnas) have been identified as key molecules in regulatory networks. the fine-tuning role of mirnas in addition to the regulatory role of transcription factors has shown that molecular events during development are tightly regulated. in addition, several mirnas play crucial roles in the response to abiotic stress induced by drought, salinity, low temperatures, and metals such as aluminium. interestingly, several mirnas have overlapping roles with regard to development, stress responses, and nutrient homeostasis. moreover, in response to the same abiotic stresses, different expression patterns for some conserved mirna families among different plant species revealed different metabolic adjustments. the use of deep sequencing technologies for the characterisation of mirna frequency and the identification of new mirnas adds complexity to regulatory networks in plants. in this review, we consider the regulatory role of mirnas in plant development and abiotic stresses, as well as the impact of deep sequencing technologies on the generation of mirna data.

Abstract: the effect of high temperature on sex ratio in the chitralada strain of nile tilapia (oreochromis niloticus) was tested using two experiments. primarily, the animals were exposed to a high (35oc) or control (27oc) water temperature for a 28-day-period, beginning ten days after hatching. a significant difference in sex ratio was found. the highest proportion of males was in the high temperature group (72.39%), compared with the control group (62.27%). no differences were found for total length or body weight between the groups. in the second experiment, the effect of exposure times (7, 14, 21 and 28 days starting ten days post hatching) at a constant temperature of 35oc was tested. no differences were found in sex ratio between groups. a significant difference was found between treatments for body weight and total length. in both experiments, the survival rate had a direct relationship with the occurrence of cannibalism, which was significantly higher at 35oc. the significant temperature effect on sex ratio found in the first experiment indicates the thermosensitivity of the chitralada strain.

Abstract: Os objetivos do presente estudo foram avaliar os efeitos da autonomiza o cirúrgica sobre a extens o da área viável e da vasculariza o de retalhos cutaneos pré-fabricados com área média de 48,2cm2 na parede abdominal de ratos. Os retalhos de pele eram pré-fabricados por implante de um pedículo femoral ligado distalmente e implantado diretamente na camada subdérmica. A pele da parede abdominal de 25 ratos Wistar foi comparada em três grupos de retalhos: grupo A, implante vascular sem autonomiza o e grupos B e C, implante vascular com autonomiza o no mesmo tempo cirúrgico. Três semanas após, os retalhos dos grupos A e B foram elevados como retalhos em ilha, nutridos pelo pedículo implantado. No grupo C, o pedículo implantado foi seccionado no momento da eleva o do retalho. Sete dias após, a extens o das áreas viáveis nos grupos A, B e C foi delimitada e o percentual da área viável, em rela o à área total do retalho, calculado por meio do Auto Cad R 14. A densidade vascular em torno do pedículo implantado, nos grupos A e B, foi avaliada por estudo histológico. O valor médio do percentual de área viável dos retalhos de pele foi de 9,6% no grupo onde os retalhos n o foram autonomizados, 44,8% no grupo onde os retalhos foram autonomizados e 0,3% no grupo onde o pedículo implantado foi seccionado. Os resultados mostraram que o procedimento de autonomiza o aumentou significativamente (p< 0,01, teste bicaudal de Mann - Whitney) os percentuais de área viável e n o alterou estatisticamente (p = 0,307, teste bicaudal de Mann - Whitney) a densidade vascular em torno do pedículo, três semanas após o mesmo ter sido implantado.
Retalho ósseo pré-fabricado com osso homógeno: estudo da matura o óssea em um modelo experimental

