Abstract: an immunoenzimatic assay (elisa) was standardized to determine the natural anti-band 3 antibodies by coupling with chemobond plates the band 3 protein obtained from the membrane of human erythrocytes by a combination of affinity and anion exchange chromatography. 3 g of band 3 protein per well were used. a level of detection of 1 g/ml of anti-band 3 antibody was achieved. the results showed the usefulness of the method to measure the levels of natural anti-band 3 antibodies in patients with drepanocytes, both in basal state and in the painful vasocclusive crises.

Abstract: Se purificaron anticuerpos naturales anti banda 3 de la clase IgG mediante cromatografía de afinidad con un aceptable nivel de pureza, rapidez y eficiencia, a partir de una mezcla de inmunoglobulinas humanas de uso parenteral. Abstract: Se estandarizó un método inmunoenzimático (ELISA) para la determinación de anticuerpos naturales anti banda 3 acoplando a placas Chemobond la proteína banda 3 obtenida a partir de la membrana de eritrocitos humanos mediante una combinación de cromatografía de intercambio aniónico y afinidad. Se emplearon 3 mg de la proteína banda 3 por pozo, lográndose un nivel de detección de 1 mg/mL de anticuerpo anti banda 3. Los resultados indican la utilidad del método para su empleo en la medición de los niveles de anticuerpos naturales anti banda 3 en pacientes con drepanocitosis, tanto en estado basal como en las crisis vasooclusivas dolorosas. Abstract: Se determinó la actividad de las vías clásica y alternativa del complemento, así como la cuantificación de algunos de sus componentes en 46 pacientes con anemia drepanocítica (AD) (hemoglobina SS), 21 sin complicaciones clínicas y 25 con estas; entre las más frecuentes encontramos: número de infecciones, úlceras maleolares, crisis vasooclusivas, hepáticas y aplásticas, considerando como límite un a o anterior a la toma de muestra de sangre. Se demostró una disminución significativa de la actividad de la vía alternativa, factor B y del componente C3 en el grupo de pacientes con complicaciones clínicas. Se observó una correlación significativa entre el número de crisis vasooclusivas e infecciones y la actividad de la vía alternativa, el factor B y el C3. Estos resultados sugieren que los fenómenos inflamatorios que pueden persistir en pacientes con AD en estado basal, fundamentalmente aquellos con historia anterior de complicaciones clínicas, pueden provocar alteraciones en la actividad del complemento The activity of the classic and alternative complement pathways, as well as the quantitation of some of their components was determined in 46 patients with sickle cell anemia (haemoglobin SS), 21 with no clinical complications and 25 with them. Among the most frequent complications we found: number of infections, malleolar ulcers, and vaso-occlusive, hepatic and aplastic crises. A year previous to the blood sample taking was considered as the limit. A significant reduction of the activity of the alternative pathway, factor B and component C3 was proved in the group of patients with clinical complications. A marked correlation was observed between the number of vaso-occlusive crises and infections and the activity of the alternative pathway, factor B and complement C3. These results suggest that the inflammatory phenomena that may persist among patients with basal sickle cell anemia, mainly those with prior history of clinical complications, may produce alterations in the complement activity

