Anna Rachel Gontijo Mazoni
Movimento , 2005,

Abstract: We propose a novel method for describing binding properties and delineating serine proteases specificity by compiling an exhaustive table of interface forming residues (IFR) for serine proteases and their inhibitors. Currently, the Protein Data Bank (PDB) does not contain all the data that our analysis would require. Therefore, an in silico approach was designed for building corresponding complexesThe IFRs are obtained by "rigid body docking" among 70 structurally aligned, sequence wise non-redundant, serine protease structures with 3 inhibitors: bovine pancreatic trypsin inhibitor (BPTI), ecotine and ovomucoid third domain inhibitor. The table (matrix) of all amino acid positions at the interface and their respective occupancy is created. We also developed a new computational protocol for predicting IFRs for those complexes which were not deciphered experimentally so far, achieving accuracy of at least 0.97.The serine proteases interfaces prefer polar (including glycine) residues (with some exceptions). Charged residues were found to be uniquely prevalent at the interfaces between the "miscellaneous-virus" subfamily and the three inhibitors. This prompts speculation about how important this difference in IFR characteristics is for maintaining virulence of those organisms. Our work here provides a unique tool for both structure/function relationship analysis as well as a compilation of indicators detailing how the specificity of various serine proteases may have been achieved and/or could be altered. It also indicates that the interface forming residues which also determine specificity of serine protease subfamily can not be presented in a canonical way but rather as a matrix of alternative populations of amino acids occupying variety of IFR positions.Serine proteases play an important role in processes such as blood clotting, digestion and in some pathways of cell development [1]. Serine proteases can hydrolyze either peptide bonds or esters. Proteases digest protei

Abstract: objective: this work was designed to validate the portuguese version of the contemplation ladder, whose purpose is to assess the motivational phase to quit smoking among tobacco users using a telephone service. method: a cross-sectional study was conducted in a nationwide drug use information hotline. in order to assess the convergent validation, the correlation between the contemplation ladder and the urica scale was calculated, which was previously validated. results: the study included 271 tobacco users. statistically significant correlations were found between the contemplation ladder scores and the scores of the urica precontemplation (r=-0.16; p<0.01), action (r=0.15; p<0.01) and maintenance (r=0.18; p<0.01) subscales. the correlation between the urica scale compound score and the contemplation ladder was also significant (r=0.31; p<0.01). conclusion: the results of our study suggest that the contemplation ladder can be an efficient substitute for the urica scale (whose application lasts at least 20 minutes), without submitting the interviewee to a heavy load of questions. the study presented evidences of convergent validity for the contemplation ladder when applied via telephone in tobacco users.

Abstract: the objective of this study is to analyze the constraints faced by the nursing assistants and technicians in a pediatric clinic sponsored by a state foundation during their work activities and identify the strategies adopted by them in order to guarantee the achievement of their production goals and job satisfaction and ensure their protection against workplace accidents. the method of ergonomic work analysis (ewa) was used for analysis and data collection of 59 tenured and temporary nursing workers with intermediate and technical level of education and to investigate the strategies related to actions and employment regulation concerning the interfaces they deal with. the results show that among the constraints assessed in this institution the most evident are: double shift, diversity of employment contracts (tenure and temporary), risks to which these professionals are exposed, and the lack of knowledge about their object of work. it can be said that these constraints increase physical, psychic, and cognitive workload. aiming to reduce such workloads, the workers try to develop strategies from their own experience, expertise, and values in order to deal with the risks in the workplace.

Abstract: smoking cessation is associated to health benefits, because individuals who stop smoking will avoid most tobacco-related disorders. our aim was to review the most important characteristics and the efficacy of the pharmacological and psychossocial treatments available for tobacco smokers, presenting meta-analysis and randomized clinical trials fundamental conclusions. pharmacological interventions involving nicotine replacement and antidepressant use, with bupropion as the first line agent, and psychossocial interventions involving cognitive behavioral therapies and face-to-face or phone-based motivational interventions are proving to be efficacious. pharmacotherapies increase two-fold the chance of abstinence and face-to-face or phone-based psychosocial interventions increase the chance of quitting smoking in 1.5 to 2.5 times in comparison to individuals who try to quit smoking by themselves. these data support health professionals to decide the most effective treatment for individual smokers, according to the available interventions tested, their efficacy and the benefits of use.