Abstract: As op es convencionais de tratamento de perdas ósseas, como enxertos e retalhos n o s o satisfatórias. Com isso uma nova categoria de retalhos, o retalho pré-fabricado (RPF), passou a ser estudada. Foram retirados 42 ossos metatársicos-I das patas traseiras de 21 ratos Wistar. O grupo I (n=21) foi descalcificado e liofilizado e o grupo II (n=21) foi n o descalcificado e congelado a -70°C. Após 3 semanas os ossos foram enxertados em 21 animais sobre os vasos epigástricos inferiores nas regi es inguinal direita e esquerda, individualizados por uma lamina de silicone. Os grupos I e II foram divididos em três subgrupos cada, de acordo com o tempo de permanência (1, 2 e 4 semanas) após a pré-fabrica o do retalho. No estudo histológico macroscópico os grupos apresentaram diferen as em morfologia e consistência: o grupo I mostrou perda da arquitetura óssea e da rigidez e grupo II mantiveram sua forma, rigidez e consistência. Na análise qualitativa altera es foram observadas nos fragmentos do grupo desmineralizado e liofilizado, especialmente no subgrupo de 2 semanas. O grupo II n o apresentou altera o no aspecto do tecido ósseo mineralizado entre os diversos subgrupos. é nítida a diferen a histológica entre os RPF com osso descalcificado e liofilizado quando comparado ao n o descalcificado e congelado. A reabsor o óssea gradual sugere que a rota o do retalho deve ocorrer precocemente. Este estudo demonstra a aplicabilidade do osso homógeno para pré-fabrica o de retalhos.

Abstract: in the course of the coffee breeding project carried out at the instituto agron?mico, outstanding progenies have been derived from mother plants selected by either of the two following methods; a) before selection, individual yield is recorded for a number of years begining with the first crop, and b) selection of mother plants is made on the basis of vegetative vigor and the crop they have at the time of selection, regardless of their previous yields. yield data and other observations presented in this paper refer to a group of progenies derived from plants selected according to the second method. with open pollinated seeds collected the from best plants found in 1943 in a private coffee plantation at urupês county, (formerly mundo novo) state of s?o paulo several progenies were planted at the experiment stations of campinas, pindorama, jaú and mococa, with different types of soils. the original coffee plantation of urupês had been formed with seeds collected in mineiros do tietê county, in a farm nearby the place where the so called sumatra coffee (coffea arabica var. typica) was first planted after it was privately imported from the island of sumatra at the end of the last century. this is probably the reason why the mundo novo coffee, as it is called nowadays, was formerly named sumatra de mundo novo. from observations on the morphological type of mundo novo coffee plants, it was noted that most plants in all progenies and also in private plantations are morphologically similar to the bourbon variety, while a few plants are similar to the typica variety. artificial crosses between the highest yielding mundo novo plants from progenies grown in campinas and the murta variety (ttnana) grave indication that these plants are of the bourbon type (ttnana) and not of the typica type (ttnana), as it is the case of the ordinary sumatra coffee. it has been assumed, therefore, that the mundo novo coffee probably originated from a primitive natural cross between the imported
Quality of life in patients with age-related macular degeneration with monocular and binocular legal blindness

Abstract: objective: to evaluate the quality of life for persons affected by age-related macular degeneration that results in monocular or binocular legal blindness. methods: an analytic transversal study using the national eye institute visual functioning questionnaire (nei vfq-25) was performed. inclusion criteria were persons of both genders, aged more than 50 years old, absence of cataracts, diagnosis of age-related monocular degeneration in at least one eye and the absence of other macular diseases. the control group was paired by sex, age and no ocular disease. results: group 1 (monocular legal blindness) was composed of 54 patients (72.22% females and 27.78% males, aged 51 to 87 years old, medium age 74.61 ± 7.27 years); group 2 (binocular legal blindness ) was composed of 54 patients (46.30% females and 53.70% males aged 54 to 87 years old, medium age 75.61 ± 6.34 years). the control group was composed of 40 patients (40% females and 60% males, aged 50 to 81 years old, medium age 65.65 ± 7.56 years). the majority of the scores were statistically significantly higher in group 1 and the control group in relation to group 2 and higher in the control group when compared to group 1. conclusions: it was evident that the quality of life of persons with binocular blindness was more limited in relation to persons with monocular blindness. both groups showed significant impairment in quality of life when compared to normal persons.
Biologia molecular de baculovírus e seu uso no controle biológico de pragas no Brasil

