Yellow fever virus envelope protein expressed in insect cells is capable of syncytium formation in lepidopteran cells and could be used for immunodetection of YFV in human sera

Abstract: Insect cells infected with vSynYFE showed syncytium formation, which is a cytopathic effect characteristic of flavivirus infection and expressed a polypeptide of around 54 kDa, which corresponds to the expected size of the recombinant E protein. Furthermore, the recombinant E protein expression was also confirmed by fluorescence microscopy of vSynYFE-infected insect cells. Total vSynYFE-infected insect extracts used as antigens detected the presence of antibodies for yellow fever virus in human sera derived from yellow fever-infected patients in an immunoassay and did not cross react with sera from dengue virus-infected patients.The E protein expressed by the recombinant baculovirus in insect cells is antigenically similar to the wild protein and it may be useful for different medical applications, from improved diagnosis of the disease to source of antigens for the development of a subunit vaccine.Yellow fever (YF) is an haemorragic disease caused by a virus and transmitted by mosquitoes through two distinct cycles: the urban YF, transmitted by Aedes aegypti and the sylvatic YF, maintained in a enzootic cycle by Haemagogus and Sabethes mosquitoes with monkeys as main hosts [1]. No cases of urban YF have been reported in Brazil since 1942 [2]. The sylvatic YF is mostly restricted to wild and rural areas but recent outbreaks amongst human visitors and travelers together with the reinfestation of urban areas with the vector mosquito Aedes aegypti have concerned health authorities about the reurbanization of YF.The main mechanisms for YF control consists of vaccination and insect vector control in urban areas. Yet, in 2008, 228 YF epizootic cases were reported and 64 cases of dead monkeys ocurred just in January. These are more than the 104 YF epizootic cases and the 17 cases of dead monkeys during the whole year of 2007. Abstract: reproductive phenology, floral morphology and the reproductive system of schinus terebinthifolius were investigated in two physiognomies of restinga vegetation at grussai/iquipari, rio de janeiro, from january to december/2004. schinus terebinthifolius had cornucopia flowering strategy, with two flowering peaks during the year, both in the rainy season; fruit was set in the dry period. morphological analysis of the flowers and pollination experiments confirmed dioicism and obligatory xenogamy in this species. high synchrony between male and female plants occurred, an important strategy for dioecious species. no difference in fruit set was found between natural and cross pollinations in the two physiognomies studied, indicating lower values than those observed in others studies.

Abstract: the aim of this work is to propose the body warming as one of the basic contents to be taught in physical education classes when concerning gymnastics. based on qualitative approaches, we analyzed texts and contexts which involve the theoretical productions in the area of physical education and health. we concluded that the issue warming is studied by different professional areas, and there are controversies when it comes to types, objectives and purposes. we consider, therefore, the process of texts construction focused on suggesting contents for the teaching of gymnastics may contribute to the organization of knowledge in the area. this may also propose reformulations in the field of pedagogical intervention and methodological innovation in physical education classes.

Abstract: Este artigo trata da rela o entre discurso e ideologia presente na teoria da análise do discurso de linha francesa (AD), elaborada por Michel Pêcheux, que se origina na matriz althusseriana de onde ele retira os conceitos de ideologia e assujeitamento. Por meio de discuss o cuja inflex o teórica é o marxismo estrutural, busca-se entender como o teórico francês reinterpreta os conceitos althusserianos, movimento que o leva a abrir a grade estrutural, reafirmar o conceito marxista de luta de classes no ambito das forma es ideológicas e, ao mesmo tempo, a repensar a no o de discurso de Foucault. This article discusses the relationship between discourse and ideology presented by the French School of Discourse Analysis (DA), elaborated by Michel Pêcheux, and originated in the Althusserian matrix from which he takes all concepts of ideology and subjection. Through a discussion whose theoretical inflection is structural Marxism, this study tries to understand how the French theoretician reinterprets Althusserian concepts, a movement that leads him to reopen the structural matrix, reaffirm the Marxist concept ofclass struggle within ideological formations and, at the same time, rethink Focault’s idea of discourse.

Abstract: Este artigo aborda o discurso a partir de duas pesquisas já concluídas que, tomando por base o texto jornalístico, interpretam tanto o processo discursivo quanto o acontecimento político pela perspectiva do encontro da língua com a história e a ideologia. Desse modo, avalia os princípios e procedimentos teórico-metodológicos da Análise de Discurso de linha francesa (AD) a partir do seu uso no campo das Ciências Sociais, em especial pela Ciência Política. Mostra que o encontro do cientista social com a AD propicia a descoberta de que ela n o constitui um campo exclusivo da Linguística, mas um entremeio formado também pela Psicanálise e o Marxismo, mantendo estreita rela o com as Ciências Sociais. Orientada pela releitura que Althusser faz de Marx, revê os conceitos de ideologia e de sujeito e a rela o que se estabelece entre eles no processo de assujeitamento. O uso desta metodologia, que também é teoria, coloca, muitas vezes, o cientista social, durante o processo de compreens o do objeto investigado, diante de desafios teórico-metodológicos que podem levá-lo à revis o desse entremeio à luz de sua própria área deforma o e do seu campo disciplinar. This paper deals with discourse based on two previous studies with newspaper texts that interpret not only the discourse processbut also the political aspect concerning the linkage of language with history and ideology. Thus, it evaluates the theoretical-methodological principles and procedures of the French Discourse Analysis (DA), based on its use in the Social Sciences field, especially byPolitical Science. It shows that the connection between the social scientist and DA allows the awareness that it is not a field exclusive to Linguistics, but is formed by both Psychoanalysisand Marxism, with close relation to the Social Sciences. Guided by Althusser’s analysis of Marx, it reviews concepts of ideology and subject as well as the relation that is established between them in the subjectivation process. The use of this methodology, which is also a theory, places the social scientist, during the process of understanding the studied object, up against theoretical-methodological challenges that may lead him/her to review thisreflection based on his/her own education and disciplinary fields.

Abstract: this paper reports the occurrence of an megabacterium-like organism at small birds from the northeast area of s?o paulo state - brazil. the results presented herein were obtained from 64 necropsy along four years (1994-1997). sixty four birds (4 budgerigars melopsittacus undulatus, 12 lovebirds agapornis spp and 48 canaries serinus canaria) were studied. at 56% of the examined birds showed at the proventricular mucus smear a presence of rod-shaped (similar to a toothpick) organisms, gram positive and acidophilic in giemsa staining, called megabacteria. different kind of culture media was testes to replicated these organism in vitro. also the dimension (large and width) of the fresh microorganism from the proventricular mucus and from the first culture was compared. the macroscopic alterations observed at the necropsies was also listed.

Abstract: representations that society associates with nurses and nursing, some of them contradictory, persist in individual and collective imaginations. this is not consistent with the development of nursing in terms of professional practice, education and scholarship. images we associate with nurses and nursing are engraved in the memory from the more or less remote past and may have historical, social and cultural justifications. revealing how social representations associated with nurses and nursing have been formed in the past can help us today to re-create the construction of our professional identity and elucidate how we as a social group produce, use, share and take on images that define our own identity. our aim is to reveal how portuguese feminist movements at the beginning of xxth century, namely the liga republicana das mulheres portuguesas (lrmp), has created a positive image of nursing, revealing a praiseworthy discourse attracting the interest of women in a “dignified profession” that could allow them economic independence in cases of abandonment or widowhood. we will use a set of newspaper articles published by the official press of lrmp - “a mulher e a crian?a”, and “a madrugada” - since the printing press was an important way to reveal and spread feminist ideals.
