Proteome-Wide Analysis of Disease-Associated SNPs That Show Allele-Specific Transcription Factor Binding

Abstract: A causative role for single nucleotide polymorphisms (SNPs) in many genetic disorders has become evident through numerous genome-wide association studies. However, identification of these common causal variants and the molecular mechanisms underlying these associations remains a major challenge. Differential transcription factor binding at a SNP resulting in altered gene expression is one possible mechanism. Here we apply PWAS (“proteome-wide analysis of SNPs”), a methodology based on quantitative mass spectrometry that enables rapid screening of SNPs for differential transcription factor binding, to 12 SNPs that are highly associated with type 1 diabetes at the IL2RA locus, encoding the interleukin-2 receptor CD25. We report differential, allele-specific binding of the transcription factors RUNX1, LEF1, CREB, and TFAP4 to IL2RA SNPs rs12722508*A, rs12722522*C, rs41295061*A, and rs2104286*A and demonstrate the functional influence of RUNX1 at rs12722508 by reporter gene assay. Thus, PWAS may be able to contribute to our understanding of the molecular consequences of human genetic variability underpinning susceptibility to multi-factorial disease.

Abstract: Biomarker-based cross-sectional incidence estimation requires a Recent Infection Testing Algorithm (RITA) with an adequately large mean recency duration, to achieve reasonable survey counts, and a low false-recent rate, to minimise exposure to further bias and imprecision. Estimating these characteristics requires specimens from individuals with well-known seroconversion dates or confirmed long-standing infection. Specimens with well-known seroconversion dates are typically rare and precious, presenting a bottleneck in the development of RITAs.
Sharing Experiences: Establishing an Expertise Centre on the Protection of Dutch Cultural Heritage

Abstract: In the Netherlands an Expertise Centre on the Protection of Dutch Cultural Heritage is being established to bring together scattered knowledge and expertise on collection safety and security issues. A website is being set up based on a broad, integrated security concept. The website will include the DICE database for incident registration. More specialised expertise will be made available on demand. The Centre has been funded for two years, whereupon its viability will be assessed. For the Centre to become a success, the entire cultural heritage field should overcome its reluctance to share sensitive information and commit itself to the Centre.
Appraising the e-readiness of online learning facilitators: key human factors

Abstract: Higher education institutions in South Africa have undergone a number of changes over the past few years. These changes brought about inter alia changed work environments and job demands. One of the new job demands is the need to incorporate technology in teaching and learning, viz. e-learning. Not all job incumbents, however, adapted successfully to these changes, particularly with regard to e-readiness. Such a lack in e-readiness is likely to influence the effectiveness with which an academic employee will fulfil his/her online learning duties. Therefore, it is important to find solutions to overcome the lack of e-readiness. This article will focus on the role of human resource appraisal, in order to assess the e-readiness of online learning facilitators with a view to improving their online skills and capacity. A number of human factors that can play a role in employee performance and motivation, namely learning styles, personal profile patterns, and pace and style of technology adoption will be briefly outlined, in order to determine the role that these factors could play in assessing the e-readiness of online learning facilitators.
De dagbladkritiek van vóór en ná Merlyn. Verschillen tussen de literatuurkritische praktijk in 1955 en 1975

Abstract: This study investigates the differences between literary reviewing in the Dutch newspapers before and after the literary magazine Merlyn. The reason to do so is the claim of many scholars that Merlyn influenced literary reviewing in the Netherlands, more specifically the kind of value judgments literary critics make. To find out whether this specific influence on the judgments is a myth or not, the reviews in the newspapers before and after Merlyn have been examined. The value judgments in prose and poetry reviews that appeared in Het Parool, de Volkskrant and Trouw in two reference years (1955 and 1975) were collected and categorized in order to make a comparison. The analysis shows several differences between 1955 and 1975 that may be interrelated with Merlyn. Although, of course, the differences found cannot be attributed to Merlyn solely, this study seems to confirm the idea that Merlyn influenced literary reviewing in the newspapers.

Abstract: It is always difficult to define exactly why a particular author has been awarded the Nobel Prize for Literature. This approach will deal with four different aspects which may have contributed to Wole Soyinka's. award. It cannot be denied that Soyinka is the author of an extensive and richly varied work, which has been appreciated by critics the world over. Especially the satirical qualities of his style have been praised on many occasions.But we have to focus our attention on two other important aspects. We can only achieve a full understanding of Soyinka's dramas when we interpret them against the background of Yoruba drama and Yoruba cultural tradition. And we can only appreciate Soyinka's work if we pay attention to the social context in which it is situated. This political commitment, as well as the international reputation Soyinka has helped to create as a director and an actor, are essential elements that have contributed to this Nobel Prize award.

