Trapa japonica Flerov Extract Attenuates Lipid Accumulation through Downregulation of Adipogenic Transcription Factors in 3T3-L1 Cells  [PDF]

Abstract: Obesity is a major human health problem associated with various diseases, including cardiac injury and type 2 diabetes. Trapa japonica Flerov (TJF) has been used in traditional oriental medicine to treat diabetes. In this study, we evaluated the inhibitory effect of and the mechanism underlying the effect of TJF extract on adipogenesis in 3T3-L1 cells. The effects of TJF extract on cell viability were analyzed using a 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay, and the anti-adipogenic effect was measured by oil red O staining. The expression of peroxisomal proliferator activated receptor (PPAR)γ, CCAAT/enhancer-binding protein-α (C/EBP)α, adenosine monophosphate-activated protein kinase (AMPK), acetyl-CoA carboxylase (ACC), adiponectin, and fatty acid binding protein (FABP)4 involved in adipogenesis was determined by western blot analysis. TJF extract effectively inhibited lipid accumulation and the expression of PPARγ and C/EBPα in 3T3-L1 cells. TJF also increased the phosphorylation of AMPK and ACC, and decreased the expression of adiponectin and FABP4. These results indicate that TJF extract exerts its anti-obesity effect through the downregulation of adipogenic transcription factors and adipogenic marker genes.
Effect of Hoechunyangkyeok-San Extract on Melanogenesis  [PDF]

Abstract: Forsythia fructus has been shown to have antioxidative, anti-inflammatory, antibacterial, anti-aging and whitening effects. Hoechunyangkyeok-san (Forsythia viridissima-prescription) is a traditional herbal medicine, which has been clinically used for treating febrile and inflammatory disorders. This work was carried out to investigate the skin whitening effects of Forsythia viridissima-prescription extract (a hydrolyzed extract of Hoechunyangkyeok-san: SID White HYC) on skin. The effects of SID White HYC were assessed the melanin contents in B161 melanoma cells and the pigmented equivalent with HMB45 and Fontana Masson staining in 3D skin model. Then, we examined the expression of major pigment enzymes regulating melanin synthesis and melanosome transport related proteins in B16F1 cells. SID White HYC significantly inhibited the melanin synthesis (56.7% and 30.6% inhibition at 100 μg/mL, intracellular and secreted, respectively) in B16F1 cells and 3D skin model. In addition, western blotting analysis showed that SID White HYC reduced the expression of melanin synthesis and melanosome transport related proteins in B16F1 cells. In clinical trials, the cream containing 0.05% SID White HYC showed skin depigmentation effect without any irritation. These results suggest that SID White HYC may be useful inhibition of melanogenesis and melanosome transport. Therefore, SID White HYC may have potential as a skin-whitening ingredient in cosmetics.
Determinants Influencing Consumers’ Trust and Trust Performance of Social Commerce and Moderating Effect of Experience

Abstract: The main purpose of this study is to investigate the effects of various antecedents of trust in Social commerce (s-commerce) (reputation, size, information quality and communication) on consumers’ trust. In addition, the study examines the moderating effects of consumers’ s-commerce experience on the relationships between these trust antecedents and trust and the effect of trust on trust performance. The results based on a sample of 466 s-commerce users and structural equation modeling with SmartPLS 2.0 indicate significant effects of all trust antecedents; significant moderating effects of consumers’ s-commerce experience and a positive effect of trust on trust performance and suggest a new theory for IS research. In addition, the results have important implications for s-commerce firms wishing to develop consumers’ trust as well as effective business models.
Ethanol Extract of Dianthus chinensis L. Induces Apoptosis in Human Hepatocellular Carcinoma HepG2 Cells In Vitro

Abstract: Dianthus chinensis L. is used to treat various diseases including cancer; however, the molecular mechanism by which the ethanol extract of Dianthus chinensis L. (EDCL) induces apoptosis is unknown. In this study, the apoptotic effects of EDCL were investigated in human HepG2 hepatocellular carcinoma cells. Treatment with EDCL significantly inhibited cell growth in a concentration- and time-dependent manner by inducing apoptosis. This induction was associated with chromatin condensation, activation of caspases, and cleavage of poly (ADP-ribose) polymerase protein. However, apoptosis induced by EDCL was attenuated by caspase inhibitor, indicating an important role for caspases in EDCL responses. Furthermore, EDCL did not alter the expression of bax in HepG2 cells but did selectively downregulate the expression of bcl-2 and bcl-xl, resulting in an increase in the ratio of bax:bcl-2 and bax:bcl-xl. These results support a mechanism whereby EDCL induces apoptosis through the mitochondrial pathway and caspase activation in HepG2 cells.
Atypical florid vascular proliferation in appendix: a diagnostic dilemma

With the exception of angiodysplasia, vascular abnormalities of the gastrointestinal tract are uncommon [1]. Seven cases of florid vascular proliferations occurring in the intestinal tract have been reported in English literature [1-3]. All reported cases were associated with intussusception or mucosal prolapse and showed microscopic features that overlap with those of angiosarcoma. Florid vascular proliferations are thought to be a secondary response to repeated mucosal trauma and ischemia and differential diagnosis from angiosarcoma is necessary [1]. We herein report on a challenging case showing unusual microscopic findings of the appendix.A 41-year old male presented with melena. He had no other symptoms, such as migratory pain, right lower quadrant abdominal tenderness with guarding, or leukocytosis. Colonoscopy showed non-specific findings, except for adhesioned blood clots in the appendiceal orifice. He underwent laparoscopic appendectomy. No characteristic features were observed on the external surface. Cut sections showed a tiny polypoid mucosa with easy contact bleeding and the appendiceal lumen was filled with blood. Microscopically, inflammatory cells infiltration, diverticulum, parasite, and fecalith were not observed. An exophytic polypoid mass with extensive surface ulceration was observed. The superficial portion of the polyp showed pyogenic granuloma-like features and the deeper portion was composed of vaguely lobulated proliferation of closely packed small capillary-sized vessels and showed infiltrative growth and extension into muscularis propria (Figure 1). Induction of Neuronal Differentiation of Rat Muscle-Derived Stem Cells in Vitro Using Basic Fibroblast Growth Factor and Ethosuximide

