A Six-Month Supplementation of Mulberry, Korean Red Ginseng, and Banaba Decreases Biomarkers of Systemic Low-Grade Inflammation in Subjects with Impaired Glucose Tolerance and Type 2 Diabetes

Abstract: We sought the long-term efficacy of traditionally used antidiabetic herbs in controlling blood glucose homeostasis and low-grade inflammation. Ninety-four subjects with either impaired glucose tolerance or mild T2D were randomized either to treatment arm or placebo arm and received 1 : 1 : 1 mixture of ginseng roots, mulberry leaf water extract, and banaba leaf water extract (6 g/d) for 24 weeks. Oral 75 g glucose tolerance test was performed to measure glucose and insulin responses. Blood biomarkers of low-grade inflammation were also determined. Results found no significant difference in glucose homeostasis control measure changes. However, plasma intracellular adhesion molecule-1 (ICAM-1) concentration was decreased showing a significant between-treatment changes (=0.037). The concentrations of vascular cell adhesion molecule-1 (VCAM-1) (=0.014) and ICAM-1 (=0.048) were decreased in the treatment group at week 24, and the oxidized low-density lipoprotein (ox-LDL) concentration was reduced at week 24 compared to the baseline value in the treatment group (=0.003). These results indicate a long-term supplementation of ginseng, mulberry leaf, and banaba leaf suppresses inflammatory responses in T2D.

Production of modified liquid glass and quartz materials from kaolinite clay of Alekseev deposit of Kazakhstan was studied. The sodium liquid glass processed by kaolinite clay, with chemical composition (g/dm3): SiO2-350; Na2O-155; Al2O3-2.1; Fe2O3- 0.3, silicate module-2.33 and density-1.49 g/cm3 has been used. The sodium liquid glass complies with the Russian Interstate standard 13078-81 for sodium liquid glass.10 samples of modified liquid glass were synthesized by introducing modifiers in an amount of 4% by weight of the solution into the liquid glass. Hard quartz material produced by using synthesized modified liquid glass was obtained. It has been established that the obtained modified liquid glass from kaolinite clay can improve the strength of the quartz material for 1.6 - 3.12 times higher than the initial liquid glass. Using liquid glass modifier to the inorganic sodium nitrate (NaNO3) increases the strength 3.12 times.

Struma ovarii often escapes recognition during intraoperative consultation because of its rarity, subtle characteristic gross appearance, and lack of clinical suspicion. An intraoperative diagnosis of benign struma ovarii enables the general gynecologic surgeon to continue the planned surgery. However, a diagnosis of malignnancy in a struma ovarii would alter the course of surgery with the involvement of a gynecologic oncology surgeon. We present here that our experience with intraoperative consultation for preoperatively undiagnosed struma ovarii presenting as an adnexal cystic or solid mass at our teaching hospital. Fifty-three cases of struma ovarii, 5.2% of all cystic teratoma of the same period, were diagnosed between January 1991 and March 2011. All intraoperative consultation reports, gross descriptions and final pathology reports were reviewed. The H&E stained slides and in selected cases, immunohistochemistry stained slides, were reviewed. Of the 53 cases of struma ovarii, intraoperative consultation was requested on 48 cases. Frozen section was done on 24 cases and only gross examination was felt appropriate in remaining 24 cases. 83% cases were diagnosed when a frozen section was done. None of the remaining 24 cases were recognized as struma by gross inspection. Our findings reveal that in a large number of cases the diagnosis of struma ovarii remained unrecognized during intraoperative consultation, indicating its often subtle/deceptive gross morphologic appearance. However, the purpose of the intraoperative consultation was served, as appropriate information was provided to the surgeon to guide the surgical management.