Abstract: African literature, like any other literature remote in space or time, can be approached in two different ways. We can read it from our own point of view and try to appreciate it with our own literary standards. We can also try to read it as it was experienced and appreciated by its immediate audience, which means by the African public at the time of publication. Reactions given by a group of Dutch-speaking readers on a novel by E. Dongala clearly illustrate that the second approach is hardly achievable, however attractive it may seem. So we have to accept that every reading of an African literary work implies a serious degree of creative treason.
Does Curriculum 2005 promote successful learning of elementary algebra?

Abstract: This article reviews literature, previous to the development of Curriculum 2005, describing possible causes and solutions for learners’ poor performance in algebra. It then analyses the Revised National Curriculum Statement for Mathematics in an attempt to determine whether it addresses these causes and suggested solutions. This analysis finds that the curriculum to a large extent does address them, but that some are either not addressed, or addressed only implicitly. Consequently, Curriculum 2005 may only partly promote successful learning of elementary algebra.
From Darwin to the Census of Marine Life: Marine Biology as Big Science

Abstract: With the development of the Human Genome Project, a heated debate emerged on biology becoming ‘big science’. However, biology already has a long tradition of collaboration, as natural historians were part of the first collective scientific efforts: exploring the variety of life on earth. Such mappings of life still continue today, and if field biology is gradually becoming an important subject of studies into big science, research into life in the world's oceans is not taken into account yet. This paper therefore explores marine biology as big science, presenting the historical development of marine research towards the international ‘Census of Marine Life’ (CoML) making an inventory of life in the world's oceans. Discussing various aspects of collaboration – including size, internationalisation, research practice, technological developments, application, and public communication – I will ask if CoML still resembles traditional collaborations to collect life. While showing both continuity and change, I will argue that marine biology is a form of natural history: a specific way of working together in biology that has transformed substantially in interaction with recent developments in the life sciences and society. As a result, the paper does not only give an overview of transformations towards large scale research in marine biology, but also shines a new light on big biology, suggesting new ways to deepen the understanding of collaboration in the life sciences by distinguishing between different ‘collective ways of knowing’.
Nucleotide Excision Repair in Caenorhabditis elegans

Abstract: Nucleotide excision repair (NER) plays an essential role in many organisms across life domains to preserve and faithfully transmit DNA to the next generation. In humans, NER is essential to prevent DNA damage-induced mutation accumulation and cell death leading to cancer and aging. NER is a versatile DNA repair pathway that repairs many types of DNA damage which distort the DNA helix, such as those induced by solar UV light. A detailed molecular model of the NER pathway has emerged from in vitro and live cell experiments, particularly using model systems such as bacteria, yeast, and mammalian cell cultures. In recent years, the versatility of the nematode C. elegans to study DNA damage response (DDR) mechanisms including NER has become increasingly clear. In particular, C. elegans seems to be a convenient tool to study NER during the UV response in vivo, to analyze this process in the context of a developing and multicellular organism, and to perform genetic screening. Here, we will discuss current knowledge gained from the use of C. elegans to study NER and the response to UV-induced DNA damage. 1. DNA Damage Response Mechanisms To preserve and faithfully transmit DNA to the next generation, cells are equipped with a variety of DNA repair pathways and associated DNA damage responses, collectively referred to as the DNA damage response (DDR). DNA is continuously damaged by environmental and metabolism-derived genotoxic agents. It is vital for cells and organisms to properly cope with DNA damage, because unrepaired damage can give rise to mutation and cell death. The importance of the DDR is illustrated by several human cancer prone and/or progeroid hereditary diseases, which are based on defects in the DDR. Over the last decades, a wealth of information on the molecular mechanism of different repair pathways has been gathered from detailed in vitro and live cell studies. Currently, this acquired knowledge is being used to develop therapeutic strategies to treat patients suffering from the consequences of unrepaired DNA damage, such as cancer and aging [1]. Damage is repaired by different DNA repair pathways depending on the type of DNA lesion, genomic location, and the cell cycle phase (for reviews see [2–4]). Lesions originating from different genotoxic sources can range from small base modifications to double-strand breaks. Small base modifications, such as oxidative lesions which do not substantially distort the double helix, are repaired by base excision repair (BER).
