Chip-Oriented Fluorimeter Design and Detection System Development for DNA Quantification in Nano-Liter Volumes

Abstract: The chip-based polymerase chain reaction (PCR) system has been developed in recent years to achieve DNA quantification. Using a microstructure and miniature chip, the volume consumption for a PCR can be reduced to a nano-liter. With high speed cycling and a low reaction volume, the time consumption of one PCR cycle performed on a chip can be reduced. However, most of the presented prototypes employ commercial fluorimeters which are not optimized for fluorescence detection of such a small quantity sample. This limits the performance of DNA quantification, especially low experiment reproducibility. This study discusses the concept of a chip-oriented fluorimeter design. Using the analytical model, the current study analyzes the sensitivity and dynamic range of the fluorimeter to fit the requirements for detecting fluorescence in nano-liter volumes. Through the optimized processes, a real-time PCR on a chip system with only one nano-liter volume test sample is as sensitive as the commercial real-time PCR machine using the sample with twenty micro-liter volumes. The signal to noise (S/N) ratio of a chip system for DNA quantification with hepatitis B virus (HBV) plasmid samples is 3 dB higher. DNA quantification by the miniature chip shows higher reproducibility compared to the commercial machine with respect to samples of initial concentrations from 103 to 105 copies per reaction.
A Model to Predict Total Chlorine Residue in the Cooling Seawater of a Power Plant Using Iodine Colorimetric Method

Abstract: A model experiment monitoring the fate of total residue oxidant (TRO) in water at a constant temperature and salinity indicated that it decayed exponentially with time, and with TRO decaying faster in seawater than in distilled water. The reduction of TRO by temperature ( °K) was found to fit a curvilinear relationship in distilled water (r2 = 0.997) and a linear relationship in seawater (r2 = 0.996). Based on the decay rate, flow rate, and the length of cooling water flowing through at a given temperature, the TRO level in the cooling water of a power plant could be estimated using the equation developed in this study. This predictive model would provide a benchmark for power plant operators to adjust the addition of chlorine to levels necessary to control bio-fouling of cooling water intake pipelines, but without irritating ambient marine organisms.
Effects of Jianpi Bushen Huoxue Recipe on hematopoietic cell apoptosis in a mouse model of aplastic anemia

Abstract: Objective: Adopting methods of cell culture to explore the effects and mechanisms of Jianpi Bushen Huoxue Prescription (JPBSHXP), a traditional Chinese compound herbal medicine for strengthening spleen, reinforcing kidney and activating blood circulation, in inhibiting hematopoietic cells apoptosis in a mouse model of aplastic anemia (AA).Methods: Blood serum of AA mice was made from an AA mouse model. Blood serums containing different traditional Chinese compound herbal medicine were made from rats after intragastric administration of JPBSHXP and its related decoctions, respectively. Bone marrow cells of normal mice were incubated by these blood serums for 24 hours, respectively. The apoptosis of the bone marrow cells were assayed by flow cytometry and transmission electron microscopy (TEM).Results: It was indicated that the bone marrow cells of normal mice incubated with blood serum of AA mice displayed typical apoptosis. The apoptosis rates of bone marrow cells of the AA mice incubated by blood serum containing different traditional Chinese herbal medicine were decreased. The effect of Bushen (reinforcing kidey) Recipe was better than Jianpi (strengthening spleen) Recipe and Huoxue (activating blood circulation) Recipe, while the effect of JPBSHXP was the best. TEM results showed that the effect of Bushen Recipe was better than that of the Jianpi Recipe and the Huoxue Recipe, while the effect of JPBSHXP was the best.Conclusion: JPBSHXP and its related decoctions can significantly decrease the apoptosis rate of bone marrow mononuclear cells of the AA mice. It is inferred that JPBSHXP can promote bone marrow hematogenesis
Effects of Bushen Huoxue Granule on motor function in patients with Parkinson’s disease: a multicenter, randomized, double-blind and placebo-controlled trial