Abstract: Several studies have demonstrated that basic fibroblast growth factor (bFGF) can induce neural differentiation of mesenchymal stem cells. In this study, we investigated the neural differentiation of muscle-derived stem cells (MDSCs) following treatment with bFGF and ethosuximide, a small molecule used as an anticonvulsant in humans. Stem cells isolated from rat skeletal muscle (rMDSCs) were pre-induced by culturing with 25 ng/mL bFGF for 24 h and then were transferred to a medium supplemented with or without 4 mM ethosuximide. Neuronal differentiation was assessed by immunocytochemical and western blotting analyses of marker expression. Immunocytochemistry of rMDSCs treated with bFGF and ethosuximide identified abundant cells expressing neuronal markers (TuJ1, neuron-specific class III β-tubulin; NeuN, neuronal nuclear antigen; and NF-MH; neurofilament M and H). Olig2 (oligodendrocyte transcription factor 2)-positive cells were also observed, indicating the presence of oligodendrocyte lineage cells. These findings were substantiated by western blotting analysis of marker proteins. In particular, the expression of NeuN and TuJ1 was significantly higher in rMDSCs treated with ethosuximide and bFGF than in cells stimulated with bFGF alone (NeuN, p < 0.05 and TuJ1, p < 0.001). Expression of the astrocyte marker GFAP (glial fibrillary acidic protein) was not detected in this study. Collectively, the results showed that treatment with bFGF and ethosuximide induced effective transdifferentiation of rMDSCs into cells with a neural-like phenotype. Notably, rMDSCs treated with a combination of bFGF plus ethosuximide showed enhanced differentiation compared with cells treated with bFGF alone, implying that ethosuximide may stimulate neuronal differentiation.
Effects of Antecedents of Collectivism on Consumers’ Intention to Use Social Commerce

Abstract: Social commerce (s-commerce), a subset of electronic commerce (e-commerce), involves social interactions and user contributions and facilitates the online buying and selling of a wide range products and services. Given that s-commerce encourages consumers to share product- and service-related information, it reflects collectivism, not individualism. Using the Technology Acceptance Model (TAM), this study employs the Structural Equation Modeling (SEM) method to investigate a research model incorporating consumers’ preferences, reliance, concern and norm acceptance as antecedents of collectivism. The results of a survey of 365 s-commerce users indicate that preferences, reliance and norm acceptance had significant effects on the perceived usefulness of s-commerce. In addition, the goodness-of-fit results indicate that collectivism and perceived ease of use accounted for 65.7% of the variance in the perceived usefulness of s-commerce. This study contributed to the literature by providing useful insights into the factors influencing consumers’ decision to adopt s-commerce.
Finite-Time-Finite-Size Scaling of the Kuramoto Oscillators

Abstract: Phase transition in its strict sense can only be observed in an infinite system, for which equilibration takes an infinitely long time at criticality. In numerical simulations, we are often limited both by the finiteness of the system size and by the finiteness of the observation time scale. We propose that one can overcome this barrier by measuring the nonequilibrium temporal relaxation for finite systems and by applying the finite-time-finite-size scaling (FTFSS) which systematically uses two scaling variables, one temporal and the other spatial. The FTFSS method yields a smooth scaling surface, and the conventional finite-size scaling curves can be viewed as proper cross sections of the surface. The validity of our FTFSS method is tested for the synchronization transition of Kuramoto models in the globally-coupled structure and in the small-world network structure. Our FTFSS method is also applied to the Monte-Carlo dynamics of the globally-coupled q-state clock model.

KIOM-79 Protects AGE-Induced Retinal Pericyte Apoptosis via Inhibition of NF-kappaB Activation In Vitro and In Vivo

Abstract: KIOM-79 is an herbal mixture of parched Puerariae radix, gingered Magnoliae cortex, Glycyrrhizae radix and Euphorbiae radix. In the present study, we determined the efficacy and possible mechanism of KIOM-79 on the advanced glycation end product (AGE)-modified bovine serum albumin (BSA)-induced apoptosis of cultured bovine retinal pericytes and rat retinal pericytes in Zucker diabetic fatty (ZDF) rats. Seven-week-old male ZDF rats were treated with KIOM-79 (50 mg/kg body weight) once a day orally for 13 weeks. KIOM-79 significantly inhibited pericyte apoptosis which were induced by the AGE-BSA treatment. The KIOM-79 treatment markedly suppressed the activation of nuclear factor-kappaB (NF-κB) through the inhibition of inhibitory κB kinase complex. In addition, the oral administration of KIOM-79 inhibited the changes in retinal vasculature (vascular hyperpermeability, acellular capillary). KIOM-79 strongly inhibited pericyte apoptosis, NF-κB activation and the expression of pro-apoptotic Bax and tumor necrosis factor-α. Our results suggest that KIOM-79 may exert inhibitory effects on AGE-induced pericyte apoptosis by blocking NF-κB activation, thereby ameliorating retinal microvascular dysfunction.