Abstract: baculoviruses are insect viruses found mainly in lepidoptera. the family baculoviridae is taxonomically divided in two genera, nucleopolyhedrovirus and granulovirus, which differ by occlusion body morphology. npvs (nucleopolyhedroviruses) have polyhedrical inclusion bodies (pibs) containing multiple viral particles, while gvs (granuloviruses) appear to be generally single particles occluded in oval shaped occlusion bodies. during the life cycle, two different viral progenies are produced: bv (budded virus) and pdv (polyhedra derived virus), which are essential for the infectious process and virus propagation in host cells. baculoviruses are being used for pest control and they are especially safe due to their specificity and invertebrate-restricted host range. baculoviruses have been used as vectors for high level protein expression ofheterologous genes from prokaryotic and eukaryotic organisms. also, recombinant dna techniques have allowed the production of genetically modified viral insecticides. this study is a review on the taxonomy, structure, replication and molecular biology of baculoviruses, as well as their use as bioinsecticides in brazil.

Abstract: recent advances for improving physicochemical and nutritional properties of lipids are reviewed, with emphasis on products attaining by biochemical processing of natural fats and oils. enzymatic interesterification provides an important route to modify physical and nutritional properties of milkfat without generating trans isomers. this process makes use of lipases, a versatile class of enzyme that is able to perform efficiently the target modification in both solvent and solvent free systems. the present review covers important features of lipases, lipase-catalyzed interesterification reactions and their effects on the composition and texture of the resulting product.

Abstract: Este estudo avaliou o desempenho zootécnico do camar o marinho Litopenaeus vannamei em diferentes tipos de substratos n o consolidados. Juvenis (0,97 ± 0,27 g) foram aclimatados na densidade de 52 m-2, em 21 caixas de polietileno com biofiltros individuais, emum sistema fechado de filtra o contínua, sendo submetidos aos tratamentos: 1) A = 25% silte + 25% argila + 25% areia muito fina + 25% areia fina; 2) B = 50% areia fina + 50% areia muito fina e 3) Controle = sem substrato, em um delineamento experimental completamente casualizado, com três tratamentos e sete repeti es cada. Os animais foram alimentados com 12% da biomassa/dia com ra o peletizada (35% de proteína bruta), ofertada em bandejas e parceladaem duas ofertas diárias (8 e 16h). O estudo teve dura o de 48 dias, sendo avaliados: consumo aparente de ra o, eficiência alimentar, taxa de crescimento específico, sobrevivência e ganho de peso. Foram acompanhados diariamente salinidade, pH, am nia e temperatura da água. A análise estatística dos dados se deu por meio dos testes Anova ou Kuskal-Wallis, em fun o da sua parametricidade. N o foram observadas diferen as significativas com rela o aos fatores avaliadosnas diferentes granulometrias de sedimento testadas, indicando que estas n o exerceram influência sobre o desempenho zootécnico de L. vannamei juvenil. This study aimed to evaluate the zootechnical performance of Litopenaeus vannamei shrimp at different types of unconsolidated substrate. Juvenile shrimp (0.97 ± 0.27 g) were acclimatized, in a stocking density of 52 m-2, to 21 polyethylene boxes (50 L) connected individually to biofilters in a closed water exchange system. The animals were submitted to the treatments: 1) A = 25% silt + 25% clay + 25% very fine sand + 25% fine sand; 2) B = 50% fine sand + 50% very fine sand and 3) Control = no sediment, with seven replications each. Photoperiod was set on 12L: 12D. Shrimps were fed twice a day (at08:00 and 16:00h) with 12% total biomass day-1. Food was offered in feeding trays 2 cm from the substrate. The experiment had a duration of 48 days, evaluating: apparent ration intake, food efficiency, specific growth rate, survival and weight gain. During this period, water quality was monitored daily. Data was analyzed by Anova or Kuskal-Wallis test, depending on normality (SigmaStat 3.1, 2004 and STATISTICA 6.0, 2001). There were no differences among groupsregarding weight gain, food intake, SGR and survival, indicating that these did not exert influence on the zootechnical performance of L. vannamei juvenil.
