Abstract:

Repeat blocks, microsatellites or simple sequence repeats (SSRs) can produce good co-dominant molecular markers for genetic diversity analysis and the determination of self-pollination rates in progenies originating from open pollination of selected genotypes. The enrichment of guarana genomic libraries was underway when it was confirmed that we are working with a complex polyploid species with 210 chromosomes. The probes (CA)12, (CT)12 and (TC)14 were used to finish the enrichment of four libraries for repeat blocks and the screening of a databank of expressed sequence tags (ESTs) from guarana seeded-fruits was accomplished as well. Fifteen clonal cultivars were genotyped with three replicas at 10 out of 27 identified loci using the 59 alleles that passed the reproducibility criterion. A large number of short repeat blocks were identified and this was considered to be a consequence of the recent polyploidization event. However, blocks with eight or more repeats ideal for genotyping were scarce. Annealing of most probes to short blocks by partial complementarity could explain the scarcity of longer blocks in genomic libraries but cannot explain why they were rare in the ESTs. Due to the complexity of the genotypes, alleles were treated as dominant traits. ESTs harboring repeat blocks had the functional annotation renewed. Locus GRN07 is inserted in a homologue of the MOTHER OF FLOWERING LOCUS T AND TFL1 (MFT), in which 3’-UTR displays clear post-transcriptional regulatory features. MFT and its variants are probably involved in the determination of seed germination and embryo growth characteristics. Other accessed loci can be involved in plant architecture and defense reactions. It was concluded that the

Abstract: the puraquequara river and its right-bank's tributary, the água branca stream, are located in the outskirts of the manaus urbanized area and their basin is still harbored by the primary rain forest cover. they exhibit 'terra firme' geologic, pedologic and climatological characteristics pertaining to the amazonian region. samples for this research were collected in november 1998 (dry season) and april 1999 (rainy season). water temperature, ph, turbidity, conductivity, alkalinity, hardness, cod, ca2+, mg2+, na+ e k+, sio2, no3-, no2-, po43-, so42-, cl-, nh4+ and fe were analysed. the water is acid (3.8 to 4.1) turning into less acid in the dry season. the alkalinity and turbidity are higher in the dry season whereas conductivity, hardness and cod are higher in the rainy season. sio2 and cl- are the main anionic elements where the former's contribution is higher in the puraquequara and lower in the água branca while the latter shows inverse contribution. na+, fe, and nh4+ are the main cations with their higher contribution in the dry season. nh4+ is the only cation that tends to increase its contribution downstream in the wet season. k+ contents is higher than that of mg+ and ca2+ yet na+ is the highest. the higher than 0.2 mg/l and 0.5 mg/l amounts of nh4+ and no3- respectively, are traces of water contamination. these characteristics define these waters as very diluted with anions predominating over cations and correlated to the black water of the region.

Abstract: This work analyzes the morphological alterations in Brachyplatystoma rousseauxii gills and evaluates whether this species can be considered an environmental monitoring biomarker of Guajará Bay. Sampling was carried out in four areas around Belém, Brazil, in four annual periods: dry/wet season, wet season, wet/dry season and dry season. Water pH, temperature and suspended material were evaluated. A total of 36 specimens were collected. The second right gill arch of each animal was removed and immediately fixed and processed for histopathology analysis with light microscopy. The physicochemical analysis of the water during the study period showed slight acidity, temperature and material in suspension within normal levels. Histopathological analysis of the gills from 14 individuals from area I presented no alterations, and only 2 individuals from this area presented some significant type of alteration. In contrast, all individuals captured in areas II, III, and IV presented at least one of the following alterations: aneurism-like alterations, epithelial elevation, infiltration, cell proliferation and cell hypertrophy. Based on the gill histopathological analysis, this organ is considered a good biomarker and the native species B. rousseauxii could be used as a bioindicator for environmental monitoring.

Abstract: The sustainability and production of collared peccary (Pecari tajacu) has been studied in the last few years; however, further information on its reproduction is necessary for breeding systems success. Understanding folliculogenesis aspects will contribute to effective reproductive biotechniques, which are useful in the preservation and production of wildlife. The aim of this study was to evaluate the ovarian folliculogenesis in collared peccary. Ovaries from six adult females of collared peccary were obtained through ovariectomy and analyzed. These were fixed in aqueous Bouin’s solution and sectioned into 7μm slices, stained with hematoxilin-eosin and analyzed by light microscopy. The number of pre-antral and antral follicles per ovary was estimated using the Fractionator Method. The follicles, oocytes and oocyte nuclei were measured using an ocular micrometer. Results showed that the length, width, thickness, weight, and the gross anatomy of the right and left ovaries were not significantly different. However, the mean number of corpora lutea was different between the phases of the estrous cycle (p<0.05), with the highest mean in the luteal phase. Primordial follicles were found in the cortex; the oocytes were enveloped by a single layer of flattened follicular cells. In the primary follicles, proliferation of the follicular cells gave rise to cuboidal cells (granulosa cells). The secondary follicle was characterized by two or more concentric layers of cuboidal cells (granulosa), beginning of antrum formation, and the presence of pellucid zone and theca cells. Antral follicles were characterized by a central cavity (antrum), the presence of cumulus oophorus and theca layers (interna and externa). In the right ovary, the values of the primordial and primary follicles were similar, but significantly different from the secondary ones (p<0.05). In the left ovary, significant differences were observed between all follicles in the follicular phase (p<0.05); the mean number of primordial and primary follicles was similar in the luteal phase. The mean number of pre-antral follicles and antral follicles in the follicular phase was higher in the left ovary (p<0.05). The mean number of antral follicles in the luteal phase was similar in both ovaries. We also found significant differences in mean diameter of preantral follicles, oocyte, granulosa layer and oocyte nucleus during the estrous cycle. In the antral follicles a significant difference was observed only in follicular diameter (p<0.05). The predominance of active primordial and primary follicles was found

Abstract: this article analyzes the history of primary health care policy in the federal district, brazil, based on the theoretical framework of historical neo-institutionalism, identifying the predominant configurations and trends in the various administrations of the state health secretariat (ses-df) from 1960 to 2007. the study indicates that the characteristics of the federal district's health policy are dependent on the history of the original health system plans for setting priorities and goals, as well as for the health system's implementation. this influence, in addition to the centralization of decision-making processes and limited political participation, can contribute to making primary care ancillary to hospital care, thus jeopardizing its potential to produce change in the health care model.
