Production, Purification, and Characterization of Polygalacturonase from Rhizomucor pusillus Isolated from Decomposting Orange Peels

Abstract: A thermophilic fungal strain producing polygalacturonase was isolated after primary screening of 40 different isolates. The fungus was identified as Rhizomucor pusilis by Microbial Type Culture Collection (MTCC), Chandigarh, India. An extracellular polygalacturonase (PGase) from R. pusilis was purified to homogeneity by two chromatographic steps using Sephadex G-200 and Sephacryl S-100. The purified enzyme was a monomer with a molecular weight of 32?kDa. The PGase was optimally active at 55°C and at pH 5.0. It was stable up to 50°C for 120?min of incubation and pH condition between 4.0 and 5.0. The stability of PGase decreases rapidly above 60°C and above pH 5.0. The apparent and values were 0.22?mg/mL and 4.34?U/mL, respectively. It was the first time that a polygalacturonase enzyme was purified in this species. It would be worthwhile to exploit this strain for polygalacturonase production. Polygalacturonase from this strain can be recommended for the commercial production because of its constitutive and less catabolically repressive nature, thermostability, wide range of pH, and lower properties. However, scale-up studies are needed for the better output for commercial production. 1. Introduction Pectin substances constitute a complex linear backbone comprised of α-1, 4-linked d-galacturonic acid residues which may be methylated and substituted with l-rhamnose, arabinose, galactose, and xylose [1–3]. Because of the large variety of pectins in plant material, they endowed with many pectinolytic enzyme systems which can degrade them [4]. The hydrolysis of pectin backbone is obtained by the synergistic action of several enzymes, including pectin methylesterase (EC., endopolygalacturonase (EC., exopolygalacturonase (EC., pectate lyase (EC., exo-pectate lyase (EC., and endopectin lyase ( [5, 6]. Pectinolytic enzymes are of prime importance for plants as they help in cell-wall extension and softening of some plant tissues during maturation and storage [7, 8]. They also aid in maintaining ecological balance by causing decomposition and recycling of waste plant materials. Plant pathogenicity and spoilage of fruits and vegetables by rotting are some other major manifestations of pectinolytic enzymes [9, 10]. They have been used in many industrial and biotechnological processes, such as textile and plant fiber processing, coffee and tea fermentation, oil extraction, treatment of industrial wastewater containing pertinacious material, purification of plant viruses, and paper making [11]. Physiological and Biochemical Changes During Seed Deterioration in Aged Seeds of Rice (Oryza sativa L.)

Abstract: The present study was carried out in five rice varieties viz. Pusa-1, Sugandha-4, Sugandha-5, Saket-4 and Saket-370, for analyzing the varietals differences during seed deterioration by subjected them to Accelerated Ageing (AA) at 45°C and 100% relative humidity (RH) for 24, 48, 72 and 96 h. Various physiological parameters such as moisture content, germination percentage, seedling vigour, vigour index and biochemical parameters such as reducing sugar based on DNS assay, soluble protein by Lowry’s method were explored. Different levels of seed vigor were obtained with different varieties and duration of accelerated aging treatments. Prolongation of ageing led to deterioration of both germinability and seed viability. The physiological results were coinciding with the biochemical parameters. Ageing conditions resulted in increased moisture content in all the varieties. The study concludes that accelerated ageing showed effect on seed quality of all the varieties of rice. All the test experiments performed concluded that Sugandha-4 is the best variety, followed by Saket-370 among all five varieties and Sugandha-5 and Saket-4 are highly sensitive varieties with respect to accelerated ageing.

Abstract: Some new 6-(substituted phenyl)-2-(4-substituted phenyl-5-thioxo-4,5-dihydro-1H-1,2,4-triazol-3-yl)-4,5-dihydropyridazin-3(2H)-one derivatives were synthesized by a sequence of reactions starting from respective aryl hydrocarbons. The final compounds (4a-i) were screened for antihypertensive activities by non-invasive method using Tail Cuff method. The compounds 4e and 4i showed appreciable antihypertensive activity.

We propose a new Quantum Key Distribution method in which Alice sends pairs of qubits to Bob; each is in one of four possible states. Bob uses one qubit to generate a secure key and the other to generate an auxiliary key. For each pair he randomly decides which qubit to use for which key. The auxiliary key has to be added to Bob’s secure key in order to match Alice’s secure key. This scheme provides an additional layer of security over the standard BB84 protocol.

Abstract: To assess the frequencies and factors of domestic injuries among children aged less than 5 years. This is a cross sectional study, in which parents who have child less than 5 years of age were targeted. A total of 246 participants had provided satisfactory information and they were approached by non-probability purposive sampling technique from January 2016 to June 2016. The data was entered in Excel and then imported to SPSS 19.0 for inferential analysis. In this study, 89.8% (n = 221) mothers and 10.2% (n = 25) father who had participated. 52.08% (n = 123) parents were from joint family. The different types of domestic injuries faced by children were fall (52%), sharp cut (36%), and burn (31%) and Temporary fever, rashes and scratches were the most common consequence of domestic injury as experienced by 57% children. No any significant difference (p > 0.05) in the storage practices of different household hazardous item was observed among the parents living in joint and nuclear family system. Protecting children from domestic injury is a multi-dimensional approach. It needs parent’s education, manufacturer guidance, Government regulation and joint community efforts for proper and safe storage of household items. These measures will aid in reducing the burden of childhood domestic injuries.
Synthesis, reactions and biological activity of 3-arylidene-5-(4-methylphenyl)-2(3H)-furanones

Abstract: 3-Arylidene-5-(4-methylphenyl)-2(3H)-furanones 2a–m were prepared from 3-(4-methyl-benzoyl)propanoic acid 1 and several aromatic aldehydes. Some of the selected furanones were reacted with ammonia gas and benzylamine to give corresponding 3-arylidene-1,3-dihydro-5-(4-methylphenyl)-2H-pyrrol-2-ones 3a–h and 3-arylidene-1-benzyl-1,3-dihydro-5-(4-methylphenyl)-2H-pyrrol-2-ones 4a–f, respectively, which were characterized on the basis of IR, 1H-NMR, mass spectral data and elemental analysis results. These compounds were tested for their anti-inflammatory and antibacterial activities. The compounds, which showed significant anti-inflammatory activity, were further screened for their analgesic and ulcerogenic activities. Three new compounds (2e, 2h and 4d), out of twenty-seven showed very good anti-inflammatory activity in the carrageenan induced rat paw edema test, with significant analgesic activity in the acetic acid induced writhing test together with negligible ulcerogenic action. The antibacterial activity is expressed as the corresponding MIC values.
A Nonlocal Wave-Particle Duality

Abstract: We propose and analyze a modified ghost-interference experiment, and show that revealing the particle-nature of a particle passing through a double-slit hides the wave-nature of a spatially separated particle which it is entangled with. We derive a nonlocal duality relation, ${\mathcal D}_1^2 + {\mathcal V}_2^2 \le 1$, which connects the path distinguishability of one particle to the interference visibility of the other. It extends Bohr's principle of complementarity to a nonlocal scenario. We also propose a ghost quantum eraser in which, erasing the which-path information of one particle brings back the interference fringes of the other.
