Immunization with a Borrelia burgdorferi BB0172-Derived Peptide Protects Mice against Lyme Disease

Abstract: Lyme disease is the most prevalent arthropod borne disease in the US and it is caused by the bacterial spirochete Borrelia burgdorferi (Bb), which is acquired through the bite of an infected Ixodes tick. Vaccine development efforts focused on the von Willebrand factor A domain of the borrelial protein BB0172 from which four peptides (A, B, C and D) were synthesized and conjugated to Keyhole Limpet Hemocyanin, formulated in Titer Max? adjuvant and used to immunize C3H/HeN mice subcutaneously at days 0, 14 and 21. Sera were collected to evaluate antibody responses and some mice were sacrificed for histopathology to evaluate vaccine safety. Twenty-eight days post-priming, protection was evaluated by needle inoculation of half the mice in each group with 103 Bb/mouse, whereas the rest were challenged with 105Bb/mouse. Eight weeks post-priming, another four groups of similarly immunized mice were challenged using infected ticks. In both experiments, twenty-one days post-challenge, the mice were sacrificed to determine antibody responses, bacterial burdens and conduct histopathology. Results showed that only mice immunized with peptide B were protected against challenge with Bb. In addition, compared to the other the treatment groups, peptide B-immunized mice showed very limited inflammation in the heart and joint tissues. Peptide B-specific antibody titers peaked at 8 weeks post-priming and surprisingly, the anti-peptide B antibodies did not cross-react with Bb lysates. These findings strongly suggest that peptide B is a promising candidate for the development of a new DIVA vaccine (Differentiate between Infected and Vaccinated Animals) for protection against Lyme disease.
Frequency and Clinical Epidemiology of Canine Monocytic Ehrlichiosis in Dogs Infested with Ticks from Sinaloa, Mexico

Abstract: Ehrlichia canis is a rickettsial intracellular obligate bacterial pathogen and agent of canine monocytic ehrlichiosis. The prevalence of this disease in veterinary medicine can vary depending on the diagnostic method used and the geographic location. One hundred and fifty-two canine blood samples from six veterinary clinics and two shelters from Sinaloa State (Mexico) were analyzed in this study. All animals were suspected of having Canine Monocytic Ehrlichiosis (CME). The diagnostic methods used were the ELISA (Snap4Dx, IDEXX) together with blood smear and platelet count. From all dogs blood samples analyzed, 74.3% were positive to E. canis by ELISA and 40.1% were positive by blood smear. The sensitivity and specificity observed in the ELISA test were 78.8% and 86.7%. In addition, thrombocytopenia was presented in 87.6% of positive dogs. The predominant clinical manifestations observed were fever, anorexia, depression, lethargy, and petechiae. Consequently, this is the first report in which the morulae were visualized in the blood samples, and E. canis-specific antibodies were detected in dogs from Sinaloa, Northwest of Mexico. 1. Introduction Ehrlichia canis is the causative agent of canine monocytic ehrlichiosis (CME). Moreover, CME is an emerging disease in veterinary medicine, and E. canis has been considered in the last decade as a potential zoonotic pathogen [1, 2]. It is a worldwide disease transmitted by a tick bite. The competent vector for its transmission is the Ixodidae ticks Rhipicephalus sanguineus and Dermacentor variabilis [3]. In dogs, the CME is a multiphase disorder that progresses in three stages: acute, subclinical, and chronic. Each phase is characterized by several clinical and hematologic abnormalities. Thrombocytopenia is a common finding in E. canis infected dogs and many clinicians tend to use it as an indication for antibiotic treatment, and it is observed in 84% of the cases and its severity varies in the different disease phases [4]. During the subclinical stage a moderate thrombocytopenia is observed, while the chronic phase is characterized by severe leukopenia and anemia. In this stage dogs show other complications such as hypocellular marrow, suppressed splenic sequestration, decreased life of platelets, and an increase of circulating migration-factor platelet inhibitor [3, 4]. The relationship between the magnitude of thrombocytopenia and prevalence of E. canis has been established in countries such as Brazil in 2004 where 84.1% of infected dogs showed thrombocytopenia [3]. These qubits are either based on the quantum state of a single particle (semiconductor qubits), or on the quantum state of the global circuit (superconducting qubits). Abstract: Corticosteroids are still the first-line treatment for active ulcerative colitis more than 50 years after the publication of trials assessing their beneficial effect, with about a 50% remission rate in cases of severe disease. The mortality related to severe attacks of ulcerative colitis has decreased dramatically, to less than 1%, in experienced centers, due to the appropriate use of intensive therapeutic measures (intravenous steroids, fluids and electrolytes, artificial nutritional support, antibiotics, etc), along with timely decision-making about second-line medical therapy and early identification of patients requiring colectomy. One of the most difficult decisions in the management of severe ulcerative colitis is knowing for how long corticosteroids should be administered before deciding that a patient is a non-responder. Studies assessing the outcome of acute attacks after steroid initiation have demonstrated that, in steroid-sensitive patients, the response generally occurs early on, in the first days of treatment. Different indexes to predict treatment failure, when applied on the third day of treatment, have demonstrated a high positive predictive value for colectomy. In contrast to this resolute approach, which is the most widely accepted, other authors have suggested that in some patients a complete and prolonged response to steroids may take longer. Either way, physicians taking care of these patients need to recognize that severe ulcerative colitis may be life-threatening, and they need to be careful with excessively prolonged medical treatment and delayed surgery.
Caballos y ciervos : Una aproximación a la evolución climática y económica del Paleolítico superior en el Mediterráneo peninsular