Abstract: La transmisión del virus de la hepatitis C (VHC) a través de la sangre constituye un problema de salud en Cuba y el mundo. Varios tipos de inmunoensayos diagnosticadores se han desarrollado para la certificación de sangre y generalmente cuentan con una elevada sensibilidad y especificidad diagnóstica en donantes sanos. Sin embargo, su comportamiento en muestras de pacientes multitransfundidos podría ser menos eficaz. Para evaluar la eficacia diagnóstica del inmunoensayo cubano UMELISA HCV 3er Gen. (TecnoSUMA SA, La Habana, Cuba) en muestras de pacientes multitransfundidos, se procesaron en paralelo 335 sueros de pacientes por los sistemas UBI HCV EIA 4.0 (United Biomedical, EE.UU.) y UMELISA HCV 3er Gen., y las muestras con resultados incongruentes se confirmaron por el sistema de PCR COBAS AmpliScreen HCV Test, v2. (Roche, EE.UU.). Cuando se comparó el sistema UMELISA HCV 3er Gen. con el UBI HCV EIA 4.0 se obtuvo una Sd de 95,8 % IC (95 %): 92,599,15 y Ed de 100 % IC (95 %): 99,7-100, con IY: 0,96 (0,93-0,99). k: 0,9582 ID (95 %): 0,9276-0,9888, p=0,000. Ambos sistemas de inmunoensayos fueron satisfactorios para el inmunodiagnóstico de pacientes multitransfundidos. Hepatitis C virus (CHV) blood-transmission is a health problem in Cuba and in the world. Some types of diagnostic immunoassays have been developed for the blood certification and in general have a high diagnostic sensitivity and specificity in healthy donors. However, its behavior in samples from multi-transfusion patients could by less effective. To assess the diagnostic effectiveness of the UMELISA HCV third generation Cuban immunoassay (TecnoSUMA, S.A. La Habana), Cuba) in samples from multi-transfusion patients, in parallel, 335 sera from patients were processed by UBI HCV EIA 4.0 (United Biomedical, EE.UU) and UMELISA HCV third generation, and the samples with incongruous results were verified by PCR COBAS AmpliScreen HCV Test, v2 system (Roche, EE.UU.) Comparing the UMELISA HCV third generation system with the UBI HCV EIA 4.0 it was achieved a Sd of 95,8% CI(95%): 92,5-99,15 and a Ed of 100% CI (95%): 99,7-100, with IY: 0,96 (0,93-0,99) with k: 0,0582 ID (95%): 0,9276-0,9888, p = 0,000. Both immunoassay systems were satisfactory for immunodiagnosis of multi-transfusion patients.

Abstract: a concentrated preparation of component c1 (q,r,s) of the complement system in an inactive form was obtained by a precipitation method, free of contamination with the rest of the initial components of the classical pathway, that is, c2 and c4. this preparation remained stable at 2°c and in the functional assays, it was able to bind to complex eac4, to activate components c2 and c4 to form convertase c3 and cause lysis of these cells. hemolysis reaction was activated by c2 and inhibited by c1 inhibitor, which indicates the specificity of the obtained c1. this preparation can be used to attain a specific antiserum to obtain subcomponents c1q, c1r and c1s or to standardize the functional methods of study of c1, c2 and c1 inhibitor

Abstract: La lectina de unión a la manosa (MBL) es una colectina que se sintetiza en el hígado y es secretada al torrente sanguíneo, la cual es capaz de unirse con estructuras repetidas de azúcares presentes en una amplia variedad de bacterias y otros microorganismos promoviendo su eliminación mediante la activación del complemento a través de serín proteasas asociadas. A las deficiencias de MBL se les considera como un importante factor de riesgo de infecciones en ni os y en individuos inmunosuprimidos. Se discute la evidencia de que la MBL contribuye de forma importante a la inmunidad innata con el incremento de la susceptibilidad a determinadas enfermedades o la incidencia en el curso de estas. Estudios preliminares del empleo de terapias sustitutivas con MBL han arrojado resultados prometedores, los que deben ofrecer evidencias acerca del significado fisiológico de esta proteína Manosse-binding lectin (MBL) is a colectin that is synthesized in the liver and is secreted to the the blood stream. It is capable of binding to other repeated structures of sugars present in a wide variety of bacteria and other microorganisms, promoting their elimination by the activation of the complement through associated serin proteases. The MBL deficiencies are considered as an important risk factor for infections in children and immunosuppressed individuals. It is discussed the evidence that MBL contributes in an important way to the innate immunity with the increase of susceptibility to certain diseases or the incidence on their course. Preliminary studies on the use of replacement therapies with MBL have yielded encouraging results, which should offer evidences about the physiological significance of this protein