Abstract: Background: The main clinical symptoms of Parkinson's disease (PD) are resting tremor, muscle rigidity, bradykinesia, and so on. There is no effective treatment for PD yet, and dyskinesia symptoms affect the life qualities of PD patients. The therapy used for reinforcing kidney and activating blood circulation in treatment of PD can achieve good clinical effects.Objective: To evaluate the efficacy and safety of Bushen Huoxue Granule (BSHXG), a compound traditional Chinese herbal medicine for reinforcing kidney and activating blood circulation in treatment of PD.Design, setting, participants and interventions: A multi-center, randomized, double-blind, placebo-controlled clinical study was undertaken. A total of 120 PD patients from Outpatient Department of General Hospital of People’s Liberation Army, Guangdong Provincial Hospital of Traditional Chinese Medicine, and Xijing Hospital and Tangdu Hospital in Xi’an, were randomly divided into BSHXG group and placebo group. There were 55 cases in BSHXG group, for 5 cases lost to follow-up, and 51 cases in placebo group, for 1 case was excluded and 8 cases lost to follow-up. The patients in two groups were all treated for three months.Main outcome measures: The movement scale, exercise testing, and muscle tension were observed before and after treatment to make a comprehensive evaluation for clinical efficacy. One month follow-up was also made.Results: At three different times (one, two and three months) after treatment, the score of Unified Parkinson’s Disease Rating Scale (UPDRS) Ⅲ, rise time of 10-meter back and forth exercise and resting muscle tension in BSHXG group were improved as compared with before treatment (P<0.05, P<0.01), and there was an interaction between treatment time and intervention (P<0.05, P<0.01). There were no differences in evaluation results of chronograph movement (times of left and right hand movement in one minute), and walking time and turn around time of 10-meter back and forth exercise between BSHXG group and placebo group, and no interaction existed between treatment time and intervention. BSHXG showed a better efficacy than the placebo (P<0.01) in improving motor function, shortening rise time of 10-meter back and forth test and relieving muscle tension. No adverse effects were found in this trial.Conclusion: BSHXG plus Western medicine is effective and safe in improving motor dysfunction of PD patients.

Spatial variability of soil nutrients in a hill-pond landscape

Abstract: 理解土壤养分空间分布的异质性对于评价和管理土地资源具有重要意义。利用地理信息系统和地统计学方法定量研究了丘塘景观土壤养分的空间异质性特征。结果表明 ,土壤全氮的有效变程最大 (为 16 5 m) ,有机碳次之 (10 2 m) ,而全磷的变程最小(90 m)。土壤有机碳含量由高到低 5个不同级别的土壤面积与丘塘景观整个面积的比值的变化范围较小 (10 .5 4 %~ 2 3.15 % ) ,土壤全氮含量比值变化范围较大 (5 .79%~ 32 .73% ) ,土壤全磷的比值的变化范围最大 (1.80 %~ 4 2 .0 6 % )。土壤有机碳和土壤全氮的分布情况较为一致 ,不同级别斑块分布也很相似 ,土壤有机碳含量高的地方土壤全氮也高。表层土壤有机碳和全磷的空间异质分布用球状模型拟合最佳 ,而全氮的空间分布规律更宜用指数模型来拟合。景观尺度的半方差拟合总体上优于斑块尺度。相对有机碳和全氮 ,全磷的空间异质性更多由随机因素 (如人类施肥活动 )引起和决定。土壤全氮的取样尺度应大于 16 5 m,而有机碳、全磷的取样距离则分别可大于 10 3m和 90 m。
Spatial distribution pattern of tree individuals in the Schrenk spruce forest, northwest China

Abstract: The spatial dispersion pattern of individual trees in forest communities is one of the central topics in community ecology. The Schrenk spruce (Picea schrenkiana) forest is the most widely spreaded forest type in Xingjiang Province, northwest China. It has profound significance in soil and water conservation, timber harvesting, and climate regulation to the region. Understanding tree dispersion pattern is important to conserve and manage the forest sustainablly. Two sample plots in the Xingjiang Agriculture University's Experimental Forest, with the area of 2.52 hm~2 and 3.0 hm~2, were set up for field investigation. Species within each plot were identified, and number of species, tree height, tree age, and DBH (diameter at breast high) were recorded for each individual tree. The spatial distribution pattern of individual trees was further analyzed using the Ripley's K function. The results showed that Schrenk Spruce tree individuals distributed in an aggregative fashion. X~2 test showed that there was no significant deviation from the Poisson expectation to a distance of 10 m, but significant clumping pattern occurred beyond this distance, and it could reach 86 m. Two measures about the population distribution pattern, Ripley's K statistic and quadrat methods were compared and the result showed that the former was better. This suggested that random distribution at the local (nearest neighbor) scales gave way to the clumpy pattern at larger scales. Furthermore, dead Schrenk trees were significantly clumped at all scales. Two other common tree species in the Schrenk spruce forest, Betula tianschanica Rupr. and Salix tianschanica Rgl. were also distributed aggregatively, and the degree of aggregation is even higher than that of spruce trees. The aggregation of Betula and Salix is different in two sample plots because of the succession stage is different. The spatial distribution pattern of Schrenk Spruce was closely related to tree ages. The younger the trees, the more aggregative the distribution pattern tended to be. But the degree of aggregation of Spruce trees with ages larger than 150 years tended to be higher than any other age classes due to the forming of forest gaps. Individual tree distribution pattern could also be influenced by its regeneration characteristics and surrounding microenvironments. Further studies should be devoted to analyze the relationship between the spatial distribution pattern and these influencing factors.
