Colonic Immune Stimulation by Targeted Oral Vaccine

Abstract: Background Currently, sufficient data exist to support the use of lactobacilli as candidates for the development of new oral targeted vaccines. To this end, we have previously shown that Lactobacillus gasseri expressing the protective antigen (PA) component of anthrax toxin genetically fused to a dendritic cell (DC)-binding peptide (DCpep) induced efficacious humoral and T cell-mediated immune responses against Bacillus anthracis Sterne challenge. Methodology/Principal Finding In the present study, we investigated the effects of a dose dependent treatment of mice with L. gasseri expressing the PA-DCpep fusion protein on intestinal and systemic immune responses and confirmed its safety. Treatment of mice with different doses of L. gasseri expressing PA-DCpep stimulated colonic immune responses, resulting in the activation of innate immune cells, including dendritic cells, which induced robust Th1, Th17, CD4+Foxp3+ and CD8+Foxp3+ T cell immune responses. Notably, high doses of L. gasseri expressing PA-DCpep (1012 CFU) were not toxic to the mice. Treatment of mice with L. gasseri expressing PA-DCpep triggered phenotypic maturation and the release of proinflammatory cytokines by dendritic cells and macrophages. Moreover, treatment of mice with L. gasseri expressing PA-DCpep enhanced antibody immune responses, including IgA, IgG1, IgG2b, IgG2c and IgG3. L. gasseri expressing PA-DCpep also increased the gene expression of numerous pattern recognition receptors, including Toll-like receptors, C-type lectin receptors and NOD-like receptors. Conclusion/Significance These findings suggest that L. gasseri expressing PA-DCpep has substantial immunopotentiating properties, as it can induce humoral and T cell-mediated immune responses upon oral administration and may be used as a safe oral vaccine against anthrax challenge.
Disintegrated Comet Trail in Southern Tibet  [PDF]

Abstract: In southern Tibet, there is a series of lakes in the region (82°30'E, 29°N and 90°30'E, 33°N). This study indicates that these lakes were formed by the impact of a single disintegrating comet that hit the region. Observation indicates that the lakes are unusually closely aligned and have a steep slope facing circular feature on the eastern side. Fractures and faults connecting these lakes can be observed over entire subject site hinting towards the multiple impact craters. The terrain is uneven and lakes are partially filled hence in some cases do not look like circular or elliptical. These lakes vary from 1 Km to 65 Km in diameter and are linearly aligned around a single line. Studies of deuterium by Yuan et al. (2011) [1] indicate an abrupt 4‰ increase in δ18O in middle Holocene period around 6000 years ago. The study indicates that the region is rich in diamond, Coesite, Platinum Group Elements (PGE), Stishovite, tektites, and other complex alloys, as would be expected from a high energy impact of an extraterrestrial object. The possibility of the impact as one of the reasons for the diamond in this region was also suggested by Wen-Ji Bai and Qing-Song Fang (2007) [2]. However in absence of definitive evidence they favoured mantle origin. Two possible reasons of absence of impact signature are source of sample and impact pattern. First is the source of the sample which is collected from the river beds of Yarlung-Zangbo Suture Zone of Tibet which is believed to be the origin of this diamond, however it ignores the fact that these rivers get water from lakes which are in the proposed impact zone and the second reason is the impact-pattern spread over close to 800 km which is not expected from the asteroid impact. It will therefore be prudent to test samples from the lake region to understand surface distortion features to confirm the impact. Based on satellite imagery major industrial development supported by road infrastructure in the subject region near lakes and rivers originating from these lakes can be observed hinting towards placer mining activity associated with lakes (craters). Abstract: SEQUEL (a Solver of circuit EQuations with User-defined ELements) is a public-domain simulation package that has been used for a variety of applications. A graphics interface has recently been added to SEQUEL, in order to make it more user-friendly and better suitable for teaching purposes. The package can be downloaded freely. In this paper, some of the features of SEQUEL are described, along with examples.

Abstract: Fundus photography in adults and cooperative children is possible with a fundus camera or by using a slit lamp-mounted digital camera. Retcam TM or a video indirect ophthalmoscope is necessary for fundus imaging in infants and young children under anesthesia. Herein, a technique of converting and using a digital video camera into a video indirect ophthalmoscope for fundus imaging is described. This device will allow anyone with a hand-held video camera to obtain fundus images. Limitations of this technique involve a learning curve and inability to perform scleral depression.

Abstract: Transesophageal echocardiography has been shown to provide unique information about cardiac anatomy, function, hemodynamics and blood flow and is relatively easy to perform with a low risk of complications. Echocardiographic evaluation of the tricuspid and pulmonary valves can be achieved with two-dimensional and Doppler imaging. Transesophageal echocardiography of these valves is more challenging because of their complex structure and their relative distance from the esophagus. Two-dimensional echocardiography allows an accurate visualization of the cardiac chambers and valves and their motion during the cardiac cycle. Doppler echocardiography is the most commonly used diagnostic technique for detecting and evaluating valvular regurgitation. The lack of good quality evidence makes it difficult to recommend a validated quantitative approach but expert consensus recommends a clinically useful qualitative approach. This review ennumerates probe placement, recommended cross-sectional views, flow patterns, quantitative equations including the clinical approach to the noninvasive quantification of both stenotic and regurgitant lesions.
