A 2D-QSPR approach to predict blood-brain barrier penetration of drugs acting on the central nervous system

Brazilian Journal of Pharmaceutical Sciences , 2010, DOI: 10.1590/S1984-82502010000400016
Abstract: drugs acting on the central nervous system (cns) have to cross the blood-brain barrier (bbb) in order to perform their pharmacological actions. passive bbb diffusion can be partially expressed by the blood/brain partition coefficient (logbb). as the experimental evaluation of logbb is time and cost consuming, theoretical methods such as quantitative structure-property relationships (qspr) can be useful to predict logbb values. in this study, a 2d-qspr approach was applied to a set of 28 drugs acting on the cns, using the logbb property as biological data. the best qspr model [n = 21, r = 0.94 (r2 = 0.88), s = 0.28, and q2 = 0.82] presented three molecular descriptors: calculated n-octanol/water partition coefficient (clogp), polar surface area (psa), and polarizability (α). six out of the seven compounds from the test set were well predicted, which corresponds to good external predictability (85.7%). these findings can be helpful to guide future approaches regarding those molecular descriptors which must be considered for estimating the logbb property, and also for predicting the bbb crossing ability for molecules structurally related to the investigated set.

Emphasis was given to studies and proposals that characterize the so-called territorial dimension of metropolitan socio-spatial processes. The analysis identified housing, especially social interest, as a central issue of such processes. Related to housing werehighlighted the issues of transport and mobility, economic development and employment, environmental protection, and joint municipal and metropolitan policies. The analysiscontributed to assess possibilities of integrated territorial management, and also to enlargethe understanding of urban socio-spatial processes.
Abstract: OBJECTIVE: Growing evidence suggests that sudden death after an acute myocardial infarction (AMI) correlates with autonomic nervous system imbalance. Parasympathomimetic drugs have been tested to reverse these changes. However, their effects on ventricular function need specific evaluation. Our objective was to analyze pyridostigmine's (PYR) effect on hemodynamic and echocardiographic variables of ventricular function. METHODS: Twenty healthy volunteers underwent Doppler echocardiographic evaluations, blood pressure (BP), and heart rate (HR) assessment at rest, before and 120 min after ingestion of 30 mg PYR or placebo, according to a double-blind, placebo-controlled, crossed and randomized protocol, on different days. RESULTS: PYR was well tolerated and did not cause alterations in BP or in ventricular systolic function. A reduction in HR of 10.9±1.3% occurred (p<0,00001). There was an A wave reduction in the mitral flow (p<0.01) and an E/A ratio increase (p<0.001) without changes in the other diastolic function parameters (p>0.05). CONCLUSION: PYR reduces HR and increases E/A ratio, without hemodynamic impairment or ventricular function change.

Revista Brasileira em Promo??o da Saúde , 2012,
Abstract: Objective: To evaluate the knowledge of pharmacists who work in commercial pharmacies in Fortaleza, Ceará, Brazil, about breastfeeding. Methods: This was an observational, descriptive and transversal study. We used a questionnaire approaching the profile of pharmacists and general knowledge about breastfeeding and Brazilian Standard for Marketing of Food for Infants and Young Child, teats, dummies and Bottles (BSMFI). Data collection occurred from January to March 2011. Through drawing, we selected 82 commercial pharmacies, corresponding to 15.5% of all pharmacies in Fortaleza, when held up contact with their pharmacists. The questionnaire was administered to 67 pharmacists. The results were statistically analyzed using the Epi Info v. And v 3.5.1 SigmaPlot. 10.0. Results: Among the participants, 72% (n = 48) were female, 35.8% (n = 24) are formed over ten years, 49.3% (n = 33) working in pharmacy business for less than five years, and 52.2% (n = 35) engaged in professional activity at other establishments. The level of knowledge was rated as fair to 71.7% (n = 48) of respondents; insufficient to 17.9% (n = 12) and good for 10.4% (n = 07). This low performance, not reaching the maximum score, was associated with exercise time in the profession, noting that trained professionals for more than ten years had lower knowledge. Conclusion: Respondents recognize the importance of breastfeeding for both the baby’s health as for the mother. However, they lack knowledge of relevant information concerning the correct management of breastfeeding, especially among those who graduated earlier.
Masculinidades e experiências masculinas em Bernardo Guimar?es

Abstract: this article aims to identify and analyze different aspects of masculinity presented in the literature of bernardo guimar?es, an important 19th century brazilian writer from minas gerais. we shall focus how the author, in his literary work, constructs the gender relations and produces representations about men and women, specially the masculine experiences, without forgetting the feminine ones. we intend, in the same way, to contribute to a broader understanding of the participation of literature in the production of representations and practices of masculinity (and of femininity) in late nineteenth-century brazil.

Abstract: musculoskeletal pain and functionality are linked to the individual health and, thus, influence the mode of interaction between individuals and between them and the environment that surrounds them. however, there are not many studies that question such health issues concerning the population in general, especially when we look at these issues from the basic care point of view. the aim of this article is to present initial data on the subject of pain and functionality in individuals with musculoskeletal complaints enrolled in a primary care unit and prompt a discussion on how complete the services provided by these units are. this study was carried out in a primary care unit and various households, among subjects aging 20 years or older. a functionality assessment tool and a visual analogue scale were applied, and social and demographic issues were also considered. by using these tools, it was made an attempt to evaluate the profile of individuals in association with their degree of musculoskeletal pain and the degree of functionality in the activities of their daily lives, ages and occupations. we found a predominance of women in both groups: 84.37% and 81.25%, respectively. nearly half of all people with musculoskeletal pain were housewives, and the average age between groups was over 55.