Piperine Causes G1 Phase Cell Cycle Arrest and Apoptosis in Melanoma Cells through Checkpoint Kinase-1 Activation

Abstract: In this study, we determined the cytotoxic effects of piperine, a major constituent of black and long pepper in melanoma cells. Piperine treatment inhibited the growth of SK MEL 28 and B16 F0 cells in a dose and time-dependent manner. The growth inhibitory effects of piperine were mediated by cell cycle arrest of both the cell lines in G1 phase. The G1 arrest by piperine correlated with the down-regulation of cyclin D1 and induction of p21. Furthermore, this growth arrest by piperine treatment was associated with DNA damage as indicated by phosphorylation of H2AX at Ser139, activation of ataxia telangiectasia and rad3-related protein (ATR) and checkpoint kinase 1 (Chk1). Pretreatment with AZD 7762, a Chk1 inhibitor not only abrogated the activation of Chk1 but also piperine mediated G1 arrest. Similarly, transfection of cells with Chk1 siRNA completely protected the cells from G1 arrest induced by piperine. Piperine treatment caused down-regulation of E2F1 and phosphorylation of retinoblastoma protein (Rb). Apoptosis induced by piperine was associated with down-regulation of XIAP, Bid (full length) and cleavage of Caspase-3 and PARP. Furthermore, our results showed that piperine treatment generated ROS in melanoma cells. Blocking ROS by tiron protected the cells from piperine mediated cell cycle arrest and apoptosis. These results suggest that piperine mediated ROS played a critical role in inducing DNA damage and activation of Chk1 leading to G1 cell cycle arrest and apoptosis.
rAAV2-mediated Gene Expression In Primary Cortical Neural Cells Following Inhibition Of DNA Synthesis and/or EGFR Tyrosine Kinase

Abstract: Aim: In an effort to enhance rAAV2 transduction efficiency in primary cortical neural cells, we examined the combined effects of chemicals known to increase rAAV2 transduction efficiency in many other cell types.Results: Unexpectedly, pretreatment of neural cells with genotoxic agents (hydroxyurea or aphidicolin) and/or the EGFR tyrosine kinase inhibitor, tyrphostin-1, did not improve transduction efficiency. SK-Hep1 cells expressing dramatic increase in rAAV2 transduction following chemical combination, experienced both significant release from G1/G0 phase and phosphorylation of p38, whereas no in neural cells.Conclusion: Our results suggest that chemical pretreatment with hydroxyurea, aphidicolin and/or tyrphostin-1 is unable to increase rAAV2 transduction efficiency in neural cells, indicating that other strategies should be investigated.
Structural evolution of bismuth sodium titanate induced by A-site non-stoichiometry: Neutron powder diffraction studies

Abstract: We performed neutron powder diffraction measurements on (Bi$_{0.5}$Na$_{0.5+x}$)TiO$_3$ and (Bi$_{0.5+y}$Na$_{0.5}$)TiO$_3$ to study structural evolution induced by the non-stoichiometry. Despite the non-stoichiometry, the local structure ($r$$\leq$ 3.5 {\AA}) from the pair distribution function analysis is barely affected by the sodium deficit of up to -5 mol%. With increasing pair distance, however, the atomic pair correlations weaken due to the disorder caused by the sodium deficiency. Although the sodium and the bismuth share the same crystallographic site, their non-stoichiometry have rather opposite effects as revealed from a distinctive distortion of the Bragg peaks. In addition, Rietveld refinement demonstrates that the octahedral tilting is continually suppressed by the sodium deficit of up to -5 mol%. This is contrary to the effect of the bismuth deficiency, which enhances the octahedral tilting.

Abstract: kakiemon-style porcelain is mainly characterized by the porcelain body of white color or milky-white color and the elegant and bright patterns of the colored overglazes on its white body. the porcelains were first developed and completed by kakiemon-kiln family in 1650-80's. the red-color overglazes and the transparent glazes of the present porcelain pieces, which were produced in the early 17th century to the middle 18th century, have been investigated by means of x-ray absorption spectrum using synchrotron radiation. the structure of α-fe2o3 as red-color emission element is slightly deformed in the red-color overglazes by the other surrounding oxides (sio2, al2o3, k2o, na2o, mgo, pbo), and/or the new complexes of (sio2-al2o3,) systems constructed by the raw materials. the fe ions in the transparent glazes are in the glass-state with the other oxides. however, the electronic valence, fe3+, of fe ions is the same with that of the α-fe2o3 in the red-color overglazes. the lead oxides (pbo, pb3o4) used in the edo period to increase the brightness in the fritted red-color overglazes were perfectly decomposed, and pb ions were in the glass-state of the porcelains.
SAP97 Controls the Trafficking and Resensitization of the Beta-1-Adrenergic Receptor through Its PDZ2 and I3 Domains