Abstract: Caballos y ciervos son las principales presas de caza mayor en los yacimientos del Paleolítico superior en la cuenca mediterranea de la Península Ibérica. Sólo las cabras monteses pueden llegar a tener mayor importancia en yacimientos situados en áreas abruptas o monta osas. A pesar de los pocos requerimientos climáticos de ambas especies, podemos comprobar una clara relación entre los episodios más fríos del final del Pleistoceno y la caza del caballo y la presencia de los restos de ciervo en los yacimientos arqueológicos durante los momentos más templados. Nuestros resultados indican la existencia de una especie de frontera bioclimática Equus/Cervus que se desplazó de sur a norte en la cuenca medietrranea de la peninsula Ibérica a lo largo del Paleolítico superior.
Density of states in a superconductor carrying a supercurrent

Abstract: We have measured the tunneling density of states (DOS) in a superconductor carrying a supercurrent or exposed to an external magnetic field. The pair correlations are weakened by the supercurrent, leading to a modification of the DOS and to a reduction of the gap. As predicted by the theory of superconductivity in diffusive metals, we find that this effect is similar to that of an external magnetic field.

Abstract: introduction: a study carried out in the city of braga, in february 2008 (shortly after the entry into force of law 37/2007 of august 14 of tobacco control), in hospitality venues revealed that there are establishments that allow smoking inside their facilities. this raised the problem of knowing what the concentration of nicotine in the air present in these venues is (an indicator of pollution by tobacco smoke). participants and methods: the air quality was assessed in march 2009, in six hospitality venues, of the city of braga: two smoke free restaurants under 100 m 2; two coffees less than 100 m 2 (a smoke free coffeehouse and another that does not allow consumption); a smoke free night bar with less than 100 m 2 and a nightclub with a smoking area and a non-smoking room, separated by a curtain of air. measurement of vapor phase nicotine in air was done by active monitoring, through the use of monitors, using the methodology applied by hammond 1. particles (pm 2.5) were measured with a side pack. results: the average concentration of nicotine inside the smoke free coffeehouses and restaurants is 6.29 mg/m3, while inside the coffeehouses where smoking is not allowed is 1.1 mg/m3. the concentration of nicotine in the night bar has a very high value (9.42 mg/m3). at the disco, the concentration of nicotine is the highest recorded in enclosed spaces (19.1 mg/m3 in the area where smoking is forbidden and 10.2 mg/m3 in a smoke free area). conclusions: the results suggest that in the hospitality venues, where smoking is permitted, there is a high air contamination by environmental tobacco smoke, which are particularly exposed workers. more studies are needed to clarify the situation of tobacco consumption in the hospitality sector.