Abstract: Previous studies have determined that the type-1 PDZ sequence at the extreme carboxy-terminus of the ?1-adrenergic receptor (?1-AR) binds SAP97 and AKAP79 to organize a scaffold involved in trafficking of the ?1-AR. In this study we focused on characterizing the domains in SAP97 that were involved in recycling and resensitization of the ?1-AR in HEK-293 cells. Using a SAP97 knockdown and rescue strategy, we determined that PDZ-deletion mutants of SAP97 containing PDZ2 rescued the recycling and resensitization of the ?1-AR. Among the three PDZs of SAP97, PDZ2 displayed the highest affinity in binding to the ?1-AR. Expression of isolated PDZ2, but not the other PDZs, inhibited the recycling of the ?1-AR by destabilizing the macromolecular complex involved in trafficking and functional resensitization of the ?1-AR. In addition to its PDZs, SAP97 contains other protein interacting domains, such as the I3 sequence in the SRC homology-3 (SH3) domain, which binds to AKAP79. Deletion of I3 from SAP97 (ΔI3-SAP97) did not affect the binding of SAP97 to the ?1-AR. However, ΔI3-SAP97 could not rescue the recycling of the ?1-AR because it failed to incorporate AKAP79/PKA into the SAP97-?1-AR complex. Therefore, bipartite binding of SAP97 to the ?1-AR and to AKAP79 is necessary for SAP97-mediated effects on recycling, externalization and functional resensitization of the ?1-AR. These data establish a prominent role for PDZ2 and I3 domains of SAP97 in organizing the ?1-adrenergic receptosome involved in connecting the ?1-AR to trafficking and signaling networks.
Magnetic penetration depth in single crystals of SrPd$_2$Ge$_2$ superconductor

Abstract: The in-plane magnetic penetration depth, $\lambda_m(T)$, was measured in single crystals of SrPd$_2$Ge$_2$ superconductor in a dilution refrigerator down to T=60 mK and in magnetic fields up to $H_{dc} = 1$ T by using a tunnel diode resonator. The London penetration depth, $\lambda$, saturates exponentially approaching $T\rightarrow 0$ indicating fully gapped superconductivity. The thermodynamic Rutgers formula was used to estimate $\lambda(0) = 426$ nm which was used to calculate the superfluid density, $\rho_s(T)=\lambda^2(0)/\lambda^2(T)$. Analysis of $\rho_s(T)$ in the full temperature range shows that it is best described by a single - gap behavior, perhaps with somewhat stronger coupling. In a magnetic field, the measured penetration depth is given by the Campbell penetration depth which was used to calculate the theoretical critical current density $j_c$. For $H \le 0.45$ T, the strongest pinning is achieved not at the lowest, but at some intermediate temperature, probably due to matching effect between temperature - dependent coherence length and relevant pinning lengthscale. Finally, we find a compelling evidence for surface superconductivity. Combining all measurements, the entire $H$-$T$ phase diagram of SrPd$_2$Ge$_2$ is constructed with an estimated $H_{c2}(0)=0.4817$ T.

Abstract: hizen celadon glazes produced in 1630's to 1790's (edo period, japan) have been investigated by means of x-ray absorption spectra (xas) near a fe-k edge by using synchrotron radiation and a m?ssbauer spectrum. the xas suggest that the local structure around fe2o3 fine powders is slightly different between the izumiyama ceramics of mainly the quartz-sio2 and ohkawachi ceramics of mainly the feldspar of (k,na)si3o8 (sanidine), and that the glazes of the hizen celadons include the fe2o3 fine powders in the glassy state, though the x-ray diffraction patterns of the glassy celadon glazes do not show any peaks of the fe2o3 structure. the m?ssbauer spectrum suggests that the celadon glaze of seiji (m) includes only fe3+ ions, but not fe2+ ions. this indicates the existence of fe2o3 in the celadon glaze. it is interpreted that the colored brightness of the hizen celadons is induced by the structural properties of the used raw celadon ceramics and the other transition-metal ions of cr, cu, zn in the celadon glazes, but not by the chemical reaction from fe2o3 to feo under the deoxidizing thermal treatment at higher temperature in a kiln.
